筋骨草中黄酮类化合物提取工艺的探究

摘    要:以乙醇回流提取法提取筋骨草中黄酮类化合物,采用比色法测定总黄酮含量,跟踪乙醇浓度、料液比、提取时间、提取温度等因素对黄酮类化合物提取量的影响,通过正交实验确定最佳的提取工艺条件。并以槲皮素为对照品,通过高效液相色谱法(HPLC)表征了溶液的提取物为黄酮类化合物。结果表明,适宜的提取工艺条件为:乙醇浓度60%,料液比1:20,提取温度为70℃,提取时间为2.5h。

Abstract:The flavonoid was extracted from the Ajuga Ciliate via ethanol reflux extraction method. The extraction content of flavonoid were followed and determined by colorimetry. The effects of the ethanol concentration, ratio of solid to liquid, temperature of extraction and time of extraction on the extraction content were followed. The optimal experimental conditions of extracting the flavonoid were determined through orthogonal experiment. It was proven that the extractive is flavonoid by high performance liquid chromatography(HPLC) with quercetin as reference substance. The results showed that the appropriate extraction process conditions were as follows: ethanol concentration is 60 %; ratio of solid to liquid is 1 : 20; temperature of extractio n is 70℃; time of extraction is 2.5 hour.
