Determination of 5-Fluorouracil in Human Plasma by High- Performance Liquid Chromatography （HPLC

5-Fluorouracil (5-FU) has a broad spectrum of anti-tumor activity, widely applied to the treatment of cancers. However, it is necessary to determine the plasma concentration of 5-FU in clinical practice due to its narrow therapeutic index. Therefore, a simple, economic and sensitive high-performance liquid chromatography (HPLC) method was developed and validated for the determination of 5-FU in human plasma. Ethyl acetate was chosen as extraction reagent. Chromatographic separation was performed on a Diamonsil C18 column (250 mm × 4.6 mm i.d., 5 μm) with the mobile phase consisting of methanol and 20 mmol/L ammonium formate using a linear gradient elution at a flow rate of 0.8 mL/min. 5-FU and 5-bromouracil (5-BU) were detected by UV detector at 265 nm. The calibration curve was linear over the concentration range of 5—500 ng/mL and the correlation coefficient was not less than 0.992 6 for all calibration curves. The intra- and inter-day precisions were less than 10.5% and 4.3%, respectively, and the accuracy was within ±3.7%. The recovery at all concentration levels was 80.1±8.6%. 5-FU was stable under possible conditions of storing and handling. This method is proved applicable to therapeutic drug monitoring and pharmacokinetic studies of 5-FU in human.     

Loading System for Full-Scale Heavy-Duty Support Node Test with Multi-Directional Loading Requirements

This paper presents the design, analysis and experimental study of a loading system for heavy-duty nodes test based on a large-scale multi-directional in-plane loading device, which has been used in a full-scale heavy-duty support node test. Test loads of the support reached 6 567 kN with multi-directional loading requirements, which outrange the capacity of the available loading devices. Through the reinforcement of a large-scale multi-directional inplane loading device, the innovative design of a self-balanced load transferring device, and other arrangement considerations of the loading system, the test was implemented and the loading capacity of the ring was considerably enlarged. Due to the heavy loading requirements, some checking computations of the ring and the load transferring device outranged the limit of the Chinese national code "Code for Design of Steel Structures (GB 50017—2003)", thus elastic-plastic finite element (FE) analysis was carried out on the two devices, and also the real-time monitoring on the whole loading systems during experiments to ensure test safety. FE analysis and test results show that the loading system worked elastically during experiments.

Effect of the scale of quantitative trait data on the representativeness of a cotton germplasm sub-core collection

A cotton germplasm collection with data for 20 quantitative traits was used to investigate the effect of the scale of quantitative trait data on the representativeness of plant sub-core collections.The relationship between the representativeness of a sub-core collection and two influencing factors,the number of traits and the sampling percentage,was studied.A mixed linear model approach was used to eliminate environmental errors and predict genotypic values of accessions.Sub-core collections were constructed using a least distance stepwise sampling(LDSS) method combining standardized Euclidean distance and an unweighted pair-group method with arithmetic means(UPGMA) cluster method.The mean difference percentage(MD),variance difference percentage(VD),coincidence rate of range(CR),and variable rate of coefficient of variation(VR) served as evaluation parameters.Monte Carlo simulation was conducted to study the relationship among the number of traits,the sampling percentage,and the four evaluation parameters.The results showed that the representativeness of a sub-core collection was affected greatly by the number of traits and the sampling percentage,and that these two influencing factors were closely connected.Increasing the number of traits improved the representativeness of a sub-core collection when the data of genotypic values were used.The change in the genetic diversity of sub-core collections with different sampling percentages showed a linear tendency when the number of traits was small,and a logarithmic tendency when the number of traits was large.However,the change in the genetic diversity of sub-core collections with different numbers of traits always showed a strong logarithmic tendency when the sampling percentage was changing.A CR threshold method based on Monte Carlo simulation is proposed to determine the rational number of traits for a relevant sampling percentage of a sub-core collection.

Experimental Study on Heat Transfer and Pressure Drop of Micro-Sized Tube Heat Exchanger简

A micro-sized tube heat exchanger(MTHE) was fabricated, and its performance in heat transfer and pressure drop was experimentally studied. The single-phase forced convection heat transfer correlation on the sides of the MTHE tubes was proposed and compared with previous experimental data in the Reynolds number range of 500—1 800. The average deviation of the correlation in calculating the Nusselt number was about 6.59%. The entrance effect in the thermal entrance region was discussed. In the same range of Reynolds number, the pressure drop and friction coefficient were found to be considerably higher than those predicted by the conventional correlations. The product of friction factor and Reynolds number was also a constant, but much higher than the conventional.

The mechanism of Fe (Ⅲ)-catalyzed ozonation of phenol

Fe (Ⅲ)-catalyzed ozonation yielded better degradation rate and extent of COD (Chemical Oxygen Demand) or oxalic acid as compared with oxidation by ozone alone. Two parameters with strong effects on the efficiency of ozonation are pH of the solution and the catalyst (Fe3 ) dosage. The existence of a critical pH value determining the catalysis of Fe (Ⅲ) in acid conditions was observed in phenol and oxalic acid systems. The best efficiency of catalysis was obtained at a moderate concentration of the catalyst. A reasonable mechanism of Fe (Ⅲ)-catalyzed ozonation of phenol was obtained based on the results and literature.

Interaction Between DEHP and Particulate in a Eutrophic Lake

Characteristics of interaction between di-2-ethylhexyl phthalate(DEHP) and particulate in a eutrophic lake were studied in this paper. DEHP concentrations ranged from 89.9 to 247 μg/L with an average value of 146 μg/L in subsurface water (SSW) samples, and from 82.0 to 390 μg/L with an average value of 211 μg/L in water surface microlayer (SM) samples. The results indicate that there was only a weak correlation between the DEHP concentrations and suspended particulate material(SPM) concentrations in both SSW and SM,while the significant correlation between DEHP concentrations and chlorophyll a concentrations was found,which suggestes that DEHP was principally bound to phytoplankton in the eutrophic lake. Correlation between DEHP concentrations and total phosphor (TP) concentrations was also found in this investigation. Enrichment factors (EF) of DEHP in SM were in the range of 0.85 to 2.12 with an average value of 1.35..DEHP EFs were significantly related to the enrichment of chlorophyll a in SM. The results suggest that the enrichment of DEHP in SM of this eutrophic lake was mainly due to DEHP accumulation in phytoplankton and was controlled by distribution of phytoplankton between SM and SSW.     

Multi-year monitoring of paddy rice planting area in Northeast China using MODIS time series data

The objective of this study was to investigate the tempo-spatial distribution of paddy rice in Northeast China using moderate resolution imaging spectroradiometer(MODIS) data.We developed an algorithm for detection and estimation of the transplanting and flooding periods of paddy rice with a combination of enhanced vegetation index(EVI) and land surface water index with a central wavelength at 2130 nm(LSWI2130).In two intensive sites in Northeast China,fine resolution satellite imagery was used to validate the performance of the algorithm at pixel and 3×3 pixel window levels,respectively.The commission and omission errors in both of the intensive sites were approximately less than 20%.Based on the algorithm,annual distribution of paddy rice in Northeast China from 2001 to 2009 was mapped and analyzed.The results demonstrated that the MODIS-derived area was highly correlated with published agricultural statistical data with a coefficient of determination(R2) value of 0.847.It also revealed a sharp decline in 2003,especially in the Sanjiang Plain located in the northeast of Heilongjiang Province,due to the oversupply and price decline of rice in 2002.These results suggest that the approaches are available for accurate and reliable monitoring of rice cultivated areas and variation on a large scale.

Development of quality function development matrix to measure the quality management performance of contractor

Quality function development (QFD) matrix was introduced as a tool to measure the quality management performance of contractors. Engineering quality, quality management system components, and their relationship were defined. An integrated engineering quality system was decomposed into seven factors and the quality management system was composed of eight factors. Importance weights of all factors and their relationship point were acquired by questionnaires and interviews. Then, QFD matrix was formulated and the calculating process was proposed. This model was verified on a case study. The result shows that it is useful for contractor in benchmarking themselves and invaluable for owners in the process of deciding contractor.     

Calcium Phosphate Coating on Al2O3 Ceramics by a Biomimetic Method Using Electric Pulse Technique

The preparation of calcium phosphate (CP) coating on alumina ceramics using electric pulse stimulating method has been investigated. The cup-shaped alumina ceramics were soaked in a simulated body fluid ( SBF), and a square pulse potential with frequency of 1 Hz and voltage of 110 V was applied between the inner and outer surfaces of the alumina cup. Surface morphology of CP coatings during different deposition periods was observed by a Philips XL-30 scanning electron microscope (SEM). Compositional analysis was examined by EDAX. The mechanism of nucleation and growth of CP coating was discussed. SEM result indicates that the coating comprises of a large number of tiny needle-like grains and has a porous microstructure. There is a strong bond between the deposited layer and Al2O3 substrate, which may be due to the gentle growth of the biomimetic method. The EDAX analysis indicates that main composition of the coating is calcium and phosphor. The formation of CP coating may be contributed to the stimulation of electric pulse and the high ions concentration which is 1.5 times of the concentration of SBF solution ( 1.5SBF solution). Such surface functionalization method by electric pulse potential can be used to prepare CP coating on various electric-insulating bioinert materials for improving their bioactive character.
