分子生物学实验教学中液相Northern杂交检测microRNA实验探索

摘    要:为了丰富分子生物学实验教学内容,加深学生对分子杂交原理等专业理论知识的理解,在分子生物学实验教学中开展液相Northern杂交检测microRNA的实验探索。利用荧光标记的寡核苷酸做探针,通过探针灵敏性、杂交温度、杂交时间和杂交液等不同杂交条件的探索,从而简便、准确、灵敏地杂交检测不同植物中的microRNA。整个实验过程仅需几个小时即可完成,且实验结果直观生动。

Abstract:In order to enrich the experimental teaching content of molecular biology and deepen students' understanding of the theoretical knowledge of molecular hybridization,the liquid-phase Northern hybridization for detection of microRNA is carried out in the experimental teaching of molecular biology. Fluorescent labeled oligonucleotides are used as probes to explore different hybridization conditions such as probe sensitivity,hybridization temperature,hybridization time and hybridization solution,so as to detect microRNA in different plants simply,accurately and sensitively.The whole experimental process only takes a few hours to complete,and the experimental results are intuitive and vivid.
