成年男性血清碱性磷酸酶活力降低的主要临床意义   总被引:1,自引:0,他引:1  
目的:进一步探讨引起血清碱性磷酸酶活力降低的原因。方法:2001-2005年采用AMP-buffer方法进行了碱性磷酸酶活性测定共2568例。其中126例低于正常,88例为患者,83例为男性。结果:重症肾病并伴有肾衰患者占36.5%,营养不良患者占18%,镁缺乏患者占8.8%,甲状腺机能减退患者占4.8%,严重贫血患者占3%,其余24例(占29.9%)没有与血清ALP活力降低有明显关系的症状。结论:血清ALP活性降低主要见于重症慢性。肾炎并伴有。肾衰、营养不良和甲状腺功能不全、镁缺乏、严重贫血等。血清ALP活性增高或降低均有重要意义。  相似文献

目的:了解燃煤型氟骨症患者血清Ca、P和ALP的变化。方法:分别测定99例燃煤型氟骨症患者和61例非病区健康人群血清Ca、P和ALP水平。结果:氟骨症患者血清Ca(2.12±0.24mmol/L)明显低于健康对照组(2.51±0.14mmol/L),血清P(1.24±0.41mmol/L)和ALP(90.22±29.21U/L)明显高于对照组(分别为1.09±0.28mmol/L和73.64±23.93U/L),差异均具有统计学意义(P<0.01)。结论:在排除其他代谢性骨病的条件下,血清Ca、P和ALP可作为氟骨症辅助诊断的生化指标。  相似文献

目的:观察慢性牙周炎的病人龈沟液(gingival crevicular fluid,GCF)中天冬氨酸转氨酶(aspartate aminotransferase,AST)、碱性磷酸酶(alkaline phosphatase,ALP)及弹性蛋白酶(elastase,EA)在牙周基础治疗结合微波治疗后的变化.方法:用滤纸条的袋内取样法取34例患者一侧104个位点的基础治疗结合微波治疗的GCF样本,同时取对侧104个位点的基础治疗的GCF样本作为对照组,用全自动生化分析仪测定GCF-AST、GCF-ALP,用底物法检测GCF-EA.结果:基础治疗结合微波治疗后的AST、ALP及EA水平较单纯的基础治疗显著下降(P<0.01),各临床指标显著改善.结论:微波治疗可以降低慢性牙周炎病人的GCF-AST、GCF-AST和GCF-EA水平,对牙周炎有确切的治疗作用,可以在临床推广应用.  相似文献

炎症反应已被证明在脑缺血再灌损伤中扮演了重要角色。炎症复合体(inflammasome)是胞浆中参与非特异性免疫的一种多蛋白复合物,其功能主要为识别外来或内源性的危险信号,进而激活含半胱氨酸的天冬氨酸蛋白水解酶-1(cysteinyl aspartate specific proteinase-1,caspase-1),调节炎症因子如:白介素1β(interleukin-1β)、白介素18(IL-18)的表达。目前,在炎症复合体中,研究最广泛的是南嗜中性白细胞碱性磷酸酶-3(neutrophilic alkaline phosphatase-3,NALP3)、凋亡相关点样蛋白(apoptosis—associated speck-like protein,ASC)及caspase-1组成的NALP3炎症复合体。然而,调节其生成和激活的分了机制仍不十分清楚。近期研究显示,被NALP3激活物诱导产生的活性氧(reactive oxygen species,ROS)是活化NALP3炎症复合体必要的二级信号。该文就ROS与NALP3炎痔复合体激活间的关系进行讨论。  相似文献

用多种离子、金属螯合剂、解离剂、酶抑制剂、肝素处理大鼠肝脏碱性磷酸酶,观察大鼠肝脏碱性磷酸酶活性变化,并用胰岛素处理大鼠研究其对酶活性的影响。研究结果表明:1.F^-和高浓度的Zn^2 可完全抑制大鼠肝脏碱性磷酸酶活性,Hg^2 、EDTA和EGTA可在很大程度上抑制该酶活性,而Mg^2 则为其激活剂;2.该同功酶是由多亚基组成的;3.该酶活性中心均含有丝氨酸、天冬氨酸及半胱氨酸;4.胰岛素和肝素均对酶活性有影响。  相似文献

目的:探讨糖皮质激素对肾病综合征患儿血清钙、骨源性碱性磷酸酶的影响,了解患儿成骨细胞功能变化.方法:以血清钙、骨源性碱性磷酸酶为参数,对肾病综合征患儿治疗前及治疗后4周血清钙及骨源性碱性磷酸酶进行动态观察.结果:强的松治疗后血清钙及骨源性碱性磷酸酶与治疗前相比有显著差异,治疗后低于治疗前水平[血清钙(P<0.01)、骨源性碱性磷酸酶(P<0.01)].结论:皮质激素治疗可显著抑制肾病综合征患儿的成骨细胞功能,导致血清钙及碱性磷酸酶水平的降低.  相似文献

碱性磷酸酯[正磷酸单酯磷酸酶(E、C3,1、3、1AKP or ALP)]是机体内一组催化各种磷酸单酯水解的膜结合蛋白。我们从新生牛犊肝、肾、骨、小肠组织中分离与纯化了碱性磷酸酶同工酶,每种组织以正丁醇抽提;水相高速离心分离;透析浓缩;而后进一步用SephadexG-100凝胶过沪;DEAE——纤维素柱层所纯化,得到比活性(S、A)增大30倍和PAGE纯的AKP同工酶。对提纯了的同工酶分别进行了酶活性、Rf值,组织特异性抑制剂、热抑制、尿素和DEAE抑制实验以鉴别每种同工酶的特征。  相似文献

本文报导了目前国内酶制剂工业的主要产品——地衣芽孢杆菌碱性蛋白酶的纯化方法的建立,并从蛋白质工程的需要对2709蛋白酶进行了酶学性质的研究。结果表明,2709蛋白酶具有比活高,热稳定性好,耐碱能力较强等优点。因此地衣芽孢杆菌(Bacillus Licheniformis)2709是一个极具有遗传工程改造潜力的优良菌株,将成为新的蛋白质工程材抖。  相似文献

化学实验室采用离子交换法制备去离子水,制水工艺和树脂的再生条件对出水质量和产水量有明显的影响。该文通过将制水设备中的交换柱全部改为阴阳离子混合柱,同时改进树脂的再生条件,使出水品质和出水量有较大的提高。装置的实际运行效果:出水速率为7 L/h,周期产水量为750 L,水的电导率为0.2-3μs/cm。  相似文献

Comparative study on the activity and kinectic properties of acid phosphatase (ACPase) of three soils amended with Hg and Cu at constant temperature and humidity was carried out. The results indicated that the inhibition on ACPase of the three sample soils by Hg and Cu varied with the content of soil organic matter and pH, where, Soil 1 was the most seriously contami- nated due to its lowest content of organic matter and the lowest pH among three samples, Soil 2 took the second place, and Soil 3 was the least contaminated. Except Soil 3, the activity of soil ACPase tended to increase along with the contact time under the same type and the same concentration of heavy metal. In particular the Vmax values of ACPase in all three samples decreased with increasing Hg and Cu concentration, whereas the Km values were affected weakly. According to the change of Vmax and Km values, Cu and Hg had the same inhibition effect on soil ACPase. Both of them may be a type of compound of non-competitive and anti-competitive inhibition. Statistic analyses indicated that activities of soil ACPase and Vmax values could serve as bioindicator to partially denote the heavy metal Hg and Cu contamination degree.  相似文献

The performance of Al-alloy anode in 4 mol/L KOH with and without stannate and o-aminophenol at 25℃and 55℃was studied by hydrogen collection,potentiodynamic polarization and electrochemical impedance spectrum,o-amino- phenol acts as a perfect inhibitor because of its adsorbability and forming chelate complex at its optimum concentration of 0.4 mol/L.Stannate enhances the inhibition of o-aminophenol and improves the activity of Al-alloy because of its reduction to tin.There is synergetic effect of stannate with o-aminophenol on the behavior of Al-alloy,and the inhibitive efficiency at 55℃is better than that at 25℃.  相似文献

We conducted two-stage acidification-bioleaching experiments to probe the feasibility of bioleaching for a kind of mixed alkaline copper oxide and sulphide mineral. We used the uniform design method for data analysis and experimental optimization, with initial pH value, pulp density, inoculation of bacteria and ferrous iron concentration selected as the influential factors. Polynomial regression shows that the four factors sequentially influence the copper recovery by 14.430%, 8.555%, 1.982% and 3.895%. Acid equilibrium in the bioleaching system is mainly influenced by alkaline gangue content, chemical reactions and bacterial activity. A maximal portion of refractory copper extracted reaches 71.08%. The dynamic analysis of copper recovery indicates that bioleaching goes through a lag leaching phase, prime leaching phase and leaching stationary phase corresponding to the growth phases of bacteria. Compared with the predicted value of 80.87%, the confirmatory experiment observes a 78.21% copper recovery under the optimal conditions of pH of 1.5, pulp density of 5%, bacteria inoculation of 30% and initial ferrous iron concentration of 9 g L^-1. Results suggest that bioleaching is technically feasible to improving total copper recovery.  相似文献

采用碱沉、D101树脂吸附两种精制方法对黄藤酸水提取液进行精制,以所得物中黄藤素的HPLC含量测定为指标,对两种方法进行比较.结果表明:两法有显著性差异,用D101树脂精制的提取率为0.9%,质量分数为58%,纯化所得到的黄藤素的质量较好,含量明显提高而且操作方便,可应用于大规模生产.  相似文献

目的 :本研究拟从人混合血浆中提取MBL并初步鉴定 ,为进一步深入研究MBL的一些特性奠定基础。方法 :取混合血浆通过前处理后经过Mannan -Agarose亲和层析的方法纯化 ,收集含蛋白的洗脱液 ,充分透析、浓缩即得 ,用SDS -PAGE对提取物质进行鉴定。结果 :用纯化后的产物经过SDS -PAGE ,在 30kDa处显示两条谱带 (对应的分子量约为 2 8kDa和32kDa处 ) ,回收率为 2 0 %。结论 :成功用二步亲和层析法从健康人血浆中提纯MBL ,取得了比较满意的结果。  相似文献

Background and objective: Liver regeneration is a complex process regulated by a group of genetic and epigenetic factors. A variety of genetic factors have been reported, whereas few investigations have focused on epigenetic regulation during liver regeneration. In the present study, valproic acid (VPA), a histone deacetylase (HDAC) inhibitor, was used to investigate the effect of HDAC on liver regeneration. Methods: VPA was administered via intraperitoneal injection to 2/3 partially hepatectomized mice to detect hepatocyte proliferation during liver regeneration. The mice were sacrificed, and their liver tissues were harvested at sequential time points from 0 to 168 h after treatment. DNA synthesis was detected via a BrdU assay, and cell proliferation was tested using Ki-67. The expressions of cyclin D1, cyclin E, cyclin dependent kinase 2 (CDK2), and CDK4 were detected by Western blot analysis. Chromatin immunoprecipitation (ChIP) assay was used to examine the recruitment of HDACs to the target promoter regions and the expression of the target gene was detected by Western blot. Results: Immunohistochemical analysis showed that cells positive for BrdU and Ki-67 decreased, and the peak of BrdU was delayed in the VPA-administered mice. Consistently, cyclin D1 expression was also delayed. We identified B-myc as a target gene of HDACs by complementary DNA (cDNA) microarray. The expression of B-myc increased in the VPA-administered mice after hepatectomy (PH). The ChIP assay confirmed the presence of HDACs at the B-myc promoter. Conclusions: HDAC activities are essential for liver regeneration. Inhibiting HDAC activities delays liver regeneration and induces liver cell cycle arrest, thereby causing an anti-proliferative effect on liver regeneration.  相似文献
