以当年采收的洋葱为试验材料,采用正交试验设计,研究不同培养基质和植物生长调节剂对洋葱根生长速度影响。结果表明:①以水为培养基质进行培养时,使洋葱根快速长长的最佳方法是用150mg/L植物生长调节剂NAA处理5分钟后室温下培养;②以沙为培养基质进行培养时,使洋葱根快速长长的最佳方法是用50mg/L植物生长调节剂6-BA处理5分钟后25℃培养。  相似文献

为研究祁术的组织培养及快速繁殖技术,选用祁术的叶片和下胚轴为外植体材料,接种于附加植物生长调节剂NAA,6-BA及其组合的MS固体培养基上.结果表明:下胚轴为外植体明显好于叶片,对于愈伤组织形成,叶片采用MS NAA(0.5mg/L) 6-BA(2mg/L);下胚轴MS NAA(0.5mg/L) 6-BA(2mg/L);MS NAA(0.5mg/L) 6-BA(1mg/L)为宜.对于叶片丛生芽愈伤组织培养基用MS NAA(0.2mg/L) 6-BA(2.8mg/L),下胚轴丛生芽愈伤组织培养基用MS NAA(0.01mg/L) 6-BA(0.1mg/L);MS NAA(0.05mg/L) 6-BA(0.1mg/L)为宜,诱导生根以1/2MS NAA0.3mg/L最佳,生根率达95%以上.  相似文献

以樱桃番茄新品种“圣女”和“金珠”的叶片、茎段和幼芽为外植体进行试管培养 ,结果表明 :叶片、茎段和幼芽均可诱导形成愈伤组织 ,愈伤组织可分化出芽丛 ,腋芽可直接萌生 ,不定芽可诱导生根成完整植株 .经试验筛选出各培养阶段的最适培养基为 :(1)愈伤组织诱导 :叶片以MS Kt4mgL IAA8mgL为最佳 ;茎段和幼芽以MS NAA0 1mgL 6 -BA1 5mgL为最佳 ;(2 )愈伤组织芽的分化 :MS IAA0 2mgL 6 -BA2mgL ;(3)腋芽萌生 :MS 6 -BA2 5mgL IBA0 1mgL ;(4)生根 :无激素的Ms或Mso .  相似文献

以“银粉金鳞”鳞芽为材料进行离体培养,研究了植物生长调节剂6-BA和NAA对鳞芽不定芽诱导分化、增殖培养的影响.结果表明:MS+Ca^2+ +6-BA1.0mg/L+NAA0.3mg/L为丛生芽诱导最佳培养基,诱导率高达81.67%;MS+Ca^2+ +6-BA1.6mg/L+NAA0.2mg/L为最佳增殖培养基.增殖系数为3.21.  相似文献

以黄荆种子作为实验材料,分别研究不同质量浓度的植物激素和植物生长调节剂对黄荆种子萌发及根茎叶等生理指标的测定。通过研究黄荆种子在不同处理下萌发的最佳条件,以筛选适合黄荆种子萌发的植物激素和植物生长调节剂,为黄荆的繁殖培育提供依据。结果表明:在棉花、土壤、纱布3种不同的基质处理中,黄荆种子均能萌发,但在土壤基质中的发芽效果最好;在不同质量浓度的植物激素和植物生长调节剂处理中,NAA、IAA、GA3、6-BA处理都有利于黄荆种子萌发,其中,200 mg/L的GA3处理对黄荆种子的发芽和生长效果最好;在不同的植物激素和植物生长调节剂浸种时间中,用GA3浸泡处理6 h可以对黄荆种子的发芽率、发芽指数、活力指数以及对黄荆幼苗的平均根长有促进作用,效果最好。  相似文献

以菊花的花瓣作外植体进行组培快繁技术研究,结果表明:外植体用0.1%升汞消毒10min为宜;诱导愈伤组织的最佳培养基为MS 6-BA 1.5mg/L NAA0.1mg/L;丛生芽形成的最佳培养基为MS 6-BA1.5mg/L NAA0.1mg/L;用1/2MS NAA0.5mg/L作生根培养基,生根率可达100%.  相似文献

以二色补血草的茎为试材,采用MS和LS两种基本培养基,组配不同浓度的植物生长激素,诱导植株再生。本文探讨了培养基和激素对诱导愈伤组织和不定芽的影响,找出了较适宜茎诱导植株再生的基本培养基为MS。最佳生长激素6-BA浓度为0.3~0.6mg/L。初步建立了二色补血草的茎诱导一次成芽培养基为MS+6-BA0.3~0.6mg/L+NAA0.05mg/L+Augar30g/L;对诱导再生植株生根的几种激素也进行了研究。结果显示:最佳生根激素为IAA,其适宜浓度为0.2mg/L。生根培养基为1/2MS〕IAA0.2mg/L+Sugar20g/L。  相似文献

以密叶罗勒的茎尖为外植体,以期筛选出诱导愈伤组织分化和丛生芽生根的最佳培养基。同时研究了密叶罗勒组织培养中材料的消毒时间对消毒效果的影响。试验结果表明,培养基MS+6-BA5.0mg/L+NAA0.1mg/L是愈伤组织诱导分化相对较好的培养基;培养基MS+6-BA0.3mg/L+NAA0.6mg/L是罗勒生根培养较好的培养基。在以75%的酒精消毒30秒后,用0.1%的升汞消毒2min,对试验材料的消毒效果相对较好。  相似文献

以牛大力茎段离体培养的无菌芽作为试验材料,以MS为基本培养基,进行双因子和三因子激素组合处理比较试验,探讨外源激素因子对牛大力离体培养芽增殖的影响。结果表明:在双因子激素组合处理中,与NAA0.2mg/L组合的6-BA适宜浓度为3.0mg/L,芽增殖倍数分别为3.70倍(30d)、5.43倍(60d);KT适宜浓度为4.0mg/L,芽增殖倍数分别为2.03倍(30d)、2.54倍(60d);与6-BA3.0mg/L组合的生长素IAA适宜浓度为0.2mg/L,芽增殖倍数分别为4.38倍(30d)、6.55倍(60d)。在三因子激素组合处理中,对牛大力芽增殖效应依次为:6-BA+IAA+TDZ组合处理>6-BA+IAA+CPPU组合处理>6-BA+IAA组合处理(作对照);适宜的三因子激素组合为6-BA3.0mg/L+IAA0.2mg/L+TDZ0.03mg/L,其芽增殖倍数分别高达6倍(30d)和9.65倍(60d)。  相似文献
