SDS-PAGE技术分析小麦高分子量谷蛋亚基组成的研究

为使十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)技术在分析小麦高分子量谷蛋亚基组成中广泛应用,以及为小麦品质遗传改良亲本选配提供参考依据,该研究利用SDS-PAGE技术对100个小麦骨干亲本及其衍生材料的高分子量谷蛋白亚基组成(HMW-GS)进行了分析,结果表明,在Glu-A1,Glu-B1和Glu-D1 3个位点上分别检测到2种、6种和2种不同的亚基类型;在Glu-A1位点Null亚基出现的频率最高,为82%,其次为100%(Glu-B1),第三点为18%(Glu-D1);在Glu-B1位点7+8亚基出现的频率最高,为56%;在Glu-D1位点2+12亚基出现的频率最高,为86%.共检测到13种HMW-GS组合类型,其中(N,7+8,2+12)和(N,7+9,2+12)出现的频率较高,分别为40%和23%.3个位点具有优质亚基的材料2个,占2%.供试材料具有的优质亚基及优质亚基组合较少,但它们的综合农艺性状较好,所以它们可作为优良亲本质源.SDS-PAGE技术分析小麦高分子量谷蛋亚基组成具有操作简便、快速、微量、价廉、分辨率高、稳定性好等特点.     

小麦谷蛋白亚基提取方法优化及定量研究

确定小麦籽粒中麦谷蛋白亚基含量是分析麦谷蛋白亚基对加工品质贡献的一个重要途径,而麦谷蛋白亚基提取方法对亚基定量影响很大。本研究在尝试多种提取条件后,分析麦谷蛋白亚基提取特性,确认20℃和14h为最大化提取HMW-GS和LMW-GS的最适温度和时间。采取此优化的方法提取188份中国小麦微核心种质的麦谷蛋白亚基,分析麦谷蛋白亚基质量分数与SDS沉降值的相关性,结果显示,HMW-GS、B区的LMW-GS和总麦谷蛋白亚基含量对品质影响最大。结合各品种麦谷蛋白亚基含量情况,初步筛选出钱交麦、内麦11、小偃6号和蚂蚱麦4个优质品种,为小麦品质改良提供参考。     

新引进CIMMYT小麦核心种质籽粒蛋白质含量的分析筛选研究初报

试验以CIMMYT小麦微核心种质164份种质资源为材料,用不破坏籽粒的DA7200方法测定籽粒蛋白质含量,比较分析了164份种质资源蛋白质含量的高低,筛选出一批宝贵的高蛋白和低蛋白的种质资源,以及一些蛋白质含量中等的种质资源.高蛋白的资源用于面包用品种的选育,低蛋白的种质资源用于糕点用品种的选育,蛋白质含量中等的种质资源用于加工馒头和面条用小麦品种的选育.     

国内小麦核心种质籽粒蛋白质含量的分析研究初报

以中科院提供的小麦核心种质210份种质资源为材料,用不破坏籽粒的DA7200方法测定籽粒蛋白质含量,比较分析了210份种质资源蛋白质含量的高低,筛选出一批宝贵的高蛋白和低蛋白的种质资源.高蛋白的资源用于面包用品种的选育,低蛋白的种质资源用于糕点用品种的选育.

小麦麦谷蛋白亚基及其基因的研究进展

作为人类蛋白质的主要来源之一,小麦种子贮藏蛋白质的组成、结构及其功能一直为研究者所关注.随着研究的深入,对于麦谷蛋白基因的染色体定位、结构特点也逐渐为人们所了解.这对于改善小麦籽粒的品质具有非常重要的意义.对近年来小麦麦谷蛋白亚基(高分子量麦谷蛋白亚基HMW-GS和低分子量麦谷蛋白亚基LMW-GS)的提取分离、凝胶电泳(SDS-PAGE)、高效液相色谱(HPLC)、毛细管电泳(CE)等分离技术及其编码基因的染色体定位、多态性变化等方面的研究进展作一简要概述.

33份小麦新品系的产量和品质性状及其相关性分析

以2014—2015年度驻马店市农业科学院小麦研究所选育的33份小麦新品系为材料,以周麦18为产量对照,以豫麦34为优质对照进行比较,研究产量和品质性状及其相互间的关系.结果表明:14个小麦新品系产量比对照增产.小麦新品系的蛋白质和湿面筋含量较高;籽粒产量与蛋白质和湿面筋含量呈显著负相关,与稳定时间等面团流变学特性相关不显著,提高产量可能会使这些品质性状变劣,但产量与多数品质性状呈正相关或微弱的负相关.面团流变学特性和产量性状间无显著相关性,较易同步改良.以千粒质量为切入点,同时协调产量各因素间的关系,有可能在优质和高产上找到平衡点.     

小麦品种品质性状的评价及聚类分析

对102个小麦品种(系)进行了蛋白含量、湿面筋含量、沉降值和籽粒硬度评价,并用最长距离法进行聚类分析.结果表明,强筋小麦占24.5%,中筋小麦占32.4%,弱筋小麦占43.1%.当遗传距离为28.42时,可将102个小麦品种聚为6群.筛选出一批高蛋白质、高硬度、高沉降值的优质资源, 为小麦生产和品质育种提供种质基础.
