Two components of anticoagulant protein were isolated from the leech Haemendipsa yanyuanensis by heparin agarose affinity chromatography and ultracentrifugation,The determination of anticoagulant activity and characterization analysis of the protein using the method of chromogenic substrate indicates that the anticoagulant protein is thrombin-specific but not factor Xa-specific.The resylts lay a foundation for the research of the anticoaguklant mechanism and application of anticoagulant protein from H.yanguanensis.  相似文献

目的:研究黑乳海参(Holothuria nobilis Selenka)中硫酸化多糖类型、理化性质及抗凝活性。方法:利用化学和物理相结合的分离方法进行纯化,采用色谱、化学、光谱等方法对其纯度、化学组成、分子量、特性黏度、旋光度、凝血活性等进行分析。结果:分离获得F—I、F—II两种多糖组分。F—I由葡萄糖醛酸、乙酰氨基半乳糖、岩藻糖和硫酸基(摩尔比1.00:0.96:1.18:2.99)组成;F-II仅由岩藻糖和硫酸基(摩尔比1.00:0.51);它们的理化性质显著不同,前者的抗凝效价约为后者的6倍。结论:黑乳海参中至少存在两种不同的硫酸化多糖,岩藻糖化糖胺聚糖和α-L-硫酸化岩藻聚糖,均具有强的抗凝血活性。  相似文献

Munchausen by proxy has been reported involving children who have been given various drugs or toxins. In addition, there is a body of adult literature regarding covert anticoagulant ingestion. This is a case of an 11-month-old female who appears to combine features of both of these syndromes. This child presented with an acute left hemorrhagic otitis media. The physical examination was unremarkable except for the following: weight, fifth percentile; left external auditory canal filled with blood with the right external canal and tympanic membrane being normal; and several scattered 1 X 2 cm firm, movable, nontender, purple nodules on extremities, chest and forehead. The coagulation studies were consistent with Vitamin K deficiency secondary to anticoagulant ingestion. A serum warfarin study confirmed our suspicions. The mother was noted to have a dependent relationship with her child and characteristics of those involved in Munchausen by proxy: falsifying information and thwarting medical assessment. In addition, she displayed some of the characteristics found commonly in anticoagulant malingerers. She was depressed, with limited medical knowledge, and had access to warfarin. The mother was admitted for inpatient psychiatric care and the patient placed with an extended family member. This case report describes the use of an anticoagulant to induce illness in a child by a psychologically ill mother. This form of child abuse must be considered in the differential diagnoses of hemorrhagic disorders.  相似文献

目的:分析检验科血液标本出现误差的原因及探讨应对措施。方法:从我院2018年1月-2020年2月所进行过血液生化检验的血样中随机选取165例血液标本作为研究对象。将检验结果进行回顾性分析,研究探讨检验科血液标本出现误差的原因及应对措施。结果:165例血液标本中有23例标本不合格,不合格率为13.9%,导致血液标本不合格的最主要因素是采集操作不当,总共11例,在不合格血液标本中的占比为47.8%,其次为抗凝剂与血液量比例不当,例数为5例,在不合格血液标本中的占比为21.7%,数据差异具有统计学意义(P<0.05)。结论:检验科血液标本出现误差的原因主要有采集操作不当、保存时间过长、抗凝剂与血液量比例不当等,而血液检验在临床具有重要意义。因此,应加强注意影响检验结果准确性的相关因素,提高检验正确率,使患者能够及时得到正确的治疗方式。  相似文献

目的 探索非特异性溃疡性结肠炎(UC)血清学诊断方法。方法 采用DE-52离子交换层折及cNBr-Sepharose 4B亲和层析技术从人结肠粘膜组织中提取一种蛋白组分,经10%SDS-PAGE、IEF、CIE及Westem blot技术,对该蛋白分子量、等电点及免疫学等进行鉴定。结果 产物PI为5.8,分子量为40KD,免疫学鉴定发现其主要与UC患者血清起反应。结论 人结肠40KD蛋白可能是UC的一种自身抗原,血清中ACA可以作为UC的一个特异性指标。  相似文献

SsrA peptide tag from Mycoplasma florum has been developed as a versatile biotechnology tool to control orthogonal degradation of tagged proteins in Escherichia coli. Here, using the systematic deletion mutants of mf-ssrA tag, we demonstrated that the residues in two separate regions have different functions in mf-Lon-mediated specific orthogonal target protein degradation in E. coli. The deletion of multiple residues, up to six amino acids, did not fatally abolish its specific degradation activity, instead of being able to improve the stability of the tagged protein in the presence of endogenous proteases before mf-Lon expression in E. coli. Except for previously identified essential residues, the region adjacent to the C-terminal of the mf-ssrA tag was involved in mf-Lon and endogenous protease-mediated degradation. Moreover, the deletion of specific residues made the mf-ssrA tag more effective and compact. The mf-ssrA tag can be implemented in synthetic biology and bioengineering for development of synthetic circuits.  相似文献

Background and objective

Rivaroxaban is a new oral anticoagulant for stroke prevention in patients with non-valvular atrial fibrillation (NVAF), which has less drug–food interaction than warfarin. We conducted this prospective randomized study to evaluate the metabolic benefits as well as the safety and efficacy with rivaroxaban versus warfarin in patients with NVAF following radiofrequency catheter ablation (RFCA).

Methods

From April to July 2014, 60 patients with NVAF undergoing RFCA were prospectively enrolled in our study. Following RFCA, all patients were randomly assigned to receive rivaroxaban (Group R, n=30) or warfarin (Group W, n=30). Metabolic indices including serum total protein, albumin, globulin, and high-density lipoprotein (HDL) as well as bleeding, stroke, and systemic thromboembolism events were evaluated and compared during follow-up after 15, 30, 60, and 90 d of RFCA procedure.

Results

Serum total protein, albumin, globulin, and HDL levels were all significantly elevated at each follow-up stage in Group R when compared to the baseline (P<0.05 respectively). In Group W, the metabolic indices decreased at first and then had an increasing trend. There were no deaths or thromboembolic complications in each group. The prevalence of total bleeding complications was similar between Group R and Group W (11/30, 36.7% vs. 10/30, 33.3%, P=0.79).

Conclusions

Patients with NVAF receiving rivaroxaban after RFCA procedures appear to benefit from a metabolic perspective compared with warfarin, providing practical clinical reference for the choice of the anticoagulant. Rivaroxaban seems to be as safe and effective in preventing thromboembolic events as warfarin for these patients.

In this paper, we described the symptoms and treatment of a patient with diabetic nephropathy accompanied by spontaneous retroperitoneal hemorrhage after hemodialysis. An elderly female patient with diabetic nephropathy presented with severe pain, numbness, and an increasing swelling in the left hip and left thigh after six sessions of hemodialysis involving the use of an antiplatelet drug and an anticoagulant agent. Her hemoglobin decreased to 46 g/L. An abdominal ultrasound showed a hematoma in the left retroperitoneal space, and computed tomography (CT) findings revealed a 6 cm×8 cm×10 cm hematoma in the left psoas muscle. After aggressive supportive therapy [the administration of packed red blood cell transfusion, carbazochrome sodium sulfonate injection, and continuous venovenous hemofiltration (CVVH)], the patient’s vital signs stabilized and her hemoglobin increased to 86 g/L. Repeat CT showed that the hematoma had been partially absorbed after two weeks. Eventually, the patient was discharged with stable vital signs. Physicians should be aware of the possibility of spontaneous retroperitoneal hemorrhage, particularly in patients with diabetic nephropathy undergoing hemodialysis involving the use of anticoagulant agents.  相似文献

在生物功能研究过程中,通常需要从混合蛋白中区分、检测目标靶蛋白,而蛋白免疫印迹技术能较好的实现该目的.蛋白免疲印迹技术是一种先择性地结合被分离混合物中特异蛋白的分析技术,具有灵敏度高、特异性强、分辨率高、分析容量大,可对蛋白进行定性、定量分析,检测蛋白的时空表达变化情况,被广泛应用于蛋白定性实验技术中.为在大学生创新实验中推广该技术,本教学实验以贵州大学本科生为对象,课程的主要内容包括了解蛋白印迹法的原理、目标蛋白载体的构建、蛋白可溶性表达、PAGE胶制作,以及蛋白质免疫印迹试验.该教学可提高学生对蛋白免疫印迹技术原理和实验操作技能的认识和掌握,同时训练学生在科研中的能力.  相似文献

彩超证实股青肿21例,在下腔静脉滤器保护下行股静脉切开Forgarty导管取栓,配以溶栓、抗凝等治疗。21例手术均获成功,患肢肿胀疼痛消退迅速,无肺动脉栓塞并发症,随访1~12个月,其中18例患肢肿胀消退,恢复正常,3例轻度肿胀。在下腔静脉滤器保护下行股静脉切开Forgarty导管取栓术治疗股青肿具有安全、疗效好、恢复快等优点,滤器宜以临时滤器为首选。  相似文献

根据GenBank数据库中猪圆环病毒Ⅱ型的基因组序列设计引物,采用PCR技术从病料基因组DNA中扩增出PCVⅡ河南地方株ORF4基因,全长180 bp,编码59个氨基酸.将该基因克隆至载体pGEX-4T-3中形成pGEX-4T-3-ORF4表达载体.经PCR、酶切和测序鉴定后,转化表达菌株BL21(DE3)诱导表达.SDS-PAGE结果显示:ORF4能够在大肠杆菌中表达,产物的分子量约为32 kD,且以包涵体形式存在.Western Blot检测结果显示,纯化后的ORF4蛋白能够与鼠抗6&#215;His标签单克隆抗体发生特异性反应,为进一步研究PCVⅡORF4蛋白的特性与功能奠定了基础.  相似文献
