癸基硫酸钠-聚丙烯酰胺凝胶电泳分析非变性蛋白质分子量的适用性鉴定

目的:鉴定癸基硫酸钠-聚丙烯酰胺凝胶电泳(SDecS-PAGE)是否适于分析非变性蛋白质的分子量。方法:以不同SDecS-PAGE凝胶、电泳缓冲液分析5种非变性标准分子量蛋白质,观察蛋白质亚基解离情况,测定分子量对数值(log Mr)与迁移率x之间的相关性。结果:5种标准分子量蛋白质的四级结构完整,log Mr与x线性相关性较低;但其中的α-牛乳白蛋白、鸡蛋清白蛋白、牛血清白蛋白的log Mr与x线性相关性较好,相关系数平方值(R2)>0.94。结论:SDecS-PAGE可用于测定有限种类的非变性蛋白质的分子量。

Evaluating applicability of sodium decyl sulphate-polyacrylamide gel electrophoresis in determination of molecule weight of non-denatured protein

Objective:To evaluate the applicability of sodium decyl sulphate-polyarcrylamide gel electrophoresis(SDecS-PAGE) in the determination of the molecule weight of non-denatured proteins.Methods:Different SDecS-PAGE gels and buffers were used for the determination of five different standard molecule weight protein,and then correlations of the logarithm values of protein molecule weights and migration rates x were calculated.Results:Quarternary structures of all five proteins were all kept intact in SDecS-PAGE gels,but the logarithm values of protein molecule weights were found to be not in linear correlation with migration ratex in all experiments if all the five proteins were taken for analysis.If only the three proteins,i.e.,α-lactalbumin from milk,albumin from chicken egg white and albumin from bovine serum,were included,the linearity seems to be quite good(R2>0.94).Conclusion:The SDecS-PAGE could only be used for protein molecule weight analysis for limited kinds of non-denatured proteins.
