蒜薹成熟期石蜡切片方法研究及解剖学观察

建立了一种适合大蒜蒜薹成熟期的石蜡制片方法和鉴别品种办法。对取材、固定、脱水、透明、浸蜡、包埋、切片、染色等重要制片环节进行了研究,得到了适合蒜薹成熟期的石蜡制片方法。蒜薹的最佳制片条件为:在白绿相间处取长0.3mm;切片厚度为10μm;1%番红溶液(50%乙醇配制)染色10 min,固绿0.5%(95%乙醇配制)染色2～3 min。镜下可见蒜薹的基本组织分为皮层和髓部两部分。皮层组织由外向里依次是角质层、表皮、分泌组织和周缘维管束;髓部由薄壁细胞和中央维管束组成。该制片方法可得染色清晰、组织结构完整的蒜薹成熟期的石蜡横切片,其显微特征可作为蒜薹的品种鉴定和质量标准制定的参考依据。     

茉莉叶石蜡制片技术的改进

以茉莉叶片为材料,对石蜡常规制片的部分步骤在时间上作了改进,并对制成的片子在光学显微镜下观察、拍照,以期寻找最佳观察结果和高效的制片方法。实验结果表明。以清洗3次（15min／次）、脱水4次（30min／次）、透明2次（30min／次）、浸蜡2次（120min／次）、脱蜡2次（30min／次）、番红染色50min的细胞结构完整,图像效果最佳,全程耗时624min,与常规石蜡制片的需要时间（2464min）比较,可节省1840min。清洗时间不低于15min／次。脱水时间不低于30min／次,对细胞结构和图像清晰度影响不大。但透明时间低于30min／次、浸蜡时间低于120min／次对细胞结构和图像清晰度有一定影响,若透明时间高于30min／次、浸蜡时间高于120min／次则浪费时间,而且造成组织块变硬变脆,影响制片质量。     

绿竹笋的石蜡切片制作

对绿竹笋组织块进行取材、固定、浸蜡、脱水、包埋、切片、染色等一系列过程的研究，掌握了绿竹笋石蜡切片制作方法，为进一步从解剖学上观察和研究绿竹笋老化机制提供了基础．     

尖叶木犀榄叶片的石蜡切片制作

对尖叶木犀榄叶片进行取材、固定、染色、脱水、透明、浸蜡、包埋、切片、展片、脱蜡、封片等一系列过程的研究,掌握了尖叶木犀榄石蜡切片制作方法,为进一步从解剖学上观察和研究尖叶木犀榄的叶片细胞结构和组成提供了基础.     

制备组织石蜡切片琼脂预包埋法的应用性探讨

为了有效地解决小块组织和易碎裂组织在制备石蜡切片标本中的困难,探索了用于预包埋小块组织和易碎裂组织的琼脂浓度和方法,并用琼脂预包埋法制备的组织石蜡切片做了各种染色.结果表明,使用5%浓度的琼脂预包埋的琼脂块保持完整,且组织未丢失或碎裂.用琼脂预包埋法制备的石蜡切片应用于各种组织学染色的结果与常规的石蜡切片相同,只是组织周围的琼脂略微可见.     

生物组织集中化包埋技术的研究与应用

目的：探讨生物组织集中化包埋技术的可行性。方法：将包埋机和冷冻台改装组合成简易的集中化包埋平台,设计一种带有弹性定位针的集中化脱水包埋盒进行集中化包埋,实现一次性注蜡批量完成组织包埋过程。结果：对于巨检取材大块组织应用集中化包埋技术制作出的蜡块与传统方法制作出的蜡块基本一致,在常规切片、HE染色及免疫组化染色质量上与传统方法无明显差别。结论：对于巨检取材大块组织应用集中化脱水包埋技术能在短时间内制作出较理想的蜡块,工作效率明显提高,且可减少差错事故的发生。     

提高病理切片制作质量的刍议

[目的]提高病理切片制作质量.[方法]通过对制作切片过程中固定、取材、包埋、透明、脱水、浸蜡、切片及染色等步骤易出现的问题研究,以教材常规方法为参考,作者总结实践经验,创新出实用方法并进行分析.[结果]镜下阅片清晰度较原来提高了30%,切片制作后的保存期限比原来提高了3a以上.[结论]制作切片过程中,应注重分析导致病理切片质量低劣的原因,改进常规制作方法,提高切片制作质量,给病理诊断提供正确的技术保障.     

银屑病mRNA原位杂交技术的研究

通过对38例银屑病（Psoriasis）皮肤组织中HB—EGF的表达,比较了核酸原位杂交过程中包括组织固定、脱水、浸蜡、包埋及切片过程中,外源性RNase控制与原位杂交结果之间的关系。结果表明,外源性RNase控制组的阳性信号高于非控制组,其原位杂交结果反映了基因表达的水平。提示在石蜡切片核酸原位杂交实验中,切片制备及保存过程中防止外源性RNA酶污染,是提高结果可信度的关键。     

石蜡切片制作技术中简化染色的尝适

探讨在石蜡切片制作染色中，以电热恒温干燥脱水取代传统工艺中“下行”“上行”酒精逐级梯度脱水。结果提示，恒温干燥脱水染色具有与传统脱水方法相同的效果，工艺简单，无特殊设备要求，重现性好，且具省时间、省人力、省试剂等优点，有重要的应用、推广价值。     

不同组织中PAS染色的特点

PAS染色定位准确,但不同组织中,PAS着色不同,现介绍如下。收集肝组织、肾穿刺组织、含粘液成分组织及含霉菌成分组织各10例。石蜡切片2~4μm,常规脱蜡至水,0.5%高碘酸作用10 m in,无色品红封闭染色5~20 m in,0.5%偏重亚硫酸钠1~2 m in,Mayer苏木素复染20~40 s,1%盐酸酒精脱水片刻,温水返蓝,透明,封片。讨论:组织质地不同,切片厚度和染色时间亦不同。肝组织的糖原成分遇水易分解丢失,因此处理组织时忌过度冲洗,无色品红着色一般3~5 m in,Mayer苏木素复染15~20 s即可。肝脏的切片厚度一般在3μm为宜,组织太厚或太薄均不利于观察细胞内空泡状结构。肾穿刺组织切片一定要薄,厚度控制在2μm,无色品红的染色时间一般在15~20m in之间,Mayer苏木素复染则应在40 s左右。含粘液成分的组织和含霉菌成分的组织切片厚度一般在4μm以内,品红着色5~10 m in,Mayer苏木素复染25~30 s之间即可。(本文编辑:朱佩筠)不同组织中PAS染色的特点@王珏$郧阳医学院形态学实验室!湖北十堰442000$郧阳医学院附属太和医院病理科!湖北十堰442000 @王...
