试验了亚精胺(Spd)与激动素(KT)复合处理、不同的N素及昼夜温度变化对离体黄瓜子叶花芽分化的影响。发现:KT 9.3×10-3mm o l.L-1 Spd5×1-0 2mm o l.L-1有利于花芽的诱导,低N素和昼夜变化促进花器官的构建,进一步完善了离体黄瓜子叶花芽分化的试验体系。  相似文献

将CMV-CP基因经农杆菌感染介质转化黄瓜子叶,将CMV-CP基因导入黄瓜细胞,在含有卡那霉素和羧卡青霉素的MS培养基上诱导子叶分子,获得一批转化处理的黄瓜再生苗,分化率为23%左右,转基因黄瓜试管苗移栽在蔬菜种植:沙:有机质为5:4;1的混合土中,其移栽成活率可达56%;委托省农科院品种引进中心进行转基因黄瓜再生植株对CMV的抗性接种试验结果表明:7号株系为高抗,2、4号株系为中抗。  相似文献

采用改进的CTAB法,成功地提取了北野豌豆及其近缘种的基因组DNA,电泳结果显示DNA质量和产量均较高,并以此DNA为模板,对ISSR的反应条件进行了优化,最终确定ISSR的反应体系为:总体积10μl,包括10ng的DNA,1倍的反应缓冲液,1.5mmol·L^-1MgCl2,2×10^-4mmol·L^-1(引物6和引物8)或2×10^-3mmol·L^-1(引物5,10和12)引物,0.2mmol·L^-1dNTP混合物和1.0U的Taq DNA聚合酶.ISSR扩增程序为:先在94℃下变性3min,然后在92℃下变性30s,50℃(引物5,6,10,12)或55.2℃(引物8)下复性30s,72℃下延伸1min,反应30个循环,最后72℃延伸5min.并基于此反应条件,最终筛选出5条ISSR引物用于进一步的研究.  相似文献

本文研究了6-BA、IAA、NAA、IBA不同浓度处理对黄瓜外植体器官分化及植株再生的影响.结果表明:6-BA单独处理即可诱导外植体芽分化,浓度为0.5mg/L时分化率最高为33.3%,三种外植体中子叶远轴端无芽分化.IAA、NAA、IBA单独处理均可诱导外植体产生愈伤组织和不定根.两激素配比表明,MS+0.8mg/L 6-BA+0.10mg/L IAA最有利于子叶近轴端分化出芽,MS+0.8mg/L 6-BA+0.15mg/L IAA有利于下胚轴分化出芽,黄瓜植株再生率达30%.  相似文献

考察了碱性介质中碘离子、硫离子和双氧水对ITO电极上电聚合鲁米诺修饰膜ECL的影响.在选定条件下,上述物质显著增敏聚鲁米诺的ECL.在1.0×10-6mol/L-8.0×10-6mol/L范围内,电化学发光强度与碘离子浓度有良好的线性关系,r=0.9896;在8.0×10-7mol/L-1.0×10-5mol/L范围内,电化学发光强度与硫离子浓度有良好的线性关系,r=0.9903;在8.0×10-mol/L-6.0×10-5mol/L范围内,电化学发光强度与双氧水浓度有良好的线性关系,r=0.9939.  相似文献

目的建立盐酸格拉司琼片剂的测定方法。方法以盐酸格拉司琼与四苯硼钠生成缔合物为电活性物,制做涂丝型PVC膜格拉司琼电极。结果在pH4.5-6.5内,电极的Nemst响应范围为1.0×10^-2 - 5.0×10^-5 mol·L^-1,斜率为56.5mV/pc,检测限为2.3×10^-5mol·L^-1。结论用于该药品的含量测定,与高效液相色谱法基本一致。  相似文献

During interview with English majors, it was found that a majority of students expressed disappointment with their experience of English literature classes. Students' dislike of learning English literature appears to be the major problem of teaching English literature in China. Reader response approach is a good way to solve this problem. In this paper, I explain the rationale behind the reader response approach, such as its origin, definition, features, assumptions, and strategies. Then I illustrate how reader response approach works as a teaching strategy by presenting several teaching models.
