<Emphasis Type="Italic">Ureaplasma urealyticum</Emphasis>-derived lipid-associated membrane proteins introduce IL-6, IL-8, and TNF-α cytokines into human amniotic epithelial cells via Toll-like receptor 2

Objective

The purpose of this study was to determine the role of Ureaplasma urealyticum-derived lipidassociated membrane proteins (LAMPs) in the host innate immune system, specifically their effect on Toll-like receptors (TLRs).

Methods

LAMPs were derived from U. urealyticum strains, and human amniotic epithelial cells (HAECs) were isolated from healthy full-term placentas. Cytokine concentrations were determined by enzyme-linked immunosorbent assay (ELISA) and TLR2 mRNA by real-time PCR. Expression of TLR2 was confirmed by Western blotting and immunohistochemistry.

Results

LAMPs induced HAECs to produce inflammatory cytokines interleukin (IL)-6, IL-8, and tumor necrosis factor (TNF)-α. Cytokine production was reduced after blocking TLR2 using TLR2 inhibitor (anti-hTLR2-IgA).

Conclusions

LAMPs isolated from U. urealyticum induced TLR2-dependent up-regulation of inflammatory genes and cytokines in HAECs.

     

Hyaluronic acid as a rescue therapy for trinitrobenzene sulfonic acid-induced colitis through Cox-2 and PGE2 in a Toll-like receptor 4-dependent way

We hypothesized whether systemic administration of high-molecular-weight hyaluronic acid (HMW HA) could rescue trinitrobenzene sulfonic acid (TNBS)-induced colitis through Toll-like receptor 4 (TLR4) signal. C3H/HeN mice and C3H/HeJ mice were used. Mice were divided into four groups: control, 50% ethanol treatment group, TNBS treatment group, and TNBS plus HA treatment group. The weight changes, clinical scores, macroscopic scores, and histological scores were recorded. Cyclooxygenase 2 (Cox-2) and prostaglandin E₂ (PGE₂) expressions were measured both in colons and peritoneal macrophages from these mice. HA was a rescue therapy for the colitis induced by TNBS only in C3H/HeN mice. The clinical score, macroscopic score, and histological score were much lower in C3H/HeN mice receiving TNBS plus HA treatment. Cox-2 and PGE₂ expressions only increased in C3H/HeN mice. These Cox-2 expressing cells were macrophages. HA can also promote the production of Cox-2 and PGE₂ in peritoneal macrophages from C3H/HeN mice. Our data demonstrated that HMW HA can rescue TNBS-induced colitis through inducing Cox-2 and PGE₂ expressions in a TLR4-dependent way. Macrophages may be the effector cells of HMW HA.

鼻粘膜细胞变应性炎的病理通路及TWH的干预

探讨TLR-NF-кB信号通路及NF-кB不同程度表达在变应性鼻炎(Allergic Rhinitis,AR)发病中的作用及雷公藤多甙(tripterygium wilfor-dii hook,TWH)的干预。应用卵清蛋白建造AR细胞模型,ELISA法测IgE含量,Real-Time PCR及Westernblot检测TLR及NF-кB的表达。结果,模型组IgE、TLR-4及NF-кB的表达较正常组均高;LPS组TLR-4及NF-кB的表达进一步增强,IgE的表达较模型组反而降低;DEX组及TWH组TLR-4及NF-кB、IgE的表达较模型组均降低(P<0.05);TLR4与NF-кB的表达高度相关。结果表明,TLR-NF-кB信号传导通路在AR发病中发挥作用,NF-кB的过高表达抑制IgE的生成,产生免疫偏移作用。     

三元金属间化合物La3Cu3Bi4的合成、结构与性能研究

The new intermetallic compound La3Cu3Bi4 was synthesized by arc melting method followed by annealing.The crystal structure refined using the Rietveld method and X-ray powder diffraction data showed to be Y3Au3Sb4 structure type with space group I43D(No.220).The unit cell parameters were a = 0.997 797(7) nm and V = 0.993 41(2) nm3.The structure was characterized by covalent Cu-Bi4 tetrahedron, which formed three-dimensional network by sharing corners and La atoms separated in the space of Cu-Bi4 tetragon.The expansion of La3Cu3Bi4 studied by using low temperature X-ray di?raction showed to be positive expansion and diminished with the temperature rising.     

Science Letters: A synthetic Toll-like receptor 2 ligand decreases allergic immune responses in a mouse rhinitis model sensitized to mite allergen

It has been proposed that activation of Toll-like receptors (TLRs) plays crucial roles in the polarization of adaptive immune responses. A synthetic Toll-like receptor 2 (TLR2) ligand, Pam3CSK4, has been reported to modulate the balance of Thl/Tn2 responses. We evaluated the modulation effect of Pam3CSK4 on allergic immune response in a mouse rhinitis model sensitized to house dust mite allergen (HDM). Mice were sensitized and challenged with Dermatophagoides farinae allergen (Der f), and then the allergic mice were treated by Pam3CSK4. Nasal allergic symptoms and eosinophils were scored. Der f-specific cytokine responses were examined in the splenocytes and bronchoalveolar lavage fluid (BALF). Serum level of total IgE was also detected. After establishing a mouse allergic rhinitis model with HDM, we have showed that Pam3CSK4 treatment not only ameliorated the nasal allergic symptoms remarkably but also decreased the eosinophils and total inflammation cells in BALF significantly. Analysis of cytokine profile found that' IFN-γ released from either BALF or stimulated splenocytes increased markedly in Pam3CSK4-treated mice, while IL-13 decreased significantly. Moreover, serum level of total IgE was significantly lower in Pam3CSK4-treated mice than in the untreated. Thus, in an allergic rhinitis mouse model developed with HDM, Pam3CSK4 was shown to exhibit an antiallergic effect, indicating its potential application in allergic diseases.

CH_3NHCH_3-BH_3体系的ab initio与DFT研究

用从头算 (abinitio)中的MP2方法和密度泛函理论中的B3LYP ,B3PW91 ,B3P86方法分别研究了CH3 NHCH3 -BH3 体系 ,对各种方法所得的构型、能量、原子净电荷等进行了分析比较 ,发现密度泛函方法对该体系可以得到与MP2相近的结果 .结果表明 ,该体系为强相互作用体系 .     

(La(0.7-x)Y0.3)2/3 Ca1/3MnO3的晶体结构研究

利用标准的固相反应法制备了(La0.7Y0.3)2/3Ca1/3MnO3和La空位(La0.62Y0.3)2/3Ca1/3MnO3锰氧化物粉末样品。X射线衍射(XRD)测量分析表明样品均为单相,并且在300K到30K温度范围内结构没有发生改变,单胞仍然具有正交对称性(Pnma)。并用Rietveld全谱拟合法对X射线衍射测量所得的晶体结构衍射谱进行了拟合。结果表明,晶体结构参数(a,b,c)和晶胞体积(v)随温度发生了明显变化,Mn-O键长随温度减小单调增大,而Mn-O-Mn键角呈单调减小,这也说明存在静态的Jahn-Teller效应。

La空位(La(1-x-y)Yy)2/3Ca1/3MnO3的结构和输运性质研究

用固相反应法制备La（1-x）2／3Ca1／3MnO3（V-LCMO）和（La（0.7-x）Y0.3）2／3Ca1/3MnO3）（V-LY-CMO）（x〈0．15）系列的La空位锰氧化物样品。X射线衍射（XRD）测量分析表明所有的样品均为单相,具有正交对称性（Prima）。体系的晶构参数以及Mn-O键长和Mn-O-Mn键角随空位浓度不同而改变,表明局域Jahn-Teller效应的存在。输运测量表明,不同的空位及空位浓度影响磁电阻效应,在X=0．04时磁电阻值在转变温度‰处达到最大值约为220％,我们认为很大的磁电阻效应与Y部分替代La有关,是双交换作用与Jahn-Teller效应竞争的结果。     

Positive temperature coefficient of resistivity effects of semiconducting (Bi1/2Na1/2) TiO3-CaTiO3-BaTiO3 ceramics sintered in air atmosphere

Y3+-doped (Bi1/2Na1/2)TiO3-CaTiO3-BaTiO3 (BNCBT) positive temperature coefficient of resistivity (PTCR) ceramics sintered in air atmosphere were investigated in this study. (Bi1/2Na1/2) TiO3 (BNT) component can remarkably increase the onset temperature Tc of PTCR ceramics with the expensive of the resistivity R25 increase. CaTiO3 (9-27 mol%) component can decrease the resistivity, and adjust the effects of BNT phase on the Tc point. For the sample containing 3 mol% CaTiO3, the Tc raises from 122° to 153° when only 0.6 mol% BNT added, while for the ones with higher CaTiO3 content (9-27 mol%), Tc is only increased by a rate of 8-9°/1.0 mol% BNT. The effects of BNT and CaTiO3 components on Rmln (negative temperature coefficient effect) are also discussed.     

Interactions between mycoplasma lipid-associated membrane proteins and the host cells

Mycoplamas are a group of wall-less prokaryotes widely distributed in nature, some of which are pathogenic for humans and animals. There are many lipoproteins anchored on the outer face of the plasma membrane, called lipid-associated membrane proteins （LAMPs）. LAMPs are highly antigenic and could undergo phase and size variation, and are recognized by the innate immune system through Toll-like receptors （TLR） 2 and 6, LAMPs can modulate the immune system, and could induce immune cells apoptosis or death. In addition, they may associate with malignant transformation of host cells and are also considered to be cofactors in the progression of AIDS.

新型合成金属3-烷基噻吩共聚物

用FeCl3氧化聚合法和GRIM置换法合成了无规和规整的3-辛基噻吩共3-(2-乙基-1-己基)噻吩共聚物.通过红外、紫外、核磁共振、凝胶色谱和荧光光谱法对其结构和性能进行了分析与表征.结果表明:头-尾相连的规整3-辛基噻吩共3-(2-乙基-1-己基)噻吩共聚物,带隙能较无规共聚物低,且2种共聚物均表现出能垒低、荧光量子效率高、溶解性好和易于加工处理的特点,可以是一类应用于发光二极管、电化学电池和太阳能电池等领域具有广泛前景的聚合物.
