Objective: To investigate the immunological function of a yeast expression system for thymosin α1(Tα1). Methods: A constructed Tα1 yeast expression system was used to investigate the immunological function of orally administered Tα1. Dried yeast containing three different concentration of Tα1 was fed to normal Balb/c mice and other Balb/c mice whose immunities were inhibited in advance by cyclophosphamide. Synthesized Tα1 peptide was used as positive control and dried yeast with empty plasmid was used as negative control. CD4 and CD8 levels were detected by flow cytometry assay. TNF-α IFN-γ IL-2, IL-6 and IL-10 levels were detected by liquid chip. Results: In normal Balb/c mice or immune inhibition Balb/c mice, CD8 levels were significantly increased. Especially in immune inhibition Balb/c mice, CD8 levels in synthesized Tα1 group (18.77%±4.72%),small dose group (13.48%±6.17%) and large dose group (22.74%±1.09%) were significantly higher than that in empty yeast control group (7.49%±2.14%). Conclusion: Orally administered Tα1 has its certain immunomodulatory function.  相似文献

The work is intended to achieve optimum culture conditions of α-galactosidase production by a mutant strain ,Aspergillusfoetidus ZU-GI in solid-state fermentation (SSF). Certain fermentation parameters involving moisture content, incubation temperature, cultivation period of seed, inoculum volume, initial pH value, layers of pledget, load size of medium and period of cultivation were investigated separately. The optimal cultivating conditions of α-galactosidase production in SSF were 60% initial moisture of medium, 28 ℃ incubation temperature, 18^h cultivation period of seed, 10% inoculum volume, 5.0-6.0 initial pH of medium, 6 layers of pledget and 10 g dry matter loadage. Under the optimized cultivation conditions, the maximum α-galactosidase production was 2037.51 U/g dry matter near the 144th hour of fermentation.  相似文献

As a part of our interest in biologically active germacranolides from the genus Carpesium (Compositae), we have investigated the constituents of Carpesium cernuum. This paper reports the isolation and structural elucidation of a new germacranolide, cernolide A (Compound 1), from the herb. The structure of Compound 1 was determined as 2α,3β-dihydroxy-9-angeloxygermacra-4-en-6,12-olide on the basis of spectral evidence. The skeleton of Compound 1 was elucidation by IR, MS, 1H and 13C NMR, COSY, HMQC and HMBC experiments. The stereochemistry of Compound 1 was deduced by ROESY spectral data. Finally, the procedures of extraction and isolation were described in detail.  相似文献

To clarify the possible mechanisms of α-anordin and probimane on calmodulin Ca++-Mg++ ATPase system, enzyme dynamic study was carried out by determining three dynamic parameters [the substrate concentration (ATP)-response curve, dose (inhibitors)-response curve and time-response curve]. Our data have shown that the inhibitory rates of α-anordrin and probimane are unrelated to substrate (APT) concentrations, but related to calmodulin concentrations. The inhibition of α-anordrin and probimane is very quick that is completed within 1 to 5 min and can maintain more than 1 hr in the same inhibitory rates. So it is possible that α-anordrin and probimane are calmodulin competitors with calmodulin-like binding whose actions can occur by affecting the reaction balance of substrate and product on target enzymes of calmodulin (Ca++-Mg++-ATPase).  相似文献

This study examined the effects of dietary α-tocopheryl acetate supplementation on antioxidant enzyme activities and fillet quality in commercial-size Sparus macrocephalus. Three hundred fish [main initial weight (350±12) g] were divided into three groups (E250, E500 and El000) and reared in 9 cages. The fish were fed for 8 weeks with three diets containing different levels of dietary α-tocopheryl acetate (289, 553, 1069 mg/kg). Over the experimental period, fish were fed to satiation and reached a final mean weight of (465±28) g without significant body weight difference and proximate composition difference. Fillet α-tocopherol was significantly (P〈0.05) different between groups, reaching levels of 14.2, 22.1, 30.9 pg/mg fillet for groups E250, E500 and El000, respectively. Total serum superoxide dismutase (SOD) activity increased significantly (P〈0.05) in fish fed the diets high in α-tocopheryl acetate, but serum glutathione peroxidase (GPX) activity was unaffected. In storage on ice, fillets of fish fed the diets high in α-tocopheryl acetate exhibited significantly lower (P〈0.05) levels of oxidation. These results suggested that increased dietary α-tocopheryl acetate could increase its flesh deposition, increase the activity of SOD and prevent lipid peroxidation ofSparus macrocephalus fillets in retail storage on ice.  相似文献

An axiomatic definition for the generalized inverse matrix Pade approximation (GMPA) is introduced. The matrix rational approximants are of the form of the matrix-valued numerator and the scalar denominator. By means of generalized inverse for matrices, the ε-algorithm for the computation of GMPA is established. The well-known Wynn identity for GMPA is proved on the basis of ε-algorithm. The η-algorithm is defined in a similar way. The equivalence relation between ε-algorithm and η-algorithm is proposed.
