摘    要:采用改良CTAB法对15个白梨品种叶片DNA提取情况进行了研究,考察了Vc和β-巯基乙醇抗氧化效果,所获得DNA样品经电泳、单酶切以及PCR检测,结果表明用氯仿/异戊醇混合萃取液多萃取几次可有效减少高蛋白质含量样品中的蛋白质的含量;用5mol/L的NaCl溶液能有效降低梨DNA样品中多糖的含量;Vc抗氧化效果优于β-巯基乙醇,Vc用量在0.1~0.30g/g样品时,可有效减少提取过程中酚类氧化。

Abstract:Genomic DNA of 15 P.bretschnider cultivators' leaves is isolated in amended CTAB method,accompanied by comparison between the effect of adding with anti-oxidation of Vc and BME.The obtained DNA is subjected to gel electrophoresis,single enzymatic digestion and PCR.The result shows that the highprotein sample handled with chloroform-isoamylalcohol(24:1) for more 2 times can effectively reduce the content of protein;the high-sugar sample adding 5mol/L NaCl can effectively decrease the content of amylose.Vc is prior to BME for the effect of anti-oxidation,and Adding 0.1-0.30g Vc per gram sample for anti-oxidation is effective for high-quality DNA.
