The effect of sodium pentosan polysulphate (SPP) in calcium oxalate stone forming rats was studied in relation to enzymatic changes in liver. A significant increase in liver glycollate oxidase (GAO) activity was observed in stone forming rats fed sodium glycollate. SPP treatment lowered the enzyme acitivity in both stone formers and 30 days drug treated control rats. Moderate elevation in LDH activity was seen in the calculogenic group and SPP had minimal effect. The lowering of alkaline and acid phosphatase activities in stone formers was normalised with drug administration. Increases in total, Na+, K+-and Ca2+-ATPase levels in the calculogenic rats was lowered considerably with SPP treatment. Inorganic pyrophosphatase and aminotransferases were slightly reduced in glycollate-fed rats. SPP administration further lowered the pyrophosphatase level. The decrease in liver GAO during SPP administration with a consequent reduction in kidney oxalate may prove beneficial in preventing recurrence.  相似文献

Changes in erythrocyte membrane glycoproteins during, leukaemia were investigated in one hundred patients. Control group contained normal ones. The total protein and protein bound total carbohydrates (neutral sugars) in the glycoproteins extracted from the erythrocyte membrane showed significant reduction in total protein and protein bound total carbohydrates before radiotherapy. However, after the radiotherapy there was further reduction in total protein by 10.41% whereas there was increase in total carbohydrates by 9.98%. Qualitative analysis reveals that with the help of lectins one can pin point the sugars which appear or disappear due to leukaemia. The test could be of a diagnostic value.  相似文献

The effect of DL α-lipoic acid, a potent antioxidant was studied in relation to certain erythrocyte membrane parameters in calcium oxalate stone forming rats. Induction of calcium oxalate lithiasis was done by feeding a diet containing 3% w/w sodium glycollate. Erythrocyte membrane (Na+, K+)-ATPase showed a significant decrease in stone formers whereas (Ca2+)-ATPase showed a significant increase. Lipoic acid administration brought about an elevation in the activity of (Ca2+)-ATPase. Changes in membrane (Mg2+)-ATPase was minimal. Membrane cholesterol and phospholipids were found raised significantly in lithogenic rats. The changes may be attributed to enhanced lipid peroxidative mechanisms and altered serum lipid profile observed in this group. Treatment with lipoic acid reduced membrane cholesterol levels. Phospholipids were also decreased moderately. The above observations suggest that lipoic acid administration to calculogenic rats reduces the erythrocyte strucutral changes observed in this condition.  相似文献

This study suggests a new erythrocyte sedimentation rate (ESR) measurement method for the biophysical assessment of blood by using a microfluidic device. For an effective ESR measurement, a disposable syringe filled with blood is turned upside down and aligned at 180° with respect to gravitational direction. When the blood sample is delivered into the microfluidic device from the top position of the syringe, the hematocrit of blood flowing in the microfluidic channel decreases because the red blood cell-depleted region is increased from the top region of the syringe. The variation of hematocrit is evaluated by consecutively capturing images and conducting digital image processing technique for 10 min. The dynamic variation of ESR is quantitatively evaluated using two representative parameters, namely, time constant (λ) and ESR-area (AESR). To check the performance of the proposed method, blood samples with various ESR values are prepared by adding different concentrations of dextran solution. λ and AESR are quantitatively evaluated by using the proposed method and a conventional method, respectively. The proposed method can be used to measure ESR with superior reliability, compared with the conventional method. The proposed method can also be used to quantify ESR of blood collected from malaria-infected mouse under in vivo condition. To indirectly compare with the results obtained by the proposed method, the viscosity and velocity of the blood are measured using the microfluidic device. As a result, the biophysical properties, including ESR and viscosity of blood, are significantly influenced by the parasitemia level. These experimental demonstrations support the notion that the proposed method is capable of effectively monitoring the biophysical properties of blood.  相似文献

Erythrocyte membrane protein glycosylation increase by 3.4 fold in diabetes. Insulin or sulfonylurea treatment did not reduce the extent of glycosylation. The serum protein glycosylation was comparable in all the groups including control. Erythrocyte membrane Na+,K+-ATPase activity decreased in the diabetics; only insulin treatment partly restored the activity. Erythrocyte membrane acetylcholinesterase activity decreased only in the sulfonylurea treated group. Serum butyrylcholinesterase activity was relatively low in the diabetic and insulin treated diabetic groups. The Km and Vmax of the two components of Na+,K+-ATPase from erythrocyte membranes were differently affected in the diabetic and the two treatment groups. The Vmax of acetylcholinesterase decreased only in the sulfonylurea treated group. Diabetic states resulted in decreased Vmax of components I and II of serum butyrylcholinesterase. In insulin-treated diabetics, component II was absent. Sulfonylurea group resembled diabetics.In vitro incubation with insulin differentially affected the Na+,K+-ATPase and serum butyrylcholinesterase activities.  相似文献

IntroductionIndirect estimation of reference intervals (RIs) is straightforward and inexpensive procedure for determination of intra-laboratory RIs. We applied the indirect approach to assess RIs for haematological parameters in capillary blood of pre-school children, using results stored in our laboratory database.Materials and methodsWe extracted data from laboratory information system, for the results obtained by automatic haematology analyser in capillary blood of 154 boys and 146 girls during pre-school medical examination. Data distribution was tested, and logarithmic transformation was applied if needed. Reference intervals were calculated by the nonparametric percentile method.ResultsReference intervals were calculated for: RBC count (4.2-5.4 x1012/L), haemoglobin (114-146 g/L), MCH (25.0-29.4 pg), MCHC (321-368 g/L), RDW-SD (36.1-43.5 fL), WBC count (4.5-12.3 x109/L), neutrophils count (1.7-6.9 x109/L) and percentage (29.0-69.0%), lymphocytes count (1.6-4.4 x109/L) and percentage (21.9-60.7%), PLT (165-459 x109/L), MPV (8.1-11.4 fL) and PDW (9.2-14.4%). Gender specific RIs were calculated for monocytes count (male (M): 0.2-1.6 x109/L; female (F): 0.1-1.4 x109/L) and percentage (M: 2.5-18.3%; F: 1.8-16.7%), haematocrit (M: 0.34-0.42 L/L; F: 0.34-0.43 L/L), MCV (M: 73.4-84.6 fL; F: 75.5-84.2 fL) and RDW (M: 12.1-14.3%; F: 11.7-13.9%), due to observed gender differences in these parameters (P = 0.031, 0.028, 0.020, 0.012 and 0.001; respectively). Estimated RIs markedly varied from the literature based RIs that are used in the laboratory.ConclusionsIndirect method employed in this study enables straightforward assessment of RIs in pre-school children. Herein derived RIs differed from the literature-based ones, indicating the need for intra-laboratory determination of RIs for specific populations and sample types.  相似文献

Objective  To investigate the protective effect of nicorandil and amlodipine in isoproterenol induced myocardial necrosis in rats. Methods  The effect of nicorandil and amlodipine on bio-chemical parameters during isoproterenol induced myocardial necrosis in rats was examined by determining the activity of creatine phosphokinase (CK), lactate dehydrogenase (LDH), and transaminases (AST, ALT) in the serum of the animals. Results  Isoproterenol (150mg/kg/day) administered rats showed statistically significant rise in activities of LDH(1.02±0.19 to 1.85±0.05), CK(4.3±0.19 to 7.37±.27), AST(0.38±.03 to 0.78±.05) and ALT(0.19±.017 to 0.346±.027) in the serum. Pre-co-treatment of rats with nicorandil and amlodipine significantly lowered the raised levels of these enzymes and thereby restoring the enzyme activity to near normal as was clear from the chart i.e. LDH was 1.10±0.04, CK was 4.37±0.19, AST and ALT were 0.39±0.028 and 0.199±0.04 respectively. Conclusion  It is demonstrated that pre-co-treatment with nicorandil and amlodipine either alone or in combination help in providing protective effect on isoproterenol induced myocardial necrosis.  相似文献

The aim of the present work is to understand the lipid peroxidation of RBC membrane and the spectrin protein content of RBC membrane cytoskeleton of thalassaemic carrier state (trait) of β and hemoglobin E variant (HbE). We have measured the hemoglobin (Hb), malondialdehyde (MDA) and spectrin content of RBC membrane of thalassaemic carrier. The spectrin content (α and β band) of both β and HbE carrier was not changed than normal individuals. However, lipid peroxidation of RBC membrane was significantly increased in both β and HbE trait, and Hb level was also decreased in thalassaemic carrier. It may be assumed that oxidative damage by excess lipid peroxidation may have no role on irreversible membrane damage in β thalassaemia and HbE thalassaemia carrier.  相似文献

Red blood cells (RBCs) are important to dictate hemorheological properties of blood. The shear-thinning effect of blood is mainly attributed to the characteristics of the RBCs. Variations in hemorheological properties alter flow resistance and wall shear stress in blood vessels. Therefore, detailed understanding of the relationship between the hemorheological and hemodynamic properties is of great importance. In this study, blood viscosity and blood flow were simultaneously measured in the same microfluidic device by monitoring the flow-switching phenomenon. To investigate blood flows according to hemorheological variations, the flow rate of blood samples (RBCs suspended in autologous plasma, dextran-treated plasma, and in phosphate buffered saline solution) was precisely controlled with a syringe pump. Velocity profiles of blood flows were measured by using a micro-particle image velocimetry technique. The shape of velocity profiles was quantified by using a curve-fitting equation. It is found that the shape of the velocity profiles is highly correlated with blood viscosity. To demonstrate the relationship under ex vivo conditions, biophysical properties and velocity profiles were measured in an extracorporeal rat bypass loop. Experimental results show that increased blood viscosity seems to induce blunt velocity profile with high velocity component at the wall of the microchannel. Simultaneous measurement of blood viscosity and velocity profile would be useful for understanding the effects of hemorheological features on the hemodynamic characteristics in capillary blood vessels.  相似文献

Introduction:

Today, the pneumatic tube transport system (PTS) is used frequently because of its advantages related to timing and speed. However, the impact of various types of PTS on blood components is unknown. The aim of this study was to examine the influence of PTS on the quality of routine blood cell counts, erythrocyte sedimentation, and certain blood coagulation tests.

Materials and methods:

Paired blood samples were obtained from each of 45 human volunteers and evaluated by blood cell count, erythrocyte sedimentation, and several coagulation tests, including prothrombin time (PT) and activated partial thromboplastin time (aPTT). Blood samples were divided into 2 groups: Samples from group 1 were transported to the laboratory via the PTS, and samples from group 2 were transported to the laboratory manually. Both groups were evaluated immediately by the tests listed above.

Results:

The blood sample test results from groups 1 and 2 were evaluated and compared. No statistically significant differences were observed (P = 0.069–0.977).

Conclusion:

The PTS yielded no observable effects on blood cell counts, erythrocyte sedimentation, or PT and aPTT test results. We concluded that the PTS can be used to transport blood samples and yield reliable results for blood cell counts, erythrocyte sedimentation, and several coagulation tests.  相似文献

Cryopreservation of human red blood cells (RBCs) in the presence of 40% glycerol allows a shelf-life of 10 years, as opposed to only 6 weeks for refrigerated RBCs. Nonetheless, cryopreserved blood is rarely used in clinical therapy, in part because of the requirement for a time-consuming (∼1 h) post-thaw wash process to remove glycerol before the product can be used for transfusion. The current deglycerolization process involves a series of saline washes in an automated centrifuge, which gradually removes glycerol from the cells in order to prevent osmotic damage. We recently demonstrated that glycerol can be extracted in as little as 3 min without excessive osmotic damage if the composition of the extracellular solution is precisely controlled. Here, we explore the potential for carrying out rapid glycerol extraction using a membrane-based microfluidic device, with the ultimate goal of enabling inline washing of cryopreserved blood. To assist in experimental design and device optimization, we developed a mass transfer model that allows prediction of glycerol removal, as well as the resulting cell volume changes. Experimental measurements of solution composition and hemolysis at the device outlet are in reasonable agreement with model predictions, and our results demonstrate that it is possible to reduce the glycerol concentration by more than 50% in a single device without excessive hemolysis. Based on these promising results, we present a design for a multistage process that is predicted to safely remove glycerol from cryopreserved blood in less than 3 min.  相似文献

This study was carried out to see the effect of the aqueous extract ofOcitum sanctum Linn (Tulsi) with Vitamin E on biochemical parameters and retinopathy in the streptozotocin-induced diabetic albino male rats. Adult albino male rats weighing 150–200 gm were made diabetic by intraperitoneal injection of streptozotocin in the dose 60 mg/kg in citrate buffer (pH 6.3). The diabetic animals were left for one month to develop retinopathy. Biochemical parameters like plasma glucose, oral glucose tolerance and glycosylated hemoglobin HbA1c, were measured along with lipid profile, and enzymes like glutathione peroxidase (GPX), lipid peroxidase (LPO), superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST) in normal, untreated diabetic rats and diabetic rats treated withOcimum sanctum L extracts and vitamin E. Fluorescein angiography test was done for assessing retinopathy. Results on biochemical parameters were analyzed statistically by using ANOVA followed by Dunnet's ‘t’-test. A p-value of <0.05 was considered as significant. Evaluation of biochemical profile in treated groups showed statistically significant reduction in plasma levels of glucose, HbA1c, lipid profile and LPO, and elevation of GPX, SOD, CAT and GST. Treatment of the diabetic animals withOcimum sanctum and Vitamin E, alone and in combination for 16 weeks showed reversal of most of the parameters studied including plasma glucose levels. Angiography showed improvement in retinal changes following combined antidiabetic treatment.  相似文献

We report an interesting buffer electric relaxation time tuning technique, coupled with a glutaraldehyde cross-linking cell fixation reaction, which allows for sensitive dielectrophoretic analysis and discrimination of bovine red blood cell (bRBC) starvation age. The buffer composition is selected such that two easily accessible dielectrophoretic crossover frequencies (cof) exist. Low concentration glutaraldehyde fixation was observed to produce a threefold decrease in the higher cof with a comparable increase in the lower cof also witnessed. More importantly, increased glutaraldehyde fixation concentration significantly increased the higher cof by a factor found to be sensitive to the bRBC starvation age.  相似文献

Studies were carried out to examine and compare the effects of alloxan-diabetes on reactive oxygen species (ROS) related parameters in the heart from male and female rats. Effects of insulin treatment were also evaluated. The diabetic state severely compromised the ROS defense mechanism in the cardiac tissue and the effects were more pronounced in the female than in the male rats. There was several fold increase in the xanthine oxidase (XO) activity in general and the magnitude of increase was higher in the females; insulin treatment resulted in further increase in the XO activity. The glucose-6-phosphate dehydrogenase (G6PDH) and catalase activities decreased and the reduced glutathione (GSH) content in mitochondria was completely depleted in diabetic state with significant decrease in the GSH levels in the post-mitochondrial fraction; the effect was more pronounced in the females. The superoxide dismutase (SOD) and glutathione peroxidase (GPox) activities increased in the diabetic state to a greater extent in male rats. Insulin treatment had restorative action only on some parameters. In conclusion, our results suggest that diabetic state may further compromise the weak ROS defense systems in the heart thus initiating a lesion at the level of mitochondria which ultimately leads to cardiomyopathy and the effects are especially more pronounced in the females. Our results also pointed out that insulin treatment was ineffective in restoring ROS related parameters.  相似文献

Plasma and erythrocyte magnesium levels were estimated in preeclampsia and eclampsia. Mean plasma as well as erythrocyte magnesium concentrations were found to be significantly lowered than the controls. Hypomagnesemia was directly related with the severity of the disease. The present study has investigated the levels of lipids, lipoprotein cholesterol (HDL and LDL cholesterol), thiobarbituric acid reactive substances (TBARS) and vitamin E in plasma and erythrocyte membranes of twenty two clinically diagnosed adult rheumatoid arthritis patients and an equal number of age matched healthy subjects. The levels of lipids and lipoprotein cholesterol were markedly reduced in patients with rheumatoid arthritis as compared to healthy subjects. The altered lipid pattern may be related to decreased lipoprotein cholesterol, fatty acids and impairment in antioxidant defence mechanism.  相似文献

The effect of oral administration of vitamin E for twenty-eight consecutive days on blood glucose, reduced glutathione levels, antioxidant enzymes (superoxide dismutase and catalase activities), and levels of malondialdehyde (as an index of free radical-mediated lipid peroxidation) was observed in the whole blood and liver of normal and alloxan-induced diabetic rats. It was found that oral administration of vitamin E significantly (p<0.05) lowered the blood glucose level and increased the body weight of the diabetic rats. The activities of superoxide dismutase and levels of reduced glutathione increased significantly (p<0.05) while the level of lipid peroxidation decreased.  相似文献

目的:探讨疏筋活血汤加减方治疗腰腿痛气滞血瘀证的临床疗效研究。方法:腰腿痛气滞血瘀证患者126例,将其随机分为观察组68例和对照组58例。对照组口服双氯芬酸钠双释放肠溶胶囊,观察组内服中药疏筋活血汤加减方,并将药渣敷于患处。观察指标为临床疗效评价,VAS评分,M-JOA评分,ODI评分,IL-6和NO,血液流变学。结果:患者经治疗后观察指标具有显著变化,临床总有效率高于对照组;VAS评分、ODI评分明显降低,M-JOA评分升高IL-6、NO和血液流变学均下降,观察组变化幅度明显高于对照组,有统计学意义(P0.05)。结论:疏筋活血汤加减方治疗腰腿痛气滞血瘀证能有效缓解患者的临床症状,改善患者腰腿部功能,疗效满意。  相似文献

本文利用中国1978~2008年30个时间序列数据,通过反映产业结构变动的斯托可夫指数探讨上海产业结构变动对失业率的影响.研究结果表明,在过去的30年中,虽然上海第二产业比第三产业更能对就业提供有利的带动作用,但上海市第三产业在全行业中占有举足轻重的作用,未来能为劳动力就业带来更大的发展空间,因此要有的放矢地发展和改善第三产业尤其是服务业的产业结构,使之发挥更大的社会的贡献.  相似文献
