HPLC法测定不同产地及不同方法炮制后牡丹皮中丹皮酚的含量

采用超声法提取牡丹皮中的有效成分,用高效液相色谱法测定不同产地、不同炮制方法样品中丹皮酚的含量.结果显示,相同色谱条件下,丹皮酚的平均回收率为97.30%, RSD为1.81%(n=6), 4个不同产地中陕西秦岭的丹皮样品中丹皮酚的总含量最高,而河南洛阳样品中丹皮酚的总含量最低,但各地区丹皮酚的总含量均大于1.2%.不同炮制方法提取的丹皮酚含量的比较:醋泡>生品>盐泡>蜜炒>酒泡>炒炭.结论:不同产地丹皮酚的含量有所差别,且不同炮制方法对提取的丹皮酚的含量也有影响,因此要采用合适的炮制方法,减少药效成分的破坏.     

紫外分光光度法测定丹皮酚-羟丙基-β-环糊精的包合率

建立了丹皮酚-羟丙基-β-环糊精的包合率测定方法。采用紫外分光光度法测定丹皮酚-羟丙基-β-环糊精包合物中丹皮酚的含量,由此计算包合率。结果表明:丹皮酚在274 nm处有最大吸收,浓度在10.0~80.0μg/mL范围内与吸光度呈良好的线性关系,回归方程为:A=0.0081c+0.0937(R2=0.999 7),平均回收率为99.81%,RSD为0.32%(n=9)。本方法操作简便易行,具有良好的重现性和精密度。     

RP-HPLC法同时测定金匮肾气片中黄芩苷和丹皮酚的含量

目的:建立同时测定金匮肾气片中黄芩苷和丹皮酚含量的高效液相色谱法。方法:采用C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.1%磷酸水溶液(52:48),检测波长278 nm,柱温30℃,流速1.0 mL/min。结果:黄芩苷在0.485~19.406μg/mL,丹皮酚在1.079~43.157μg/mL线性关系良好。平均回收率(n=9)分别为99.6%,100.5%,含量测定结果表明,不同批次金匮肾气片之间黄芩苷和丹皮酚含量水平差异不大。结论:经方法学验证,该法可用于金匮肾气片中黄芩苷和丹皮酚含量的快速测定。     

丹皮酚曼尼希碱肟衍生物的合成

以丹皮酚为原料,在丹皮酚的3-位/5-位经曼尼希反应引入不同的胺甲基,得到相应的丹皮酚曼尼希碱,再对其1-位酮羰基进行肟化得到相应的目标化合物.目标化合物的结构经过核磁共振氢谱(¹H NMR)和质谱(HR-MS)确证.考察了丹皮酚曼尼希碱合成中投料比及反应温度对反应收率的影响,得到最佳反应条件为：丹皮酚、多聚甲醛与有机胺物质的量之比为3∶4∶4,反应温度为85℃,此条件下丹皮酚曼尼希碱收率可达70.0%～73.4%.     

解热镇痛类药物中乙酰水杨酸含量的红外光谱法测定

建立了红外光谱测定解热镇痛药中乙酰水杨酸含量的方法。选择铁氰化钾为内标物,在乙酰氷杨酸含量为0.53%~5.03%范围内,测得其红外光谱特征峰(1605.3±0.5cm~(-1))和铁氰化钾特征峰(2116.6±0.5cm~(-1))的峰高比(y=hi/hs)与两者质量比(x=mi/ms)有良好的线性关系,其线性回归方程为y=1.2252x+0.0046,线性相关系数r=0.9982,据此测得小儿氨酚匹林片和阿咖酚散样品中乙酰氷杨酸的含量分别为49.6%和44.8%,测定的相对标准偏差分别为0.8%和2.5%,加标回收率分别为97.0%~104%和96.0%~103%,测定结果与药典法相近.     

黄花菜多糖的测定方法研究

研究一种准确测定黄花菜中多糖的方法。采用地衣酚-盐酸法测定黄花菜中多糖含量,以木糖作为标准对照品,检测波长650 nm。结果表明,黄花菜中多糖平均含量为15.82%,平均回收率为98.77%,RSD=2.25%（n=6）。该测定方法操作简便,重现性和稳定性较好,既适用于黄花菜多糖含量的测定,也可为其他戊糖含量高的多糖测定提供一种可行方法。     

丹皮酚Schiff碱的合成及结构表征

以丹皮酚为先导化合物,合成了五种丹皮酚Schiff碱,并采用红外及紫外光谱对其进行了结构表征.这些丹皮酚Schiff碱的合成,丰富了丹皮酚衍生物的种类和性质,为进一步研制开发新的丹皮酚类药物提供理论基础.     

HPLC-荧光法测定中成药中厚朴酚与和厚朴酚含量

建立了一种用高效液相色谱(HPLC)荧光检测法同时测定中成药中厚朴酚与和厚朴酚含量的新方法.使用BDSC18(100mm×4.0mm,i.d.,3μm)色谱柱,流动相为乙腈0.3%-H3PO4(50∶50,V/V),荧光激发波长为220nm,发射波长为380nm.厚朴酚在0.08～246.0μg/mL范围内线性关系良好,检出限为18.4ng/mL,平均回收率为90.2%￣102.7%,日内RSD为0.80%～4.36%,日间RSD为2.18%～5.92%(n=5);和厚朴酚在0.1～231.0μg/mL范围内线性关系良好,检出限为8.7ng/mL,平均回收率为90.0%～98.1%,日内RSD为0.43%～5.53%,日间RSD为3.97%～6.50%(n=5).该法灵敏度高、选择性好、简便、易重复,为香砂养胃丸、藿香正气软胶囊、国公酒中厚朴酚与和厚朴酚的含量测定提供了有效方法.     

氮酮对丹皮酚巴布剂体外透皮吸收的影响

为研究不同浓度的氮酮对丹皮酚巴布剂体外透皮吸收的影响,采用改良Franz扩散池法,液相色谱外标法检测丹皮酚的透过率;在丹皮酚巴布剂基质的制作过程中,分别加入2％、3％、4％的氮酮,检测不同浓度的氮酮对丹皮酚体外透皮吸收的影响;与空白组对照,2％、3％、4％的氮酮对丹皮酚的体外透皮吸收均有显著影响（P ＜0．01,P ＜0．01,P ＜0．01）,对丹皮酚透皮速率提高大小顺序为3％的氮酮＞2％的氮酮＞4％的氮酮;得出氮酮可作为丹皮酚巴布剂的透皮促进剂。     

卡铂壳聚糖/葡甘聚糖复合微球的制备及其性能测试

以液体石蜡作连续相,Span-80作乳化剂,以戊二醛为交联剂,在W/O型乳化体系中制备一种新型微球—卡铂壳聚糖/葡甘聚糖复合微球,探讨了制备工艺条件对微球形状性能的影响;通过显微镜观察,药物包裹率测定,药物释放量检测等分析了微球特性,评价了微球中卡铂的缓释性能.结果表明:制备的复合微球球形良好,球体表面光滑,微球中卡铂的包封率为39.13%,微球中的卡铂在生理盐水中具有一定缓释性能.
