[目的]通过测定心肌梗死、不稳定心绞痛患者的血清肌钙蛋白I(CTNI)、肌酸激酶(CK)及其同工酶(CK-MB),探讨其对早期心肌损伤性疾病的诊断价值.[方法]CTNI的测定利用BeckmanAccess全自动免疫分析仪,采用免疫散射比浊法;CK-MB的测定利用日本OLYMPUSAU400全自动生化分析仪,采用酶法.测定56例急性心肌梗塞(AMI)患者、43例不稳定性心绞痛(UAP)患者和50例健康体检者血清肌钙蛋白I(CTNI)、CK、CK-MB;并作相关统计学分析.[结果]AMI组血清CTNI明显高于正常对照组(P<0.01),敏感度为92.98%,特异性为97.96%,UAP组血清CTNI亦明显高于正常对照组(P<0.01).[结论]肌钙蛋白I联合检测CK、CK-MB,在早期心肌损伤的诊断、疗效检测方面具有重要意义.  相似文献

目的:检测CD44v6mRNA和HSP70mRNA在胃癌组织中的表达,探讨CD44v6mRNA和HSP70mRNA在胃癌发生发展中的作用.方法:利用核酸原位杂交技术对39例胃癌组织进行检测.结果:CD44v6mRNA和HSP70mRNA在胃癌组织表达的阳性率都为61.54%(24/39),CD44v6mRNA和HSP70mRNA在高分化管状腺癌与低分化管状腺癌中的表达都有显著性差异(P<0.05),但HSP70mRNA在胃癌组织的表达与性别、年龄、浸润深度及淋巴结有无转移都无关(P>0.05),而CD44v6mRNA与淋巴结有无转移有关(P<0.05).结论:CD44v6mRNA和HSP70mRNA在胃癌组织都有较高的表达,两者均可作为评估胃癌预后的指标.  相似文献

目的探讨普通型2019冠状病毒病(COVID-19)患者的临床表现与高分辨率CT(HRCT)的影像特征。方法回顾性分析我院收治的25例COVID-19确诊患者的临床资料及胸部CT检查,进一步分析其临床特征、实验室指标及HRCT影像学特征。结果我院收治的25例COVID-19患者均为普通型,发热(19/25,76%),干咳(18/25,72%);所有患者白细胞计数均正常,淋巴细胞计数不同程度减少(15/25,60%),血沉增快(12/25,48%),C反应蛋白升高(10/25,40%),出现降钙素原降低(2/25,8%);平均住院日为19.8±3.9天,从出现临床症状到CT检查阳性平均时间为4.9±2.7天。首次胸部HRCT病灶分布为单侧9例(36%,左肺4/25,16%;右肺5/25,20%),双侧16例(64%);20例(80%)病灶分布位于周边,5例(20%)位于周边为主及中心同时受累。HRCT表现中单纯磨玻璃影(GGO)为25例(100%),GGO伴局灶性实变为5例(20%),4例(16%)伴片状实变影。在第二次HRCT检查中,大部分患者病灶增多、范围变广,局部病灶融合,并见支气管充气征。结论普通型COVID-19患者临床表现相对较轻,HRCT是其诊断和评估的重要手段。  相似文献

[目的]了解组织蛋白酶-D在大肠癌中的表达.[方法]免疫组织化学S-P法,检测30例大肠癌中组织蛋白酶-D的表达情况.[结果]组织蛋白酶-D在大肠癌中的阳性率为66.67%,它的高表达与大肠癌的淋巴转移和肠外转移成正相关(P<0.05)而与患者的性别、年龄、肿瘤的大体类型、分期和分化程度等无关.[结论]组织蛋白酶-D可作为预测大肠癌复发与转移的重要指标.  相似文献

[目的]新的大肠癌相关性抗原EID3的基因克隆及其诊断价值研究.[方法]利用大肠癌病人体内血清中所含的对肿瘤抗原产生的特异性抗体筛选睾丸组织cDNA噬菌体表达文库和大肠癌组织cDNA噬菌体表达文库(SEREX),并用RT-PCR技术研究EID3 mRNA在正常组织和大肠癌传代细胞表达.[结果]睾丸组织cDNA噬菌体表达文库筛选得到了可以诱导大肠癌病人抗体免疫应答的新抗原EID3基因(Gen-bank NM_001008394.1).它们定位于染色体19q13.2,EID3含1个外显子.通过RT-PCR分析发现,EID3基因在43例大肠癌传代细胞株中,39例阳性,阳性率为90.7%.在正常组织中,除睾丸组织外不表达或有极低水平转录.[结论]EID3 mRNA表达检测用于诊断大肠癌,可能具有高特异性和高敏感性的特点.EID3蛋白被首次发现在大肠癌病人中能够诱导机体的抗体免疫应答,为一个新的大肠癌相关性抗原分子.其功能可能与抑制细胞的恶性增殖相关,并可进一步研究其用于治疗和诊断大肠癌的可行性.  相似文献

目的:构建抑癌基因PTEN的表达载体并在LOVO细胞中表达。方法:从人外周血中提取总RNA,通过RT-PCR扩增出PTEN基因片段,将PTEN基因连入pLenti6/V5载体。测序鉴定后,经脂质体转染入LOVO细胞,对细胞株进行RT-PCR和Western blot检测。结果:DNA测序证实pLenti6/V5-PTEN表达载体为阳性克隆;该表达载体转染LOVO细胞后上调了PTEN mRNA和蛋白的表达。结论:成功构建了pLen-ti6/V5-PTEN表达载体,为进一步研究PTEN在LOVO细胞中的功能奠定了基础。  相似文献

Objectives: To detect the serum proteomic patterns by using SELDI-TOF-MS (surface enhanced laser desorption/ ionization-time of flight-mass spectrometry) technology and CM10 ProteinChip in colorectal cancer (CRC) patients, and to evaluate the significance of the proteomic patterns in the tumour staging of colorectal cancer. Methods: SELDI-TOF-MS and CM10 ProteinChip were used to detect the serum proteomic patterns of 76 patients with colorectal cancer, among them, 10 Stage Ⅰ, 19 Stage Ⅱ, 16 Stage Ⅲ and 31 Stage Ⅳ samples. Different stage models were developed and validated by support vector machines, disctiminant analysis and time-sequence analysis. Results: The Model Ⅰ formed by 6 protein peaks (m/z: 2759.58, 2964.66, 2048.01, 4795.90, 4139.77 and 37761.60) could be used to distinguish local CRC patients (Stage Ⅰ and Stage Ⅱ) from regional CRC patients (Stage Ⅲ) with an accuracy of 86.67% (39/45). The Model Ⅱ formed by 3 protein peaks (m/z: 6885.30, 2058.32 and 8567,75) could be used to distinguish locoregional CRC patients (Stage Ⅰ, Stage Ⅱ and Stage Ⅲ) from systematic CRC patients (Stage IV) With an accuracy of 75.00% (57/76). The Model Ⅲ could distinguish Stage Ⅰ from Stage Ⅱ with an accuracy of 86.21% (25/29). The Model Ⅳ could distinguish Stage Ⅰ from Stage Ⅲ with accuracy of 84.62% (22/26). The Model Ⅴ could distinguish Stage Ⅱ from Stage Ⅲ with accuracy of 85.71% (30/35). The Model Ⅵ could distinguish Stage Ⅱ from Stage Ⅳ with accuracy of 80.00% (40/50). The Model Ⅶ could distinguish Stage Ⅲ from Stage Ⅳ with accuracy of 78.72% (37/47). Different stage groups could be distinguished by the two-dimensional scattered spots figure obviously. Conclusion: This method showed great success in preoperatively determining the colorectal cancer stage of patients.  相似文献

目的探讨孕妇外周血分离胎儿有核红细胞与FISH分析技术在无创性产前诊断中的应用。方法选取于2018年1月至2019年1月期间,20例11~24周妊娠高危孕妇,对外周血均按照常规密度梯度法,对单个核细胞进行分离,并选择红细胞特异性抗体CD71,HbF结合运用流式细胞仪设备行FACS分选富集NRBC,并行FISH分析技术。结果完成20例孕妇的外周血标本NRBC分选,达100%的分选率,其中CD71及HbF双阳性细胞(3.46±1.65)×10-2;对其中18例孕妇运用FISH分析技术均成功分析,未发现任何异常问题。结论经孕妇外周血内分离富集所得胎儿有核红细胞,运用FISH分析技术对胎儿特定染色体进行检测,能够实现无创性产前诊断及时发现遗传性疾病,可为临床改善母婴分娩预后结局提供指导。  相似文献

采用S-P免疫组织化学的方法,配对检测了37例大肠癌组织及同一病人的正常粘膜上皮中p53蛋白的表达情况。结果表明,p53蛋白表达阳性19例,阴性18例,阳性率为51.4%,而配对的大肠正常粘膜检测结果均呈p53阴性;p53蛋白表达与病人性别、年龄、Dukes分期等因素无明显相关性,而与淋巴结转移、浸润深度和不同分化程度等存在明显相关性。提示p53蛋白的检测可以作为大肠癌病人病情诊断和预后分析的辅助手段。  相似文献

The initiators caspase-9(CASP9) and caspase-10(CASP10) are two key controllers of apoptosis and play important roles in carcinogenesis.This study aims to explore the association between CASPs gene polymorphisms and colorectal cancer(CRC) susceptibility in a population-based study.A two-stage designed population-based case-control study was carried out,including a testing set with 300 cases and 296 controls and a validation set with 206 cases and 845 controls.A total of eight tag selected single nucleotide polymorphisms(SNPs) in CASP9 and CASP10 were chosen based on HapMap and the National Center of Biotechnology Information(NCBI) datasets and genotyped by restriction fragment length polymorphism(RFLP) assay.Multivariate logistic regression models were applied to evaluate the association of SNPs with CRC risk.In the first stage,from eight tag SNPs,three polymorphisms rs4646077(odds ratio(OR) AA+AG:0.654,95% confidence interval(CI):0.406-1.055;P=0.082),rs4233532(OR CC:1.667,95% CI:0.967-2.876;OR CT:1.435,95% CI:0.998-2.063;P=0.077),and rs2881930(OR CC:0.263,95% CI:0.095-0.728,P=0.036) showed possible association with CRC risk.However,none of the three SNPs,rs4646077(OR AA+AG:1.233,95% CI:0.903-1.683),rs4233532(OR CC:0.892,95% CI:0.640-1.243;OR CT:1.134,95% CI:0.897-1.433),and rs2881930(OR CC:1.096,95% CI:0.620-1.938;OR CT:1.009,95% CI:0.801-1.271),remained significant with CRC risk in the validation set,even after stratification for different tumor locations(colon or rectum).In addition,never tea drinking was associated with a significantly increased risk of CRC in testing set together with validation set(OR:1.755,95% CI:1.319-2.334).Our results found that polymorphisms of CASP9 and CASP10 genes may not contribute to CRC risk in Chinese population and thereby the large-scale case-control studies might be in consideration.In addition,tea drinking was a protective factor for CRC.  相似文献

利用酶联素和组化法和ABC免疫组化法进行88例消化道恶性肿瘤的雌激素受体(ER)、孕激素受体(PR)和癌胚抗原(CEA)的相关性研究。结果表明:(1)ER、PR水平与组织学分级呈负相关性。(2)低分化腺癌CEA阳性率最高,且淋巴结转移者CEA均为阳性,提示CEA与肿瘤的恶性度呈正相关。因此性激素受体和CEA提供了肿瘤组织不同的生物学信息,同时检测ER、PR及CEA水平对分泌治疗,手术范围及判断预后更有价值。  相似文献

目的:检测子宫内膜异位症患者手术前后外周血T淋巴细胞亚群变化,了解病人细胞免疫功能状况。方法:采用间接免疫荧光法检测子宫内膜异位症患者体内CD3~ 、CD4~ 、CD8~ 细胞水平,并取同龄正常妇女作为对照。结果:(1)手术前与正常妇女相比,子宫内膜异位症患者体内存在CD8~ 细胞比例升高,CD4~ 细胞比例和CD4~ /CD8~ 比值下降;(2)手术后病人CD8~ 细胞逐渐回降,而CD4~ 细胞和CD4~ /CD8~ 比值呈现回升趋势。结论:子宫内膜异位症患者存在T细胞免疫异常,这种异常可在手术治疗后得以恢复。  相似文献

Study on the neurotoxic effects of low-level lead exposure in rats   总被引:10,自引:0,他引:10  
Objective: To investigate effects of developmental lead exposure on nitric oxide synthase (NOS) activity in different brain regions and on N-methyl-D-aspartate (NMDA) receptor mRNA expression in the hippocampus of rats. On the basis of these observations, we explored possible mechanisms by which lead exposure leads to impaired learning and memorizing abilities in children. Methods: A series of rat animal models exposed to low levels of lead during the developing period was established (drinking water containing 0.025%, 0.05% and 0.075% lead acetate). NOS activities in the hippocampus, the cerebral cortex, the cerebellum and the brain stem were determined with fluorescence measurement and levels of mRNA expression of the NMDA receptor 2A (NR2A) subunit and NMDA receptor 2B (NR2B) subunit in the rat hippocampus were measured with Retro-translation (RT-PCR). Results: There were no differences in the body weight of rat pups between any of the groups at any given time (P>0.05). The blood lead level of Pb-exposed rat pups showed a systematic pattern of change: at 14 d of age, it was lower than that at 7 d of age, then rising to the peak level at 21 d and finally falling to lower levels at 28 d. The hippocampal NOS activities of lead-exposed groups were all lower than that of the control group on the 21st and 28th day (P<0.01). NOS activities in the cerebellum of lead-exposed groups were all lower than that of the control group on the 21st and 28th day (P<0.001) and the NOS activity of the 0.025% group was significantly lower than that of the 0.05% and 0.075% groups on the 28th day (P<0.05). NOS activity in the cerebral cortex of the 0.075% group was significantly lower than that of the control, 0.025% and 0.05% groups on the four day spans (P<0.001). There was no significant difference of NOS activity in the brain stem between any lead-exposed group and the control group on the four day spans. In the 0.05% and the 0.075% groups, the level of NR2A mRNA expression was higher than that in the control group at 7 d and 14 d of age (P<0.05). In the 0.025% group, the level of NR2A was found to be higher than that in the control group at 7 d of age only (P<0.05). No significant differences were found for the levels of NR2B mRNA expression between any of the groups at any given time. Conclusions: NOS activity in the hippocampus, the cerebral cortex and the cerebellum are inhibited by lead exposure. The degree of the inhibitory effect depends on the time span of exposure and the lead concentration. Developmental low-level lead exposure was found to raise the level of NR2A mRNA expression in the hippocampus of rats. Developmental low-level lead exposure does not affect the level of NR2B mRNA expression in the hippocampus.  相似文献

Orthotopic liver transplantation (OLT) is the only proven effective treatment for both end-stage and metabolic liver diseases. Hepatocyte transplantation is a promising alternative for OLT, but the lack of available donor livers has hampered its clinical application. Hepatocyte-like cells (HLCs) differentiated from many multi-potential stem cells can help repair damaged liver tissue. Yet almost suitable cells currently identified for human use are difficult to harvest and involve invasive procedures. Recently, a novel mesenchymal stem cell derived from human menstrual blood (MenSC) has been discovered and obtained easily and repeatedly. In this study, we examined whether the MenSCs are able to differentiate into functional HLCs in vitro. After three weeks of incubation in hepatogenic differentiation medium containing hepatocyte growth factor (HGF), fibroblast growth factor-4 (FGF-4), and oncostain M (OSM), cuboidal HLCs were observed, and cells also expressed hepatocyte-specific marker genes including albumin (ALB), α-fetoprotein (AFP), cytokeratin 18/19 (CK18/19), and cytochrome P450 1A1/3A4 (CYP1A1/3A4). Differentiated cells further demonstrated in vitro mature hepatocyte functions such as urea synthesis, glycogen storage, and indocyanine green (ICG) uptake. After intrasplenic transplantation into mice with 2/3 partial hepatectomy, the MenSC-derived HLCs were detected in recipient livers and expressed human ALB protein. We also showed that MenSC-derived HLC transplantation could restore the serum ALB level and significantly suppressed transaminase activity of liver injury animals. In conclusion, MenSCs may serve as an ideal, easily accessible source of material for tissue engineering and cell therapy of liver tissues.  相似文献

Objective:To investigate the relationship between the fatty acid-binding protein 4(FABP4)and colorectal cancer(CRC).Methods:Using an enzyme-linked immunosorbent assay(ELISA),we measured the expression of FABP4 in plasma of50 patients who underwent surgery for CRC from October 2017 to May 2018 and 50 healthy controls.The content of the visceral fat area(VFA)as seen with abdominal computed tomography(CT)scanning was measured by ImageJ software.The expression levels of FABP4,E-cadherin,and Snail proteins in CRC and adjacent tissues were determined by immunohistochemistry.Results:The mean concentration of plasma FABP4 of CRC patients was higher than that of the control group(22.46 vs.9.82 ng/m L;P<0.05).The concentration of plasma FABP4 was related to the tumor,node,metastatis(TNM)stage and lymph node metastasis and was independent of age,body mass index(BMI),tumor size and location,and the degree of differentiation of CRC.The concentration of plasma FABP4 was positively correlated with high VFA and lipoprotein-a(LPA)(P<0.05);but it was not correlated with total cholesterol(TG),total triglyceride(TC),low-density lipoprotein(LDL),high-density lipoprotein(HDL),or apolipoprotein AI(Apo-AI).The expression of FABP4 protein in CRC tissues was positively correlated with the degree of CRC differentiation,tumor stage,and lymph node metastasis.The level of FABP4 protein was negatively correlated with E-cadherin protein(r=-0.3292,P=0.0196)and positively correlated with Snail protein(r=0.5856,P<0.0001).Conclusions:High LPA and VFA were risk factors for increased plasma FABP4 in CRC patients.FABP4 protein was highly expressed in CRC tissues and associated with TNM stage,differentiation,and lymph node metastasis of CRC.The level of FABP4 in CRC tissue was correlated with E-cadherin and Snail expression,suggesting that FABP4 may promote CRC progression related to epithelial-mesenchymal transition(EMT).  相似文献
