Expression of matrix metalloproteinase-9 in nasopharyngeal carcinoma and association with Epstein-Barr virus infection

Objective: To evaluate the expression of matrix metalloproteinase-9 (MMP9) in nasopharyngeal carcinoma and the association between MMP9 and Epstein-Barr virus infection. Methods: The MMP9 expression was studied by immunohistochemical analysis; and Epstein-Barr virus encoded small nuclear mRNA-1 (EBER-1) produced by in situ hybridization were examined in 41 nasopharyngeal carcinoma sections, and the relation between them, and the associations of MMP9 with clinical features were statistically analyzed. Results: Positive expression rate of MMP9 was 73.17%. The expression of MMP9 showed significant positive correlation with the expression of EBER- 1 (γ=0.483, P=0.001). There was significant association of MMP9 expression with lymph nodes metastasis and clinical stage (P＜0.001), non-significant association with age, gender, pathological classification and T classification. Conclusions: The highly pronounced expression of MMP9 is associated with cervical lymph nodes metastasis. Epstein-Barr virus can enhance NPC metastasis by up-regulating the expression of MMP9.     

Target binding and residence: a new determinant of DNA double-strand break repair pathway choice in CRISPR/Cas9 genome editing

The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)is widely used for targeted genomic and epigenomic modifications and imaging in cells and organisms,and holds tremendous promise in clinical applications.The efficiency and accuracy of the technology are partly determined by the target binding affinity and residence time of Cas9-single-guide RNA(sgRNA)at a given site.However,little attention has been paid to the effect of target binding affinity and residence duration on the repair of Cas9-induced DNA double-strand breaks(DSBs).We propose that the choice of DSB repair pathway may be altered by variation in the binding affinity and residence duration of Cas9-sgRNA at the cleaved target,contributing to significantly heterogeneous mutations in CRISPR/Cas9 genome editing.Here,we discuss the effect of Cas9-sgRNA target binding and residence on the choice of DSB repair pathway in CRISPR/Cas9 genome editing,and the opportunity this presents to optimize Cas9-based technology.     

MMP-9和CD15s与胃癌血管生成的相关性研究及其临床意义

目的:探讨基质金属蛋白酶9(MMP-9)和唾液酸化Lewis X(CD15s)抗原与胃癌侵袭转移的相关性。方法:采用CD105,MMP-9,CD15s进行免疫组化S-P法染色,对47例胃癌根治性切除手术标本病理组织切片进行标记,检测CD105的微血管密度(MVD),并对MVD与MMP-9、CD15s的关系进行统计分析。结果:MMP-9、CD15s表达都呈阳性的胃癌组织微血管密度高于MMP-9和CD15s单阳性及二者均阴性的胃癌组织。结论:MMP-9高表达有利于癌组织局部浸润,CD15s高表达有利于癌细胞血行转移,MMP-9及CD15s检测对临床诊治胃癌及判断预后具有一定意义。     

基于人的视觉系统的小波性能评价

We have constructed a compactly supported biorthogonal wavelet that approximates the modulation transfer function(MTF) of human visual system in the frequency domain.In this paper,we evaluate performance of the constructed wavelet,and compare it with the widely used Daubechies9-7,Daubechies 9-3 and GBCW-9-7 wavelets.The result shows that coding performance of the constructed wavelet is better than Daubechies9-3,and is competitive with Daubechies 9-7 and GBCW-9-7 wavelets.Like Dauechies 9-3 wavelet,the filter coefficients of the constructed waveklet are all dyadic fractions,and the tap is less than Daubechies 9-7 and GBOW 9-7,It has an attractive feature in the realization of discrete wavelet transform.     

苄基取代茚基二价稀土配合物的合成与表征

无水EuCl3与1—苄基茚基钾在四氢呋喃(THF)中以1／2的摩尔比反应，尔后用Na／K合金原位进行还原，得到了一种新的中性的二(1—苄基茚基)稀土Ⅱ配合物(1—PhCH2C9H6)2Eu(THF)2；而用SmI2与1—苄基茚基钾在THF中以1／2的摩尔比反应，合成了一种新的苄基取代茚基稀土二价配合物(1—PhCH2C9H6)2Sm(THE)2，并对其进行了元素分析及红外光谱的表征。     

实验性兔脑血管痉挛血清MMP-9及VEGF的变化研究

观察血清基质金属蛋白酶-9（MMP-9）及血管内皮生长因子（VEGF）在实验性新西兰白兔蛛网膜下腔出血后的浓度变化,探讨它们与脑血管痉挛发生、发展及转归过程的关系,为临床诊断及治疗提供理论依据。实验结果表明血清MMP-9及VEGF的浓度变化可以作为预测蛛网膜下腔出血后脑血管痉挛的发作、判断其发展及预后的参考指标之一,为临床诊断与治疗提供依据。MMP-9及VEGF参与了蛛网膜下腔出血后的病理生理过程,与迟发性脑血管痉挛的发病机制有密切关系。

“Acting Out” in the Case of Parental Violence. Two cases of abuse in infants of less than 3 months is discussed in the light of the mother's own past history. One of the mothers was abandoned in infancy and her foster family did not respect her cultural identity. The second mother identified herself to her own mother. The following mechanisms were involved: child considered as an object rather than as a subject, sadomasochistic mechanisms, compulsion to use child abuse as a defence against anxiety and memories. The therapist initially is treated like the mother's parents by the mother (“acted” relationship). This leads to the proposal of a two-step intervention: first a socioeducative support at home followed by psychotherapy when the mother expresses a wish for it.

Résumé

Nous présentons deux cas cliniques de maltraitance infantile, inscrits dans l'histoire de la mére. Toutefois, l'un se réfère à la non reconnaissance de l'identité culturelle par la famille d'accueil de la mère dans sa petite enfance après abandon maternel, l'autre a rapport à l'identification de la mère à sa propre mère. Nous avons souligné les mécanismes en jeu: non reconnaissance de l'autre comme sujet, persistance d'une relation d'objet partiel renvoyant à un mécanisme de perversion sado-masochiste. Compulsion à la répétition où le passage à l'acte apparaît comme un mode de défense contre l'angoisse, et le souvenir. Sur le plan thérapeutique, nous constations dans un premier temps, la répétition avec le thérapeute de la relation de la mère avec ses propres parents sous forme d'agi. Nous proposons donc une prise en charge en deux temps (aide sociale et éducative à domicile favorisant ensuite une demande psychothérapique).     

Development of motor-life-skills: variations in children at risk for motor difficulties from the toddler age to preschool age

This article explores variations in development of everyday motor-life-skills in 661 children (329 girls and 332 boys) in Norwegian kindergartens of ages 2:9 (T1) and 4:9 (T2) years:months. The particular focus is on children at risk for problems in motor development (the 10% weakest children in the sample). The methodological approach chosen is authentic assessment, applying the Early Years Movement Skills Checklist (EYMSC). All correlations between motor-life-skills at ages 2:9 and 4:9 are statistically significant (p < 0.01), varying between r = 0.26 to 0.38 for the four section scores of EYMSC (Self-help skills, Desk skills, General classroom skills and Recreational and playground skills) and r = 0.39 for the EYMSC total score. The group composition of children assumed to be at risk for motor difficulties changes considerably between ages 2:9 and 4:9. Approximately, two-thirds of the 10% weakest at T1 do not belong to the 10% weakest at T2. Logistic regression failed to identify children at risk at T1 being among the 10% weakest at T2. However, for two sections of EYMSC (Self-help skills; Recreational and Playground skills), it was possible to distinguish between stable and flux groups.     

Adjuvant effects mediated by the carbohydrate recognition domain of <Emphasis Type="Italic">Agrocybe aegerita</Emphasis> lectin interacting with avian influenza H<Subscript>9</Subscript>N<Subscript>2</Subscript> viral surface glycosylated proteins

Objective

To evaluate the potential adjuvant effect of Agrocybe aegerita lectin (AAL), which was isolated from mushroom, against a virulent H₉N₂ strain in vivo and in vitro.

Methods

In trial 1, 50 BALB/c male mice (8 weeks old) were divided into five groups (n=10 each group) which received a subcutaneous injection of inactivated H₉N₂ (control), inactivated H₉N₂+0.2% (w/w) alum, inactivated H₉N₂+0.5 mg recombinant AAL/kg body weight (BW), inactivated H₉N₂+1.0 mg AAL/kg BW, and inactivated H₉N₂+2.5 mg AAL/kg BW, respectively, four times at 7-d intervals. In trial 2, 30 BALB/c male mice (8 weeks old) were divided into three groups (n=10 each group) which received a subcutaneous injection of inactivated H₉N₂ (control), inactivated H₉N₂+2.5 mg recombinant wild-type AAL (AAL-wt)/kg BW, and inactivated H₉N₂+2.5 mg carbohydrate recognition domain (CRD) mutant AAL (AAL-mutR63H)/kg BW, respectively, four times at 7-d intervals. Seven days after the final immunization, serum samples were collected from each group for analysis. Hemagglutination assay, immunogold electron microscope, lectin blotting, and co-immunoprecipitation were used to study the interaction between AAL and H₉N₂ in vitro.

Results

IgG, IgG1, and IgG2a antibody levels were significantly increased in the sera of mice co-immunized with inactivated H₉N₂ and AAL when compared to mice immunized with inactivated H₉N₂ alone. No significant increase of the IgG antibody level was detected in the sera of the mice co-immunized with inactivated H₉N₂ and AAL-mutR63H. Moreover, AAL-wt, but not mutant AAL-mutR63H, adhered to the surface of H₉N₂ virus. The interaction between AAL and the H₉N₂ virus was further demonstrated to be associated with the CRD of AAL binding to the surface glycosylated proteins, hemagglutinin and neuraminidase.

Conclusions

Our findings indicated that AAL could be a safe and effective adjuvant capable of boosting humoral immunity against H₉N₂ viruses in mice through its interaction with the viral surface glycosylated proteins, hemagglutinin and neuraminidase.

     

Correlation of matrix metalloproteinase-2, −9, tissue inhibitor-1 of matrix metalloproteinase and CD44 variant 6 in head and neck cancer metastasis

This study aimed to explore the molecular mechanism in tumor invasion and metastasis. The expression of matrix metalloproteinase-2, −9 (MMP-2, MMP-9), tissue inhibitor-1 of matrix metalloproteinase (TIMP-1), cell adhesion molecule 44 variant 6 (CD44v6), HER2/neu and p53 was investigated in 154 patients with head and neck squamous cell carcinoma (SCC) by ABC and ImmunoMax immunohistochemical method. Their clinical relevance and correlation were analysed. The expression of MMP-2, MMP-9, TIMP-1, CD44v6, HER2/neu and p53 was found in cancer cells in 87.01%, 85.71%, 68.18%, 98.05%, 55.19% and 50.65% cases respectively. Linear regression and correlation analysis revealed that there was close positive relationship (P<0.05) between the expression of MMP-2 and MMP-9, TIMP-1 and CD44v6, HER2/neu and MMP-9, MMP-2 and p53. Up-regulation of MMP-2 was accompanied by advanced T stage (P<0.01). There was also a trend of MMP-2 expression being related with tumor metastasis. Increased expression of HER2/neu was found in patients with tumor recurrence (P<0.05). The expression of TIMP-1 was higher in laryngeal cancer than that in pharyngeal cancer, and higher in keratinizing and non-keratinizing SCC than that in basaloid SCC (P<0.05). These findings suggested that MMP-2 and MMP-9, HER2/neu and MMP-9, MMP-2 and p53 had a coordinate function in aggression of tumor; that MMP-2 had a more important function than MMP-9 in tumor invasion and metastasis; and that HER2/neu might serve as a biomarker for poor prognosis in HNSCC. 衬托型反馈行为迁移现象的研究

有关＂衬托型反馈行为＂（back-channeling）的研究是近年来国外不少学者关注的一项研究课题,但目前在国内鲜见这方面的研究成果。我们从英语专业、日语专业及非外语专业刚入学本科新生中选取27名学生（每专业9名）并让他们用母语（汉语）进行两次自由会话,前后两次会话时间间隔为一年。我们将整个会话过程拍摄成录像并转述成文字后,有t-检验统计方法进行分析,发现这三组学习者交际中Bc行为未见国外学者所发现的显著迁移现象。我们通过仔细研究分析后认为,造成这种现象的原因很可能跟国内的语言教学与学习缺乏国外的习得环境有关,也可能跟课堂教学只重语言知识传授轻语用能力培养有关,当然,还有一个重要的原因,就是Bc行为语用迁移本身的艰难性。     

Visualization analysis of CRISPR/Cas9 gene editing technology studies

The clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) system is an adaptive immune defense system that resists the invasion of viruses and plasmids heterologous genetic material in bacteria and archaea. Taking the literature related to gene editing technology of CRISPR/Cas9 from the Web of Science database from 2002 to 2015, we use the software CiteSpaceV to analyze co-cited literature in order to establish the research hotspots and fronts recently in this field by knowledge mapping.

A new science and engineering career interest survey for middle school students

This study describes the development and validation of a science and engineering (S/E) career interest survey (CIS). This 56 question survey was developed to measure the overall S/E career interests of 7th through 9th grade students. In the CIS, a S/E career is characterized as one which requires the completion of at least a four-year college program with a major in science, science education, or engineering. The CIS is divided into four major parts. In Part I (30 questions), students are expected to select from occupational activities, while in Part II (20 questions) they are to select from various occupations. Part III (5 questions) and Part IV together make up the CIS internal verification scale. The CIS test-retest reliability coefficients for one week and eight months were calculated as 0.96 (n = 57, grades 7–9) and 0.78 (n = 1937, grade 8), respectively. The KR-21 estimate for the CIS was calculated as 0.92. Criterion-related validity coefficients were calculated in two ways: (a) CIS scores were correlated with the Kuder GIS science subscale (r = 0.75, n = 45, grades 7–9), and (b) CIS scores were correlated with a CIS internal verification scale (r = 0.59, n = 127, grades 7–9). Evidence to support the construct validity of the CIS was collected by two methods: (a) for 7–9 grade students (n = 45), the CIS score was found to correlate 0.75 with the scientific subscale and ?0.42 with the artistic sub-scale, of the Kuder GIS. (b) the second method compared the scores of known groups. Test results for students in grades 7-9 (n = 127; n = 1937) showed a statistically significant difference between the scores of boys and girls on S/E career interest. The readability of the CIS was seventh grade level.     

Executive functions in kindergarten children at risk for developmental coordination disorder

Executive functioning (EF) is a key cognitive process for development. Little is known about EF in Kindergarten children at risk for developmental coordination disorder (DCD), despite this age being one of the most critical and intensive period of motor and cognitive development. In our investigation we compared EF in kindergarten children at risk for DCD with Typically Developing (TD) children. Participants were 36 Italian children, 18 at risk for DCD (9 boys and 9 girls) who had a mean age of 4.6 years and 18 TD (9 boys and 9 girls) who had a mean age of 4.6. Executive functions were measured by tasks targeting cold executive functioning (working memory, fluency, inhibitory control) and two assessments of hot executive functioning (Snack Delay and Gift Wrap). Significant differences were found between children at risk for DCD and TD children on cold EF tasks of visuo-spatial working memory abilities, fluency and inhibitory control, but not on hot EF tasks. The findings suggest that it is advisable to implement preschool cognitively challenging physical activities programmes.     

Protective mechanisms of hypaconitine and glycyrrhetinic acid compatibility in oxygen and glucose deprivation injury

This study investigated the protective effect of the compatibility of hypaconitine (HA) and glycyrrhetinic acid (GA) on H9c2 cells under oxygen and glucose deprivation (OGD)-induced injury, and the possible mechanisms. We found that HA+GA significantly improved pathology and morphology of the nucleus and ultrastructure of H9c2 cells under OGD as determined by Hoechst 33342 staining and transmission electron microscopy (TEM) tests. It also reduced the releases of lactate dehydrogenase (LDH), creatine kinase-myocardial band isoenzyme (CK-MB), and aspartate transaminase (AST) from the cultured supernatant of H9c2 cells, which were tested by enzyme-linked immune sorbent assay (ELISA) kits. In addition, it lessened the apoptotic rate as determined by a fluorescein isothiocyanate-annexin V/propidium iodide (FITC-AV/PI) double staining assay. It was also found that HA+GA might regulate the protein expression associated with the phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway. Overall, the study demonstrated that HA+GA protected H9c2 cells against OGD-induced injury, and the signaling mechanism might be related to the PI3K/Akt signaling pathway.
