液-质联用在西罗莫司和依维莫司血药浓度监测中的应用进展

西罗莫司和依维莫司是一种新型的免疫抑制剂,相对于环孢霉素(CsA),西罗莫司和依维莫司服用方便、吸收好、起效快,但也存在着用药剂量小、个体吸收差异大、治疗窗窄.因此,测定个体血药浓度以调整用药的剂量和次数、制定个体化治疗方案在临床上有着至关重要的意义.本文就近几年液-质联用在西罗莫司和依维莫司血药浓度监测中的应用研究作以简要综述,以期为临床合理用药提供参考.

桃红四物汤水提液中总酚酸含量测定

目的:测定桃红四物汤水提液中总酚酸(TPA)的含量.方法:总酚酸的含量测定采用可见分光光度法,以没食子酸为对照品,0.6%FeCl3～0.9%K3\(1∶0.9)混合液为显色剂,在720nm波长处进行测定.结果:分光光度法测定总酚酸在所试浓度范围内(0.1756～0.8780μg/mL)线性关系良好,r=0.9968.总酚酸的平均回收率为99.96%,RSD为4.1%. 结论:本法简单快捷,重现性好,可作为桃红四物汤中总酚酸的质控方法.

DCD肾移植中DGF高风险受者应用EC-MPS联合低剂量他克莫司治疗的预后分析（英文）

目的:分析心脏死亡供体(DCD)肾移植中移植物功能延迟恢复(DGF)高风险受者应用米芙(EC-MPS)联合低剂量他克莫司治疗术后1年的有效性及安全性,指导临床用药。创新点:对比DGF高风险受者术后发生DGF及正常恢复受者的免疫抑制剂药代动力学特征。方法:将本中心进行肾移植的61例DGF高风险受者按照实际病情纳入DGF组及正常恢复组,均行米芙联合低剂量他克莫司免疫抑制治疗。对比两组免疫抑制剂血药浓度及预后各项指标。结论:DGF组及正常恢复组间米芙及他克莫司剂量无显著差异。术后1周DGF组EC-MPS血药浓度曲线下面积小于30(mg·h)/L的比率显著高于正常恢复组,同时他克莫司谷浓度显著低于正常恢复组,DGF组经活检证实的急排反应的发生率显著高于正常恢复组,术后1周的EC-MPS血药浓度曲线下面积低于30(mg·h)/L的受者中经活检证实的急排反应的发生率显著高于其他受者。1年移植物存活率、移植物功能及常见不良反应在两组间未见明显差异。     

高效液相色谱法测定人血浆中氯氮平浓度

目的建立测定人血浆中氯氮平浓度的高效液相色谱法。方法以DiamonsilTM C18反相柱(150mm×4.6mm,5μm)为色谱柱,流动相为0.03mol.L-1醋酸铵-甲醇(25:75);流速:0.8 ml.min-1;柱温:40°C;检测波长:254nm。以乙酸乙酯与二氯甲烷(80:20)为提取剂。结果氯氮平的高、中、低(1000.0,400.0,10.0 ng.mL-1)3种浓度平均回收率分别为105.36%,98.63%,97.46%,日内、日间差RSD均低于7%(n=5);分析方法的检测限为5.0 ng.mL-1;线性范围为10.0-1000.0 ng.mL-1。曲线方程:C=25.69F 3.47,r=0.9997(n=10)。结论:该方法灵敏、准确、简单、快速,可用于临床血药浓度监测和药动学研究。     

荧光黄荧光法测定香烟中尼古丁的含量

在pH=7.3的水溶液中,尼古丁与荧光黄(FL)由于静电吸引而发生缔合反应,使荧光黄在λex/λem=450/510nm的荧光增强,据此建立了荧光黄荧光光度测定尼古丁的新方法.尼古丁含量在0.2～8.0 mg·L-1时,荧光强度与尼古丁的含量成线性关系,方法检测限为0.12 mg·L-1,可用于各种不同品牌香烟中尼古丁含量的测定,平行6次测定回收率为91.1%～108.8%,相对标准偏差为1.8%～4.8%,结果满意.     

HPLC法测定西洋参类保健食品中人参皂苷成分的含量

目的建立HPLC法测定以西洋参为主要原料的保健食品中人参皂苷成分含量的方法。方法采用高效液相色谱法测定。色谱柱:岛津C18柱,流动相:乙腈-水(梯度),流速:1.0mL/min,检测波长:203nm。结果人参皂苷Rg1在36.18～361.8ng(r=0.999),人参皂苷Re在0.2254～2.254μg(r=0.9999),人参皂苷Rb1在0.67498～6.7498μg(r=0.9999)之间线性关系良好;人参皂苷Rg1的平均回收率为99.3%,RSD=4.62%;人参皂苷Re的平均回收率101.7%,RSD=1.63%;人参皂苷Rb1的平均回收率100.3%,RSD=1.49%。结论试验表明,该方法操作简单,分离效果好,灵敏度高,可以用于以西洋参为主要原料的保健食品的质量控制。     

槐角丸中两种有效成分含量的HPLC测定法

建立测定槐角丸中游离和结合态槲皮素和染料木素的反相高效液相色谱法,为槐角丸生产企业提供参考。方法采用高效液相色谱仪,以shim-packVp-ODS(4.6mm×150mm,5.0μm)为色谱柱;CH3CN-0.1%H3PO1(5644)为流动相;检测波长为260nm;柱温32℃。结果槲皮素在0.00324-0.01994μg、染料木素在0.05325-0.3192μg范围内线性关系良好,r值分别为0.9988、0.9990,2组分平均回收率在98.0%-100.2%之间,RSD分别为0.81%、1.24%(n=5)。结论该方法一次完成2种有效成分测定,方便、准确、灵敏度高,重现性好,可适用于槐角制剂的质检和生产企业的质控。     

气相色谱内标法测定饮料中环已基氨基磺酸钠

使用柱前衍生化对饮料中的环已基氨基磺酸钠进行处理,用含内标的正己烷萃取衍生化产物,以GC-FID测定其质量分数.结果表明:环已基氨基磺酸钠质量浓度在0.05~5.00 mg/mL范围内,与峰面积线性关系良好(r=0.9999);检出限(S/N=3∶1)为0.95 mg/kg,定量限(S/N=10∶1)为4.75 mg/kg;平均回收率为96.0%,RSD为2.3%.该方法重复性好,适合饮料中环已基氨基磺酸钠的检测.
