Hydantoinase and N-carbamoylase play important roles in the production of optically pure amino acids from racemic 5-monosubstituted hydantoins. In this report, hydantoinase and the N-carbamoylase from Burkholderia cepecia, njut01 were purified to homogeneity by chromatography (Pharmacia Explorer 100 system). The substrate specificity, enantioselectivity, pH dependence of activity and temperature stability of the activity were characterized. The results show that the hydantoinase and N-carbamoylase induced from Burkholderia cepecia, njut01 are both strict D-stereo selective enzymes. They both hydrolyze substrates with side chains containing aliphatic and aromatic residues with higher activity and affinity toward aromatic than aliphatic substituted substrates. The hydantoinase is a homotetramer with subtmit molecular weight near 52,000 and is active between pH 6.5 and 10 with an optimum near pH 9.0. The enzyme is active at temperatures up to 60~C, however, it appears instable at higher temperatures. The subunit molecular weight of N-carbamoylase is about 35KD. The N-carbamoylase is active in the pH range from 6.0 to 9.5. The optira-pH is 7.2 and the optinfizing bioconversion temperature of the N-carbamyolase is 52℃.  相似文献

The paper aims to show how competence as an educational concept for the 21st century is struggling with theoretical problems for which the concept of Bildung in the European tradition can offer alternatives, and to discuss the possibility of developing a sustainable educational concept from the perspectives of competence and Bildung. The method of the study is conceptual analysis of ‘competence’ and Bildung. The paper concludes that (1) competence must be abandoned as an educational concept, as its problems cannot be solved due to the lack of a theory of educational content. With competence, the content aspect of education is obscured and hidden from public debate, and human autonomy is threatened. (2) Bildung can be revised as an educational concept by reinventing educational content as subject to interpretation and open debate by autonomous individuals on all levels from the transnational to the classroom. (3) A revised ‘mimetic’ concept of Bildung can prepare students for the knowledge society, as imagining is a type of higher order thinking essential for innovation and creativity. Instructional content in school is meaningful to students if they are able to imagine the representational object ‘as if’ it is both subject matter and real to them.  相似文献

The paper discusses an on-going research project that studies students' and teachers' conceptions and dispositions of mathematics. Conceptions are the views that students hold on a subject, and what they believe is required in learning and doing the subject. Dispositions are the beliefs or tendencies to exhibit a frequent, conscious and voluntary behavior directed towards learning a subject. The participants are high school students, higher education students and mathematics teachers. The study utilizes self-reporting tools as well as focus groups and interviews in a three-stage research model. This is a work on progress. Results to date are briefly discussed.  相似文献

Pupils’ appraisals of their own ability have been found to be important in their school progress. This paper examines the accuracy and stability of academic self assessments from 7 to 16 years, and the effect of two factors on developments over time: domain/school subject (mathematics or reading/English) and social comparison group (general to children of the same age or specific to own class). Results come from a major longitudinal study of pupils in London schools. An overall lowering in self assessments from 7 to 16 years was accompanied from 11 years by greater accuracy. But systematic differences were found between domains; overall ability ratings were higher in reading/English, and there was more stability in ratings from 11 to 16 years. There was a marked drop in self assessments from the general to the own class social comparison group. But again there were domain differences: there was a greater drop at 11 and 16 years in reading/English.

This paper analyses pedagogic practices in four fields in art and design higher education. This paper examines how and what teachers learn from their older and younger colleagues. Data were gathered from interviews and written reports from 27 Belgian and Finnish teachers. Thematic qualitative analysis was used. The results revealed differences in what teachers learn from older and younger colleagues. Teachers reported learning innovative teaching methods and ICT skills from younger colleagues, whereas practical information, classroom management skills, self-regulation and community building were learned mainly from older colleagues. Attitudes regarding teaching and different ways of being a teacher were learned from both younger and older colleagues alike. Similarities were also found in how teachers learned from their older and younger colleagues. Informal activities and relationships, different forms of mentoring, and working in subject teams or seminars were important sources of learning. An intergenerational learning perspective is important with respect to demographic changes in school staff and in preventing knowledge loss and teacher dropout.  相似文献

Edmund Husserl’s first important move about phenomenology is the“phenomenological reduction”which means that we should reduce the external world to the contents of our consciousness alone. However, Hans-Georg Gadamer holds the opinion that all interpretation of a past work consists in a dialogue between past and present(Eagleton, T. 2009:62). Gadamer’s famous theory is fusion of horizons which means that the event of understanding comes about when our own“horizon”of historical meanings and assumptions“fuses”with the“horizon”within which the work itself is placed. The present thesis takes Hawthorne’s YoungGoodmanBrown as an example to illustrate different understandings when readers apply the two different theories.  相似文献
