Study of androgen and atherosclerosis in old-age male

Objective: To observe the difference of androgen and inflammatory cytokines level in atherosclerosis and analyse their relations. Method: Both carotid arteries and arteries of lower extremity were subjected to ultrasonic examination by Doppler's method. Those with much atheromatous plaque formation were ranged into case group, and those with normal result formed control group. Total, free testosterone and estradiol were assayed by radioimmunoassay. C reactive protein (CRP) was assayed by nepheloturbidity. Tumor necrosis factor-α (TNF-α), Interleukin-6 (IL-6), Interleukin-8 (IL-8), Interleukin-10 (IL-10), Interleukin- 18 (IL- 18), soluble intercellular adhesion molecule- 1 (sICAM- 1) and soluble vascular cell adhesion molecule- 1 (sVCAM- 1)were assayed by ELISA. The mean difference between two groups and the correlation between free testosterone and cytokines were analysed. Results: Free testosterone was (6.337±3.371) pg/L in case group and (11.375±4.733) pg/L in control group, P＜0.01.No differences were found in total testosterone and estradiol. CRP was (27.294±10.238) mg/L in case group and (12.843±6.318)mg/L in control group, P＜0.01. IL-6 was (41.700±31.385) pg/L in case group and (25.396±20.772) pg/L in control group, P＜0.05.IL-8 was (89.249±58.357) pg/L in case group and (67.873±31.227) pg/L in control group, P＜0.05. sICAM-1 was (470.491±134.078) pg/L in case group and (368.487±97.183) pg/L in control group, P＜0.01. sVCAM-1 was (537.808±213.172)pg/L in case group and (457.275±157.273) pg/L in control group, P＜0.05. There were no differences in TNF-α, IL-10 and IL-18.Correlation analysis showed that FT (free testosterone) had negative correlation with CRP, IL-6 and sICAM-1. Among them FT had well correlation with CRP, correlation index was -0.678. Conclusion: Free testosterone was in negative correlation with atherosclerosis in old-age male. Free testosterone may have the role of anti-atherosclerosis, and this effect was not achieved by its transformation to estradiol. Low free testosterone level was followed by increased level of inflammatory cytokines. Low free testosterones coexist with inflammation and they both affect the process of atherosclerosis in old-age male.

Intestinal microflora in rats with ischemia／reperfusion liver injury

Objectives:To investigate the intestinal microflora status related to ischemia/reperfusion(I/R)liver injury and explorethe possible mechanism.Methods:Specific pathogen free grade Sprague-Dawley rats were randomized into three groups:Controlgroup(n=8),sham group(n=6)and I/R group(n=10).Rats in the control group did not receive any treatment,rats in the I/R groupwere subjected to 20 min of liver ischemia,and rats in the sham group were only subjected to sham operation.Twenty-two hourslater,the rats were sacrificed and liver enzymes and malondialdehyde(MDA),superoxide dismutase(SOD),serum endotoxin,intestinal bacterial counts,intestinal mucosal histology,bacterial translocation to mesenteric lymph nodes,liver,spleen,andkidney were studied.Results:Ischemia/reperfusion increased liver enzymes,MDA,decreased SOD,and was associated withplasma endotoxin elevation in the I/R group campared to those in the sham group.Intestinal Bifidobacteria and Lactobacillidecreased and intestinal Enterobacterium and Enterococcus     

Albumin resuscitation protects against traumatic/hemorrhagic shock-induced lung apoptosis in rats

Objective: To determine the effects of albumin administration on lung injury and apoptosis in traumatic/hemorrhagic shock (T/HS) rats. Methods: Studies were performed on an in vivo model of spontaneously breathing rats with induced T/HS; the rats were subjected to femur fracture, ischemia for 30 min, and reperfusion for 20 min with Ringer’s lactate solution (RS) or 5% (w/v) albumin (ALB), and the left lower lobes of the lungs were resected. Synergistic interaction of NaCl and Cd on growth and photosynthetic parameters in soybean genotypes differing in salinity tolerance

The effects of salinity(50 mmol/L NaCl) and Cd(1 μmol/L CdCl2) as sole and combined on growth and photosyn-thetic parameters were studied using two soybean genotypes,Huachun 18 and NGB.The concentrations of Cd2 ,Zn2 ,Ca2 ,Mg2 ,K and Na were also determined in seeds and pods.Huachun 18 suffered a more serious decrease than NGB in net photosynthetic rate(Pn) in the treatments of salinity stress alone and combined stress(NaCl Cd),showing that it is relatively sensitive to salinity.The decrease in Pn caused by salt stress in Huachun 18 was mainly due to the reduced total chlorophyll content and photosynthetic efficiency(the ratio of variable fluorescence to maximal fluorescence,Fv/Fm),whereas the decease in NGB was mainly related to reduced stomatal conductance(Gs).The combined stress of both Na and Cd did not induce further decrease in photosynthesis and fluorescence in the two genotypes relative to salt or Cd stress alone.Greater change in the pod concentrations of Zn2 ,Ca2 ,Mg2 ,K and Na was detected under salt stress for Huachun 18 than for NGB.The results suggested that the interactive effect of NaCl-Cd on growth and nutrient uptake differs between the two soybean genotypes.

Effect of tramadol on immune responses and nociceptive thresholds in a rat model of incisional pain

Objective: To evaluate the effects oftramadol on the proinflammatory responses in a rat model of incisional pain by investigating its effects on nociceptive thresholds and serum interleukin-6 (IL-6) and IL-2 levels. Methods: Forty-two male Sprague-Dawley (SD) rats scheduled for plantar incision were randomly divided into 7 groups (n=6 in each group). Rats in Group 1 receiving general anesthesia with no incision were served as control; At 30 min before skin incision, Groups 2～5 were given 5 ml normal saline or 1, 10, and 20 mg/kg tramadol, respectively, intraperitoneally (i.p.); Group 6 received 10 mg/kg tramadol after operation; Group 7 received 10 mg/kg tramadol before incision, followed by 200 μg/kg naloxone after operation. Mechanical allodynia was measured by electronic von Frey filament to evaluate the nociceptive thresholds 1 h before incision, and 1 h and 2 h after operation. Serum IL-6 and IL-2 levels were measured by enzyme-linked immunosorbent assay (ELISA) 2 h after operation. Results: Mechanical thresholds decreased significantly and serum IL-6 level increased significantly after operation in Group 2 compared with control (P＜0.01), and these changes were reversed respectively by tramadol in a dose-dependent manner (P＜0.05 and P＜0.01, respectively). IL-2 level remained unchanged after operation in Group 2, but decreased in Group 3 (P＜0.05), then gradually returned to the normal level in Groups 4 and 5. The intraperitoneally injected tramadol (10 and 20 mg/kg) produced a potent and dose-dependent antinocicptive effect on the lesioned paw. The antinocicptive effects of tramadol were partially an-tagonized by naloxone (200 μg/kg), suggesting an additional non-opioid mechanism. Conclusion: The results suggest that tramadol could be a good choice for the treatment of pain under the conditions that immunosuppression may be particularly contraindicated.

Separation of acearylenes from high-temperature coal tar

High-temperature coal tar was extracted with petroleum ether(PE) under ultrasonic irradiation and the extracts were analyzed with gas chromatograph/mass spectrometer.The acearylene fractions including acenaphthylene,aceanthrylene and cyclopentapyrene were enriched together and named E,which was then transferred to a cartridge in Isolera-One flash chromatography.Three groups of compounds were eluted out with ethyl acetate/PE mixed solvent(volume ratio 1:9) and named E 1,E 2 and E 3 according to their main components.Acenaphthylene accounted for 78.2% in E 1,aceanthrylene 71.6% in E 2 and cyclopentapyrene 75.9% in E 3,respectively.The three groups of acearylenes were purified by Sephadex LH-20 column chromatography with ethanol/cyclohexane mixed solvent(volume ratio 1:4),and then confirmed with nuclear magnetic resonance spectrometer.This method indicates that flash chromatography has a good effect on separating the compounds with a similar structure after extraction under ultrasonic irradiation.     

Allograftic bone marrow-derived mesenchymal stem cells transplanted into heart infarcted model of rabbit to renovate infarcted heart

Objective: To investigate the directed transplantation of allograftic bone marrow-derived mesenchymal stem cells (MSCs) in myocardial infarcted (MI) model rabbits. Materials and Methods: Rabbits were divided into 3 groups, heart infarcted model with MSCs transplanted treatment (MSCs group, n=12), heart infarcted model with PBS injection (control group, n=20), sham operation with PBS injection (sham group, n=17). MSCs labelled by BrdUrd were injected into the MI area of the MSCs group. The same volume of PBS was injected into the MI area of the control group and sham group. The mortality, LVIDd, LVIDs and LVEF of the two groups were compared 4 weeks later. Tropomyosin inhibitory component (Tn I) and BrdUrd immunohistochemistry identified the engrafted cells 4 weeks after transplantation. Result: The mortality of the MSCs group was 16.7% (2/12), and remarkably lower than the control group's mortality [35% (7/20) (P<0.05)]. Among the animals that survived for 4 weeks, the LVIDd and LVIDs of the MSCs group

Influence of nitrogen and sulfur fertilization on quality of canola (<Emphasis Type="Italic">Brassica napus</Emphasis> L.) under rainfed conditions

Field experiments were conducted at Cereal Crops Research Institute, Pirsabak, Nowshera, Pakistan, during winter 2003~2004 and 2004~2005 to evaluate the effect of nitrogen and sulfur levels and methods of nitrogen application on canola (Brassica napus L. cv. Bulbul-98) under rainfed conditions. Four levels of S (0, 10, 20, and 30 kg/ha) and three levels of N (40, 60, and 80 kg/ha) and a control treatment with both nutrients at zero level were included in the experiments. Sulfur levels were applied at sowing while N levels were applied by three methods (100% soil application, 90% soil 10% foliar application, and 80% soil 20% foliar application). The experiments were laid out in randomized complete block (RCB) design having four replications. Oil content increased significantly up to 20 kg S/ha but further increase in S level did not enhance oil content. Glucosinolate content increased from 13.6 to 24.6 μmol/g as S rate was increased from 0 to 30 kg/ha. Protein content increased from 22.4% to 23.2% as S rate was increased from 0 to 20 kg/ha. Oil content responded negatively to the increasing N levels. The highest N level resulted in the highest values for protein (23.5%) and glucosinolate (19.9 μmol/g) contents. Methods of N application had no significant impact on any parameters under study.

Maize seed coatings and seedling sprayings with chitosan and hydrogen peroxide: their influence on some phenological and biochemical behaviors

Objective:To evaluate the effect of chitosan(CH) and hydrogen peroxide(H2O2) seed coatings and seedling sprinklings on two different maize varieties by measuring their phenology,the H2O2 presence,the catalase(CAT) activity,and the protein quantity.Methods:Seven groups of ten seeds for each maize variety were treated with CH(2%(20 g/L) and 0.2%(2 g/L)) or H2O2(8 mmol/L) by coating,sprinkling,or both.Germination and seedling growth were measured.One month after germination,the presence of H2O2 in seedlings in the coated seed treatments was evaluated.Protein content and CAT activity were determined under all treatments.Results:H2O2 seed coating enhanced the germination rate and increased seedling and stem length in the quality protein maize(QPM) variety.Seedlings had a higher emergence velocity under this treatment in both varieties.CH and H2O2 sprinklings did not have an effect on seedling phenology.Exogenous application of H2O2 promoted an increase of endogenous H2O2.CH and H2O2 seedling sprinkling increased the protein content in both maize varieties,while there was no significant effect on the CAT activity of treated seeds and seedlings.Conclusions:CH and H2O2 enhance some phenological and biochemical features of maize depending on their method of application.
