The reuse of wastewater for the irrigation of farmlands is gaining popularity, and the nutrient leaching associated with wastewater irrigation is becoming a matter of concern. The variability of nitrogen and phosphorus fractions in wastewater-irrigated soil was investigated in both horizontal and vertical directions in Kongjiaxiang, Tongliao, Inner Mongolia, China. The results showed that wastewater irrigation resulted in the concentrations of available N and P being 40.36% and 66.49% higher, respectively, than those with groundwater irrigation. Different forms of N and P exhibited significantly different distribution patterns. Higher concentrations of NO3- and total available N, as well as of those of Ca-P and total available P were observed near wastewater irrigation channels. Ca-P has a spatial distribution pattern similar to that of available P. The concentrations of NO3- and NH4+ were the highest in top soil and decreased with depth. The complex interactions between nitrate leaching and nitrogen transformation processes (e.g., nitrification, denitrification, and mineralization) determined the vertical profile of NO3- . The significant amount of NH4+ loss inhibited its deep seepage. The Ca-bound compound contained more P than other inorganic fractions as a result of high Ca levels throughout the soil profile. The differences in the concentrations of Ca-bound P at different depths could be due to the upward flux or translocation of Ca from subsurface to surface soil and the sequestration of P.  相似文献

Objective

We investigated the influence of β-receptor blocker metoprolol on return of spontaneous circulation (ROSC) during cardiopulmonary resuscitation (CPR) in rats with induced myocardial infarction (MI).

Methods

Male Sprague-Dawley rats were randomly divided into three groups: the sham-operated group, the MI group without metoprolol, which was fed the vehicle, and the MI+metoprolol group receiving intragastric metoprolol. Each group was further divided randomly into three subgroups, depending on the dosage of epinephrine administered during subsequent CPR applied after the induction of asphyxial cardiac arrest.

Results

The ROSC rate was significantly decreased in the low dose subgroup of MI group, unchanged in the medium dose subgroup of MI group, and significantly decreased in the high dose subgroup of MI group, compared with the same dose subgroup of sham-operated group. MI+metoprolol group had a lower ROSC rate than MI group in the medium dose subgroup, and a higher ROSC rate than MI group in the high dose subgroup. There was no difference in blood K+ values of successful rats between MI group and MI+metoprolol group. The rats with successful CPR had lower blood K+ values than rats with unsuccessful CPR in each of the three treatment groups.

Conclusions

Metoprolol administered to MI rats over a long period significantly improved ROSC rates under an appropriate dose of epinephrine during CPR. An increasing high blood K+ value would attenuate the rate of a successful CPR.  相似文献

In recent years, excessive use of chemical nitrogen (N) fertilizers has resulted in the accumulation of excess ammonium (NH 4 + ) in many agricultural soils. Though rice is known as an NH 4 + -tolerant species and can directly absorb soil intact amino acids, we still know considerably less about the role of high exogenous NH 4 + content on rice uptake of soil amino acids. This experiment examined the effects of the exogenous NH 4 + concentration on rice uptake of soil adsorbed glycine in two different soils under sterile culture. Our data showed that the sorption capacity of glycine was closely related to soils;’ physical and chemical properties, such as organic matter and cation exchange capacity. Rice biomass was significantly inhibited by the exogenous NH 4 + content at different glycine adsorption concentrations. A three-way analysis of variance demonstrated that rice glycine uptake and glycine nutritional contribution were not related to its sorption capacity, but significantly related to its glycine:NH 4 + concentration ratio. After 21-d sterile cultivation, the rice uptake of adsorbed glycine accounted for 8.8%;–22.6% of rice total N uptake, which indicates that soil adsorbed amino acids theoretically can serve as an important N source for plant growth in spite of a high NH 4 + application rate. However, further studies are needed to investigate the extent to which this bioavailability is realized in the field using the 13C, 15N double labeling technology.  相似文献

对烟草根系施用5种配方肥料,并分别检测烟草移栽前、后20,40,60,120 d的根际土壤微生物,比较研究了不同肥料对烟草根际微生物的影响.实验结果显示:随着有机肥比例的增加,烟草根际土壤中的真菌、细菌等数量均比不添加有机肥的处理(N0处理)呈增加趋势;其中,50%无机氮+50%有机氮处理(N3处理)能显著增加烟草根际土壤中的微生物数量,真菌、细菌、霉菌数量分别达到2.46×10^5,2.96×10^8,2.95×10^4个/g .在烟草种植中,施用50%无机氮+50%有机氮的有机肥有利于提高烟草根际微生物的数量.  相似文献

Objective:To evaluate the effect of chitosan(CH) and hydrogen peroxide(H2O2) seed coatings and seedling sprinklings on two different maize varieties by measuring their phenology,the H2O2 presence,the catalase(CAT) activity,and the protein quantity.Methods:Seven groups of ten seeds for each maize variety were treated with CH(2%(20 g/L) and 0.2%(2 g/L)) or H2O2(8 mmol/L) by coating,sprinkling,or both.Germination and seedling growth were measured.One month after germination,the presence of H2O2 in seedlings in the coated seed treatments was evaluated.Protein content and CAT activity were determined under all treatments.Results:H2O2 seed coating enhanced the germination rate and increased seedling and stem length in the quality protein maize(QPM) variety.Seedlings had a higher emergence velocity under this treatment in both varieties.CH and H2O2 sprinklings did not have an effect on seedling phenology.Exogenous application of H2O2 promoted an increase of endogenous H2O2.CH and H2O2 seedling sprinkling increased the protein content in both maize varieties,while there was no significant effect on the CAT activity of treated seeds and seedlings.Conclusions:CH and H2O2 enhance some phenological and biochemical features of maize depending on their method of application.  相似文献

Phytoremediation is emerging as a potential cost-effective solution for remediation of contaminated soils, and bioavailability of metal in the soil for plant uptake is an important factor for successful phytoremediation. This study aimed at investigating the ability of EDTA and citric acid for enhancing soil bioavailability of Cu and phytoremediation by Elsholtzia splendens in two types of soils contaminated with heavy metals [i.e. mined soil from copper mining area (MS), and paddy soil (PS) polluted by copper refining]. The results showed that addition of 2.5 mmol/kg EDTA significantly increased the H2O extractable Cu concentration from 1.20 to 15.78 mg/kg in MS and from 0.26 to 15.72 mg/kg in PS, and that shoot Cu concentration increased 4-fold and 8-fold as compared to the control. There was no significant difference between the treatment with 5.0 mmol/kg EDTA and that with 2.5 mmol/kg EDTA, probably because that 2.5 mmol/kg EDTA was enough for elevating Cu bioavailability to the maximum level. As compared with the control, citric acid had no marked effect on both soil extractable Cu and shoot Cu concentration or accumulation. The results indicated that EDTA addition can increase the potential and efficiency of Cu phytoextraction byE. splendens in polluted soils. Objective: This study was designed to investigate the pharmacokinetics of borneol in the pathological conditions of stroke and evaluate the pharmacokinetic differences of borneol caused by stroke after oral administration of borneol and Xingnaojing (XNJ). Methods: The rats were divided into two groups, ischemia-reperfusion (IR) and sham-operated (SO) rats. Each group contained two subgroups: pure borneol and XNJ subgroups. After administration with the same dosages of borneol 162.0 mg/kg, plasma samples were collected. The cerebral ischemia-reperfusion model was created by reversible middle cerebral artery occlusion (MCAO). The blood samples were collected punc- tually after oral administration and a specific gas chromatographic system-flame ionization detector (GC-FID) method was developed and employed to determine the level of borneol in the plasma. The pharmacokinetic parameters were analyzed using non-compartmental methods with Kinetica. Results: After administration of borneol, the maximum plasma concentration (Cmax) and area under the curve (AUC) values in stroke rats significantly increased by 302% and 275%, respectively, compared with the SO rats, and the same phenomenon appeared after administration of XNJ. In the rats with the same physiological conditions, the Cmax and AUC had higher values in the borneol subgroup (P〈0.05). Conclusions: These results suggest that the pathological damages of ischemia-reperfusion have a significant impact on the pharmacokinetic traits of borneol and that there are some components in XNJ inhibiting the absorption of borneol.  相似文献

Background and objective

It has been shown that macrophages play an important role in the development of severe acute pancreatitis (SAP), and eventually lead to multiple organ failure (MOF). Clodronate-liposome selectively depleted macrophages. This study was to investigate the role of renal macrophage infiltration in acute renal injury in rats with SAP and to evaluate the potential of superparamagnetic iron oxide (SPIO)-enhanced magnetic resonance imaging (MRI) for diagnosis.

Methods

Superparamagnetic Fe3O4 nanoparticles were prepared by chemical coprecipitation. SPIO-liposomes and SPIO-clodronate-liposomes were prepared by the thin film method. SAP models were prepared by injection of sodium taurocholate into the subcapsular space of rat pancreas. Sprague-Dawley rats were randomly divided into a control group, SAP plus SPIO-liposome (P) group, and SAP plus SPIO-clodronate-containing liposome (T) group. Kidney injury was evaluated by T2-weighted MRI scan. The levels of serum amylase (SAM), blood urea nitrogen (BUN), and serum creatinine (SCr) were measured by an automated enzymatic method. Serum tumor necrosis factor-α (TNF-α) was measured by enzyme-linked immunosorbent assay (ELISA). Pathological changes in the pancreas and kidney were observed using hematoxylin and eosin (H&E) staining, while cell apoptosis was detected with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining. In addition, the macrophage markers (CD68) of the renal tissue were detected with immunohistochemistry.

Results

The pathological changes in the pancreas and kidneys of rats in the T group were milder than those in the P group. The MRI signal intensity of the kidneys in the P and T groups was significantly lower than that in the control group. There were significant changes in the two experimental groups (P<0.01). The levels of SAM, Bun, SCr, and TNF-α in rats in the P group were higher than those in the control group (P<0.01) and in the T group (P<0.01). The apoptosis of the kidney in the T group was higher than that in the P group at 2 and 6 h (P<0.01).

Conclusions

Clodronate-containing liposomes protected against renal injury in SAP rats, and SPIO can be used as a tracer for MRI examination to detect renal injury in SAP rats. SPIO-aided MRI provided an efficient non-invasive way to monitor the migration of macrophages after renal injury in rats with SAP.  相似文献

研究目的:探讨萎叶(PB)提取物对5-氟尿嘧啶(5-FU)抑制结肠癌细胞HT29和HCT116生长的影响。研究方法:HT29和HCT116细胞分别给予PB、5-FU以及两种药物联合治疗24小时,应用等效线图法分析PB和5-FU的药效学相互作用,AnnexinV/PI染色法检测HT29和HCT116细胞的凋丁L=情况,高效液相色谱法排除PB和5-FU间任何可能的相互化学作用。重要结论:联合PB,低剂量5-FU可以在短时间内起到细胞毒作用,而单独应用PB或5-FU治疗较联合治疗可以诱导更多细胞发生凋亡。进一步采用等效线图法分析显示PB和5-Fu的联合作用在抑制结肠癌细胞HT29和HCT116的生长中分别体现出协同和拮抗作用。因此可以认为在HT29细胞中,PB使得较低剂量5-FU发挥最大抑制结肠癌细胞生长效果,然而在HCT116细胞中,PB没有显著降低5-FU的药物浓度,说明PB和5-FU的相互作用不仅仅体现在诱导细胞凋亡方面。  相似文献

Considering the great importance of the elastic shear modulus G0 of unsaturated soils to the serviceability of many geo-structures in geo-energy and geo-environmental engineering, some semi-empirical models have been reported for the G0 of unsaturated soils. Existing models require at least three parameters and the calibration of the model parameters requires extensive time-consuming unsaturated soil tests. In this study, a simple semi-empirical model is proposed for the hysteretic G0 of unsaturated soils, requiring only two parameters. The constitutive variables of the mean Bishop’s stress and a bonding variable are adopted for considering the average stress between soil particles and the additional normal forces between soil particles provided by water menisci. The derived equation is applied to simulate the G0 of unsaturated silts and sands. Comparisons between the measured and calculated results demonstrate that the proposed equation is able to describe the influences of various factors on G0, including mean net stress, suction, wetting-drying, and void ratio.  相似文献

研究目的:在小鼠模型中利用吲哚美辛阻断COX-1/2通路,探讨非甾体类抗炎药对肾缺血再灌注损伤的保护作用。创新要点:非甾体类抗炎药被认为具有肾毒性,本研究首次在小鼠模型中探讨非甾体类抗炎药对肾缺血再灌注损伤的保护作用。研究方法:小鼠左侧肾蒂夹闭后,通过腹腔注射不同剂量的吲哚美辛,在肾缺血再灌注损伤24小时后,获取血液和肾脏标本。利用酶联免疫(ELISA)试剂盒测定血清肌酐和细胞因子浓度来评估肾功能,肾组织样本进行苏木精-伊红染色和免疫组化分析。重要结论:腹腔注射吲哚美辛5 mg/kg组的小鼠血清肌酐值与对照组相比显著降低,肾小管损伤也显著减轻(见图1和2);腹腔注射5和7 mg/kg吲哚美辛组的小鼠血清肾肿瘤坏死因子-α和白介素-6的浓度显著降低(见图3a和3b);腹腔注射3和5 mg/kg吲哚美辛组的小鼠血清血栓素B2与6-酮前列腺素F1α的比值明显降低(见图3c);腹腔注射5 mg/kg吲哚美辛组小鼠肾组织COX-1和COX-2染色较弱(见图4)。因此,吲哚美辛对小鼠肾缺血再灌注损伤的作用与其剂量相关,在某个特定的剂量范围内具有肾保护作用。吲哚美辛对小鼠肾缺血再灌注损伤的保护作用与阻断COX-1/2有关。  相似文献

Objective

The ideal medication for the treatment of acid-related diseases, e.g., peptic ulcers, stress-related gastric bleeding, functional dyspepsia, and gastroesophageal reflux disease, should have a rapid onset of action to promote hemostasis and relieve the symptoms. The aim of our study was to investigate the inhibitory effects on gastric acid secretion of a single oral administration of a proton pump inhibitor, omeprazole 20 mg, and an H2-receptor antagonist, roxatidine 75 mg.

Methods

Ten Helicobacter pylori-negative male subjects participated in this randomized, two-way crossover study. Intragastric pH was monitored continuously for 6 h after single oral administration of omeprazole 20 mg and roxatidine 75 mg. Each administration was separated by a 7-d washout period.

Results

During the 6-h study period, the average pH after administration of roxatidine was higher than that after administration of omeprazole (median: 4.45 vs. 2.65; P=0.0367). Also during the 6-h study period, a longer duration of maintenance at pH above 2, 5, and 6 was observed after administration of roxatidine 75 mg than after administration of omeprazole 20 mg (median: 90.6% vs. 55.2%, P=0.0284; 43.7% vs. 10.6%, P=0.0125; 40.3% vs. 3.3%, P=0.0125; respectively).

Conclusions

In Helicobacter pylori-negative healthy male subjects, oral administration of roxatidine 75 mg increased the intragastric pH more rapidly than that of omeprazole 20 mg.  相似文献

Eleven red soils varying in land use and fertility status were used to examine the effect of land use on microbial biomass-C,-N and-P. Microbial biomass-C in the red soils ranged from about 68 mg C/kg to 225 mg C/kg, which is generally lower than that reported from other types of soil, probably because of low organic matter and high acidity in the red soils. Land use had considerable effects on the amounts of soilC mic. TheC mic was the lowest in eroded fallow land, followed by woodland, tea garden, citrus grove and fallow grassland, and the highest in vegetable and paddy fields. There was significant correlation betweenC mic and organic matter content, suggesting that the influence of land use onC mic is mainly related to the input and accumulation of organic matter. Microbial biomass-N in the soils ranged from 12.1 Nmg/kg to 31.7 Nmg/kg and was also affected by land use. The change ofN mic with land use was similar to that ofC mic. The microbial C/N ratio ranged from 5.2 to 9.9 and averaged 7.6. TheN mic was significantly correlated with soil total N and available N. Microbial biomass-P in the soils ranged from 4.5 mg P/kg to 52.3 mg P/kg. The microbial C/P ratio was in the range of 4–23. TheP mic was relatively less affected by land use due to differences in fertilization practices for various land use systems. Objective

Heavy ion beam, which has emerged as a new mutagen in the mutation breeding of crops and ornamental plants, is expected to result in the induction of novel mutations. This study investigates the morphological and biochemical responses of Oryza sativa toward different doses of carbon ion beam irradiation.

Methods

In this study, the dry seeds of O. sativa were irradiated at 0, 20, 40, 60, 80, 100, and 120 Gy, followed by in-vitro germination under controlled conditions. Morphological and biochemical studies were conducted to investigate the morphological and physiological responses of O. sativa towards ion beam irradiation.

Results

The study demonstrated that low doses (10 Gy) of ion beam have a stimulating effect on the height, root length, and fresh weight of the plantlets but not on the number of leaves. Meanwhile, doses higher than 10 Gy caused reductions in all the morphological parameters studied as compared to the control samples. The highest total soluble protein content [(2.11±0.47) mg/g FW] was observed in plantlets irradiated at 20 Gy. All irradiated plantlets were found to have 0.85% to 58.32% higher specific activity of peroxidase as compared to the control samples. The present study also revealed that low doses of ion beam (10 and 20 Gy) had negligible effect on the total chlorophyll content of O. sativa plantlets while 40 Gy had a stimulating effect on the chlorophyll content. Plantlets irradiated between 40 to 120 Gy were shown to be 0.38% to 9.98% higher in total soluble nitrogen content which, however, was not significantly different from the control samples.

Conclusions

Carbon ion beam irradiation administered at low to moderate doses of 10 to 40 Gy may induce O. sativa mutants with superior characteristics.  相似文献

Low phosphorus (P) availability is one of the most important factors limiting plant growth in red soils across southeastern China. Many non-symbiotic microorganisms in rhizosphere can enhance P solubility, but little is known about the magnitude of their phosphorus-solubilizing ability (PSA) and the difference in phosphorus-solubilizing microorganisms (PSM) among plant species. The number of phosphorus-solubilizing microorganisms and their PSA in rhizosphere soils of 19 weed species in a citrus orchard on red soil at Changshan, Zhejiang, China, were investigated. Inorganic P (powdered phosphate rock, PR) and organic P (lecithin, OP) were respectively used as the sole P-source to examine the PSA of isolated microbes. The PS actinomycetes community varied greatly among the different weed rhizospheres while the PS fungus community showed to be most stable to the weed rhizosphere. The highest number of PR-PS and OP-PS bacteria was found in rhizosphere soil of Mollugo pentaphyll, and the highest number of PR-PS and OP-PS actinomycetes was found in rhizosphere soil of Polygonum lapathifolium. The highest number of PR-PS fungi was found in Erigeron annuus and Mollugo pentaphyll rhizosphere soil, and the highest number of OP-PS fungi was found in rhizosphere soil of Mazus stachydifolius. Mazus stachydifolius showed the strongest PR-PS ability (6340.75μg) while Eragrostis pilosa showed the strongest OP-PS ability (1301.84μg). The PR-PS ability and OP-PS ability of Mollugo pentaphyll was 4432.87μg and 1122.05μg respectively. A significant correlation between the number of PR-PSM and OP-PSM was found. Significant correlation was only found between the PR-PS fungi number and its PSA(r=0.75, P<0.05) and between the number of OP-PS fungi and its PSA(r=0.87, P<0.01}). It indicated that plant species had significant influence on components of the non-symbiotic PSM community and their activity in its rhizosphere soil. Fungi play a leading role in phosphorus solubilization in weed rhizopshere. It suggested that weed conservation could benefit soil microbe development in agroecosystems, especially in the initial stage of agroecosystem development because there is less organic carbon in bare soil. Objective

To investigate stretch-induced electrophysiological changes in chronically infarcted hearts and the effect of streptomycin (SM) on these changes in vivo.

Methods

Sixty Wistar rats were divided randomly into four groups: a control group (n=15), an SM group (n=15), a myocardial infarction (MI) group (n=15), and an MI+SM group (n=15). Chronic MI was obtained by ligating the left anterior descending branch (LAD) of rat hearts for eight weeks. The in vivo blockade of stretch-activated ion channels (SACs) was achieved by intramuscular injection of SM (180 mg/(kg·d)) for seven days after operation. The hearts were stretched for 5 s by occlusion of the aortic arch. Suction electrodes were placed on the anterior wall of left ventricle to record the monophasic action potential (MAP). The effect of stretching was examined by assessing the 90% monophasic action potential duration (MAPD90), premature ventricular beats (PVBs), and ventricular tachycardia (VT).

Results

The MAPD90 decreased during stretching in both the control (from (50.27±5.61) ms to (46.27±4.51) ms, P<0.05) and MI groups (from (65.47±6.38) ms to (57.47±5.76 ms), P<0.01). SM inhibited the decrease in MAPD90 during inflation ((46.27±4.51) ms vs. (49.53±3.52) ms, P<0.05 in normal hearts; (57.47±5.76) ms vs. (61.87±5.33) ms, P<0.05 in MI hearts). The occurrence of PVBs and VT in the MI group increased compared with that in the control group (PVB: 7.93±1.66 vs. 1.80±0.86, P<0.01; VT: 7 vs. 1, P<0.05). SM decreased the occurrence of PVBs in both normal and MI hearts (0.93±0.59 vs. 1.80±0.86 in normal hearts, P<0.05; 5.40±1.18 vs. 7.93±1.66 in MI hearts, P<0.01).

Conclusions

Stretch-induced MAPD90 changes and arrhythmias were observed in chronically infarcted myocardium. The use of SM in vivo decreased the incidence of PVBs but not of VT. This suggests that SACs may be involved in mechanoelectric feedback (MEF), but that there might be other mechanisms involved in causing VT in chronic MI.  相似文献

Objective

To screen mutations in FERM domain-containing protein 7 (FRMD7) gene in two Chinese families with X-linked idiopathic congenital nystagmus (XLICN).

Methods

Common ophthalmic data and peripheral blood of two Chinese XLICN families (families A and B) were collected after informed consent. Genomic DNA was prepared from the peripheral blood of members of the two families and from 100 normal controls. Mutations in the FRMD7 gene were determined by directly sequencing polymerase chain reaction (PCR) products.

Results

We identified a novel mutation c.980_983delATTA compound with c.986C>A mutation in the 11th exon of FRMD7 in family B, and a previously reported splicing mutation c.782G>C (p.R261G) in family A. The mutations were detected in patients and female carriers, while they were absent in other relatives or in the 100 normal controls.

Conclusions

Our results expand the spectrum of FRMD7 mutations in association with XLICN, and further confirm that the mutations of FRMD7 are the underlying molecular mechanism for XLICN.  相似文献

A laboratory incubation experiment was conducted to study the effect of copper as cupric sulfate (CuSO4.5H2O), and lead as lead acetate (Pb (OAc)2) on the size of the microbial biomass in red soil. The metals were applied, separately at six different levels: Cu at 50, 100, 200, 300, 400, and 600 μg·g−1 soil and Pb at 100, 200, 400, 600, 800, and 1000 μg·g−1 soil. In comparison to uncontaminated soil, the microbial biomass carbon (Cmic) and biomass nitrogen (Nmic) decreased sharply in soils contaminated with Cu and Pb. The microbial biomass C:N ratio in the metal contaminated soil was observed to be considerably higher than that in untreated control. 研究目的:探索小分子酪氨酸激酶抑制剂舒尼替尼(sunitinib)对结肠肿瘤微环境中的肿瘤相关成纤维细胞的作用及其机制。创新要点:舒尼替尼通过抑制肿瘤间质成纤维细胞的生长,间接发挥抗肿瘤效应,为结肠癌综合治疗的提供新途径。研究方法:通过细胞周期分析和细胞增殖测定进行舒尼替尼体外抑制肿瘤细胞的研究。采用Western-blot检测磷酸化血小板衍生生长因子β受体(PDGFR-β)、蛋白激酶B(Akt)及细胞外信号调节蛋白激酶(ERK)的蛋白水平。通过注射肠腺癌细胞株SW620和结肠成纤维细胞构建的裸鼠移植瘤模型来研究舒尼替尼的体内抑瘤效果。重要结论:舒尼替尼可有效抑制结肠癌来源的原代结肠成纤维细胞生长,该抑制作用主要通过抑制血小板衍生生长因子(PDGF)信号通路得以实现。  相似文献
