目的:探讨生姜醇提物对糖尿病小鼠肾损害的保护作用。方法:将30只KM种雄性小鼠随机分为三组,正常对照组(C组,n=10)、模型组(DM组,n=10)及生姜醇提物干预组(ZGB组,n=10)。DM组及ZGB组小鼠尾静脉注射2%四氧嘧啶(90mg/kg)建立糖尿病肾损害模型。造模成功后,ZGB组给予生姜醇提物溶液0.5mL(300mg/kg·d)灌胃14d,DM组及C组给予生理盐水灌胃。14d后小鼠断头取血,测定血糖、血清肌酐、尿素氮含量,检测肾组织丙二醛(MDA)及超氧化物歧化酶(SOD)活力的变化,光镜下观察肾组织病理学改变。结果:与DM组比较,ZGB组血糖、血肌酐及尿素氮水平明显低于DM组(P〈0.01);肾组织MDA含量明显低于DM组(P〈0.01),而T—SOD活力则较DM组升高(P〈0.01)。光镜下ZGB组肾脏组织形态学的异常改变较模型组减轻。结论:生姜醇提物对糖尿病小鼠的肾损害有一定的保护作用。  相似文献

The present study aimed to evaluate the anti-diabetic property of peanut shell polyphenol extracts (PSPEs). Diabetic rats were oral-administrated with PSPE at doses of 50, 100, and 200 mg/kg body weight (BW) per day for 28 consecutive days, with metformin (Met) as a positive control. The results showed that, similar to the Met treatment, administration of PSPE caused significant decreases in food intake, water intake, fasting blood glucose, total cholesterol, triglyceride, low-density lipoprotein cholesterol, and methane dicarboxylic aldehyde in serum, and significant increases in BW, insulin level, high-density lipoprotein cholesterol, superoxide dismutase, glutathione, and liver glycogen. Further, glucose tolerance was markedly improved in the PSPE-treated diabetic groups. Histopathological results showed that PSPE improved cellular structural and pathological changes in liver, kidney, and pancreatic islets. Collectively, the results indicated that the hypoglycemic effects of PSPE on high-fat diet/streptozotocin (HFD/STZ)- induced diabetes are comparable to Met, though their exact mechanism actions are still under investigation. Therefore, the current study suggests that PSPE could be a potential health-care food supplement in the management of diabetes.  相似文献

目的研究免疫初乳对正常小鼠和四氧嘧啶致糖尿病小鼠血糖的影响.方法用正常小鼠和糖尿病模型小鼠研究免疫初乳调节血糖的作用.连续30d给予正常小鼠和四氧嘧啶糖尿病模型小鼠免疫初乳,30d后眼眶取血,取其血清测定小鼠空腹血清血糖.结论免疫初乳能显著降低四氧嘧啶致高血糖小鼠的血糖水平并提高其糖耐量,且高剂量免疫初乳组的效果好于低剂量组的效果.但免疫初乳对正常小鼠的血清血糖及糖耐量没有影响.  相似文献

Objective

The present study aims at determining the stability of a popular type 2 diabetes rat model induced by a high-fat diet combined with a low-dose streptozotocin injection.

Methods

Wistar rats were fed with a high-fat diet for 8 weeks followed by a one-time injection of 25 or 35 mg/kg streptozotocin to induce type 2 diabetes. Then the diabetic rats were fed with regular diet/high-fat diet for 4 weeks. Changes in biochemical parameters were monitored during the 4 weeks.

Results

All the rats developed more severe dyslipidemia and hepatic dysfunction after streptozotocin injection. The features of 35 mg/kg streptozotocin rats more resembled type 1 diabetes with decreased body weight and blood insulin. Rats with 25 mg/kg streptozotocin followed by normal diet feeding showed normalized blood glucose level and pancreatic structure, indicating that normal diet might help recovery from certain symptoms of type 2 diabetes. In comparison, diabetic rats fed with high-fat diet presented decreased but relatively stable blood glucose level, and this was significantly higher than that of the control group (P>0.05).

Conclusions

This model easily recovers with normal diet feeding. A high-fat diet is suggested as the background diet in future pharmacological studies using this model.

目的:考察慈姑多糖对四氧嘧啶性糖尿病模型小鼠血糖的影响。方法:采用皮下一次性注射190mg/kg四氧嘧啶致实验性糖尿病小鼠,观察慈姑多糖不同剂量(0.75g/kg,0.375g/kg)对糖尿病小鼠空腹血糖的影响。结果:慈姑多糖高、低剂量组能显著性降低血糖水平(P〈0.05)。结论:慈姑多糖具有一定的降血糖作用。  相似文献

为了研究补充维生素C(VC)对冷应激环境下雄性小鼠血清生化指标和部分内脏器官的影响。本试验选购清洁级雄性昆明小鼠30只,随机分为冷应激组、冷应激+VC组和对照组(n=10),冷应激和冷应激+VC组每天在0℃下给2h的冷刺激,冷应激+VC组补充300μg/g VC,试验期1w。结果表明,在冷应激环境中,小鼠血清AST、UREA、UA、Glu、TP和CREA水平均出现显著变化(P<0.05);肾器官指数显著增加(P<0.05),睾丸和附睾重量显著降低(P<0.05);肾、睾丸和附睾出现不同程度损伤,但肝的重量、器官指数和组织结构以及无明显变化。补充VC后,血清UA明显降低(P<0.05),Glu、TP和CREA含量显著增加(P<0.05),而AST和UREA含量无明显变化(P>0.05);肾器官指数显著降低(P<0.05),睾丸重量明显增加(P<0.05);肾小球和肾小囊结构正常,睾丸生精细胞和精子数量增加,附睾管脱落上皮细胞数量减少。  相似文献

[目的]〗研究知母多糖(Anemam,AM)对家兔血糖的影响.[方法]制备家兔高血糖模型,用不同剂量组的AM和D860后,测定对照组、AM组、D860组的血糖.[结果]AM1次灌胃2mg/kg,20mg/kg可显著降低四氧嘧啶引起的家兔血糖升高;AM1次灌胃20mg/kg对正常家兔血糖无明显影响,200mg/kg可降低正常家兔血糖.[结论]AM与D860同样具有实验性降低血糖作用.  相似文献

Objective:To investigate the effect of Anethum graveolens(AG) extracts on the mounting frequency,histology of testis and epididymis,and sperm physiology.Methods:Male rats induced by cold immobilization before treating with vehicle or AG extracts [50,150,and 450 mg/kg body weight(BW)] via gastric tube for consecutive 1,7,and 14 d were examined for mounting frequency,testicular phosphorylation level by immunoblotting,sperm concentration,sperm acrosome reaction,and histological structures of testis and epididymis,respectively.Results:AG(50 mg/kg BW) significantly increased the mounting frequency on Days 1 and 7 compared to the control group.Additionally,rat testis treated with 50 mg/kg BW AG showed high levels of phosphorylated proteins as compared with the control group.In histological analyses,AG extract did not affect the sperm concentration,acrosome reaction,and histological structures of testis and epididymis.Conclusions:AG extract enhances the aphrodisiac activity and is not harmful to sperm and male reproductive organs.  相似文献

Stress affects the male reproductive system and can cause sub-fertility or infertility. Although Phyllanthus emblica L. (PE) extract has been shown to have high antioxidant capacity and protective properties in damaged tissue, the preventive effects of PE extract on testicular function from stress-related impairment have never been demonstrated. This study aimed to investigate the effects of PE aqueous leaf extract on testicular impairment and protein marker changes in rats suffering from chronic stress. Adult male rats were divided into four groups: a control group, a chronic stress (CS) group, and two groups with CS that received different doses of PE extract (50 or 100 mg/kg body weight (BW)). In the treatment groups, the animals were given PE extract daily before stress induction for 42 consecutive days. Stress was induced through immobilization (4 h/d) followed by forced cold swimming (15 min/d). Sperm quality and the histology of the testes and caudal epididymis were examined, as were levels of serum corticosterone, testosterone, and malondialdehyde (MDA). The expressions of testicular steroidogenic acute regulatory (StAR) and tyrosine-phosphorylated proteins were investigated using immuno-Western blot analysis, as these proteins are assumed to play important roles in spermatogenesis and androgen synthesis. The results showed that PE (50 mg/kg BW) significantly increased sperm concentration and testosterone levels, while decreasing corticosterone levels, MDA levels, sperm head abnormalities, and acrosome-reacted sperm in CS rats. In addition, PE at both doses was found to diminish testicular histopathology in the CS rats. We also found that 50 mg/kg BW of PE significantly improved StAR protein expression and altered the intensities of some tyrosine-phosphorylated proteins in testis. We conclude that PE leaf extract at 50 mg/kg BW can prevent testicular damage in rats with CS.  相似文献

The effects ofpolychlorinated biphenyls (PCBs) on reproduction of adult cocks were studied by gavaging peanut oil or PCBs (Aroclor 1254, 50 mg/kg) once a week for six consecutive weeks. Physiological parameters were recorded and gonads were removed at the end of experiment for histological examination. The results showed that there was no significant difference between the control and treatment group in body weight, respiration rate, heart rate, body temperature, and the numbers of red and white blood cells. However, there was a marked decrease in the testicular weight and serum testosterone level after PCB treatment. Morphological studies manifested severe damage of the seminiferous tubules by PCB. The number of the germ cells at the different developmental stages was decreased and condensed nuclei were observed in most of these cells. This study revealed that the reproductive function of the adult cocks is sensitive to PCBs, which inhibited mainly spermatogenesis and testosterone secretion.  相似文献

Objective

To evaluate the potential adjuvant effect of Agrocybe aegerita lectin (AAL), which was isolated from mushroom, against a virulent H9N2 strain in vivo and in vitro.

Methods

In trial 1, 50 BALB/c male mice (8 weeks old) were divided into five groups (n=10 each group) which received a subcutaneous injection of inactivated H9N2 (control), inactivated H9N2+0.2% (w/w) alum, inactivated H9N2+0.5 mg recombinant AAL/kg body weight (BW), inactivated H9N2+1.0 mg AAL/kg BW, and inactivated H9N2+2.5 mg AAL/kg BW, respectively, four times at 7-d intervals. In trial 2, 30 BALB/c male mice (8 weeks old) were divided into three groups (n=10 each group) which received a subcutaneous injection of inactivated H9N2 (control), inactivated H9N2+2.5 mg recombinant wild-type AAL (AAL-wt)/kg BW, and inactivated H9N2+2.5 mg carbohydrate recognition domain (CRD) mutant AAL (AAL-mutR63H)/kg BW, respectively, four times at 7-d intervals. Seven days after the final immunization, serum samples were collected from each group for analysis. Hemagglutination assay, immunogold electron microscope, lectin blotting, and co-immunoprecipitation were used to study the interaction between AAL and H9N2 in vitro.

Results

IgG, IgG1, and IgG2a antibody levels were significantly increased in the sera of mice co-immunized with inactivated H9N2 and AAL when compared to mice immunized with inactivated H9N2 alone. No significant increase of the IgG antibody level was detected in the sera of the mice co-immunized with inactivated H9N2 and AAL-mutR63H. Moreover, AAL-wt, but not mutant AAL-mutR63H, adhered to the surface of H9N2 virus. The interaction between AAL and the H9N2 virus was further demonstrated to be associated with the CRD of AAL binding to the surface glycosylated proteins, hemagglutinin and neuraminidase.

Conclusions

Our findings indicated that AAL could be a safe and effective adjuvant capable of boosting humoral immunity against H9N2 viruses in mice through its interaction with the viral surface glycosylated proteins, hemagglutinin and neuraminidase.

应用黄芪水提物与不同浓度的α-葡萄糖苷酶,不同浓度的底物PNP-glucoside进行反应,研究中药黄芪中α-葡萄糖苷酶抑制剂的抑制活性及其对正常小鼠血糖和糖耐量的影响.通过绘制酶量-反应速度曲线和双倒数曲线,确定其抑制剂类型,并对正常小鼠以40~160 mg.kg-1剂量灌胃后,观察其对血糖值和糖耐量的影响.结果表明,黄芪中α-葡萄糖苷酶抑制剂为可逆、非竞争性抑制;可降低正常小鼠血糖值,并且以80 mg.kg-1的剂量时血糖值最低,还可显著提高小鼠糖耐量.黄芪水提物中含有α-葡萄糖苷酶抑制剂成分,可用于糖尿病治疗.  相似文献

The purpose of this study was to clarify effects of selected oligosaccharides on concentrations of cecal short-chain fatty acids (SCFAs), total large bowel wet weight and wall weight, and cecal microbiota levels in mice. Mice were respectively given gavage of selected fructooligosaccharides (FOS), galactooligosaccharides (GOS), mannanoligosaccharides (MOS), and chitooligosaccharides (COS) [1000 mg/(kg body weight.d)]. Control group was given physiological saline solution. After 14 d treatment, SCFAs and lactate in mice cecum were significantly increased (P<0.05) by intake of oligosaccharides, especially FOS and GOS. Thus, providing these oligosaccharides as ingredients in nutritional formulas may benefit the gastrointestinal tract.  相似文献

研究DHEA对淮南王鸡脂质代谢与抗氧化的影响。240只8日龄健康雏鸡随机分为4组:对照组(饲喂基础饲料)以及试验Ⅰ、Ⅱ、Ⅲ组(分别饲喂含有5、15和45 mg DHEA/kg的饲料)。42和56日龄各组随机抽取发育正常的12只个体,空腹称重,按常规方法屠宰,测定肝脏、腹脂、皮下脂肪相对重以及胸肌、腿肌肌内脂肪含量,并测定56日龄时血清丙二醛(MDA)、葡萄糖(Glu)、甘油三酯(TG)和胆固醇(TC)含量以及总抗氧化能力(T-AOC)、脂蛋白脂酶(LPL)和肝脂酶(HL)活性等。结果显示:在42日龄,试验Ⅱ组肝脏以及试验III组腹脂、皮下脂肪相对重显著低于对照组(P<0.05);在56日龄,试验Ⅱ组肝脏、腹脂相对重以及试验Ⅲ组的腹脂、皮下脂肪相对重皆显著降低(P<0.05);DHEA处理有降低血糖、血脂的趋势,其中试验Ⅱ组血清血糖、TC、TG和VLDL含量均显著低于对照组,差异显著(P<0.05);试验Ⅱ、Ⅲ组血清LPL活性显著升高(P<0.05),试验Ⅰ组血清HL活性显著增加(P<0.05);试验Ⅰ、Ⅱ组血清MDA含量显著降低(P<0.05),而T-AOC水平显著增加(P<0.05)。结论:DHEA具有降低鸡的脂质代谢和增强抗氧化的作用。  相似文献

目的:观察脂肪肝大鼠在生化、病理、脂肪代谢、细胞因子、胰岛素及瘦素各方面指标的变化.方法:雄性Wistar大鼠22只,按体重层次随机分为2组,对照组(A)11只,脂肪肝组(B)11只.对照组饲以普通鼠饲料,脂肪肝组饲以高脂饲料.8周后,全部处死,取血及肝组织,测定血中AIT、AST、TG、CHO、INS、FBS、FFA、Ieptin、II-6、TNF.肝组织匀浆测定肝组织TC,TG,SOD、MDA,观察肝脏组织学变化.结果:脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素表达增高;细胞因子升高.结论:实验性高脂饮食脂肪肝大鼠存在瘦素抵抗及胰岛素抵抗.  相似文献

Background

Ketoconazole (KET), an antifungal drug, has adverse effects on the male reproductive system. Pre-treatments with antioxidant plant against testicular damage induced by KET are required. The flowers of Clitoria ternatea (CT) are proven to have hepatoprotective potential. However, the protective effect on KET-induced testicular damage has not been reported.

Objective

To investigate the protective effect of CT flower extracts with antioxidant activity on male reproductive parameters including sperm concentration, serum testosterone level, histopathology of the testis, and testicular tyrosine phosphorylation levels in rats induced with KET.

Methods

The antioxidant activity of CT flower extracts was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. Male rats were treated with CT flower extracts (10, 50, or 100 mg/kg BW) or distilled water via a gastric tube for 28 d (preventive period: Days 1–21) and induced by KET (100 mg/kg BW) via intraperitoneal injection for 7 d (induction period: Days 22–28). After the experiment, all animals were examined for the weights of the testis, epididymis plus vas deferens and seminal vesicle, serum testosterone levels, sperm concentration, histological structures and diameter of testis, and testicular tyrosine phosphorylation levels by immunoblotting.

Results

The CT flower extracts had capabilities for DPPH scavenging and high reducing power. At 100 mg/kg BW, the extract had no toxic effects on the male reproductive system. Significantly, in CT+KET groups, CT flower extracts (50 and 100 mg/kg BW) alleviated the reduction of reproductive organ weight parameters, testosterone levels, and sperm concentration. In addition, CT flower extracts gave protection from testicular damage in KET-induced rats. Moreover, in the CT100+KET group, CT flower extracts significantly enhanced the expression of a testicular 50-kDa tyrosine phosphorylated protein compared with that of other groups.

Conclusions

C. ternatea flower extracts possessing antioxidant activity are not harmful to the male reproductive system and can protect against testicular damage in KET-induced rats.  相似文献

目的:研究牛奶和鸡蛋对运动能力的影响,探讨牛奶和鸡蛋对运动疲劳恢复影响的可能机制.方法:将普通成体雄性ICR小白鼠(18只)随机均分对照组(CG)、额外补充牛奶组(MG),额外补充鸡蛋组(EG);采用10d递增负荷的游泳运动方式使小白鼠力竭.分析牛奶与鸡蛋对小白鼠力竭运动后血糖、血红蛋白(Hb)、血肌酐(CRE)含量影响及体重变化,记录力竭时间、观察行为状态.结果:与实验前比较,各组小鼠实验后血糖、血红蛋白(Hb)浓度下降,血肌酐(CRE)浓度上升;两实验组小鼠体重增加均显著高于CG组;两实验组与CG相比,运动时间显著延长.结论:小鼠运动疲劳后,机体出现各种疲劳指标的变化,额外补充牛奶和鸡蛋可以减少这些疲劳指标的改变.可能是通过增加糖储备来提高运动能力,保证机体在长时间运动时的能量来源;减缓运动过程中对蛋白质的过度利用;增加体内磷酸肌酸的贮备,提高磷酸肌酸的代谢能力等方面缓解运动疲劳的,尚待进一步研究.  相似文献

144只一日龄的AA肉仔鸡随机分为4组(n=36),预饲一周后分别在试验组基础日粮中添加50mg/kg、100mg/kg、150mg/kg的半胱胺,分别于2、4、6周龄采颈静脉血,分离血清,测定四组血清钙、磷和葡萄糖含量及ALT、AST、ALP活性,研究半胱胺对肉鸡部分血清化学参数及酶活性的影响.结果表明,饲料中添加适量的Cs能升高AA肉鸡的血钙水平,而对鸡血清磷、葡萄糖含量没有明显影响;添加50~100mg/kg的Cs对ALT、AST活性没有明显的影响;而饲料中添加Cs对ALP活性有一定影响.  相似文献

观察氮-硝基-左旋精氨酸(L-NNA)和左旋精氨酸(L-Arg)对糖尿病大鼠胃排空的影响。实验结果表明,糖尿病大鼠早期静脉给L-NNA后,各测定值与糖尿病对照组差异显著(P<0.01),与正常对照组无差异(P!0.05);糖尿病大鼠延迟应用L-Arg后,各值也与糖尿病对照组差异显著(P<0.01),但未能恢复到正常对照组水平(P<0.01)。所以,糖尿病早期应用L-NNA能明显改善糖尿病大鼠胃排空延迟,保护调节胃动力。  相似文献
