Application of cDNA array for studying the gene expression profile of mature appressoria of Magnaporthe grisea

Appressorium is an infection structure of the phytopathogenic fungus Magnaporthe grisea. Analysis of gene expression profiles ofappressorium development provides insight into the molecular basis of pathogenicity and control of this fungal plant disease. A cDNA array representing 2927 unique genes based on a large EST （expressed sequence tag） database ofM. grisea strain Y34 was constructed and used to profile the gene expression patterns at mycelium and appressorium maturation stages. Compared with mycelia, 55 up-regulated and 22 down-regulated genes were identified in mature appressoria. Among 77 genes, 16 genes showed no similarity to the genome sequences of M. grisea. A novel homologue of peptidyl-prolyl cis-trans isomerase was found to be expressed at low-level in mature appressoria of M. grisea. The results indicated that the genes such as pyruvate carboxylase, phospholipid metabolism-related protein and glyceraldehyde 3-phosphate dehydrogenase involved in gluconeogenesis, lipid metabolism and glycolysis, showed differential expression in mature appressoria. Furthermore, genes such as PTHll, beta subunit of G protein and SGTI involved in cell signalling, were expressed differentially in mature appressoria. Northern blot analysis was used to confirm the cDNA array results.     

Bilateral shear strength of rectangular frame column based on simplified space truss-arch model

To investigate the bilateral shear strength of rectangular frame column subjected to oblique horizontal load, we presented a simplified space truss-arch model developed from unilateral truss-arch model. Main parameters in the new model were the cross-sectional area, transverse reinforcement raito, axial load, and material strength of the column. The reduction coefficient of concrete sterength owing to the severe cracking of column was also introduced in the model. Finally, 14 specimens under oblique horizontal load were tested to verified the feasibility and applicability of the space truss-arch model.     

Aversive parenting in China: associations with child physical and relational aggression

This study assessed the combined and differential contributions of Chinese mothers and fathers (in terms of spouse-reported physically coercive and psychologically controlling parenting) to the development of peer-reported physical and relational aggression in their preschool-age children (mean age of 5 years). Results of the two-group (boys and girls) latent sum and difference structural equation model showed that combined parenting effects were slightly more prevalent than differential effects in predicting aggression. Furthermore, physical coercion was predictive of aggression in boys whereas psychological control was primarily associated with aggression in girls. Findings extend our understanding of relational aggression and the meaning of aversive parenting, particularly within the Chinese cultural context.     

Computational prediction of microRNA genes in silkworm genome

MicroRNAs （miRNAs） constitute a novel, extensive class of small RNAs （-21 nucleotides）, and play important gene-regulation roles during growth and development in various organisms. Here we conducted a homology search to identify homologs of previously validated miRNAs from silkworm genome. We identified 24 potential miRNA genes, and gave each of them a name according to the common criteria. Interestingly, we found that a great number of newly identified miRNAs were conserved in silkworm and Drosophila, and family alignment revealed that miRNA families might possess single nucleotide polymorphisms, miRNA gene clusters and possible functions of complement miRNA pairs are discussed.

Group Cooperative Learning—Making Learning a Deeper Process

Sample size and power estimates for a confirmatory factor analytic model in exercise and sport: a Monte Carlo approach

Monte Carlo methods can be used in data analytic situations (e.g., validity studies) to make decisions about sample size and to estimate power. The purpose of using Monte Carlo methods in a validity study is to improve the methodological approach within a study where the primary focus is on construct validity issues and not on advancing statistical theory. The purpose of this study is to demonstrate how Monte Carlo methods can be used to determine sample size and to estimate power for a confirmatory factor analytic model under model-data conditions commonly encountered in exercise and sport. Because the purpose is pursued by way of demonstration with the Coaching Efficacy Scale II-High School Teams, related sample size recommendations are provided: N > or = 200 for the theoretical model; N > or = 300 for the population model. Technical terms (e.g., coverage) are defined when necessary.
