研究目的:开发具有种属特异性序列特征性扩增区域(SCAR)标记物来监测哈茨木霉在其入侵的试验菌群中的定殖和生长,为哈茨木霉应用于生物防治等生态和生物技术中提供支撑。 创新要点:多种木霉属真菌能与各种微观和宏观的生物有机体建立相互作用。利用这些相互作用,木霉可做为原生种群的入侵物种而用于生物防治。本文通过使用试验菌群为研究模型,利用随机扩增多态性DNA(RAPD)技术和序列特征性扩增区域(SCAR)标记物来监测菌群中哈茨木霉的生长状态。 研究方法:利用随机扩增多态性DNA(RAPD)技术,从16个10聚体引物进行多态性筛选,其中1个引物扩增出对应哈茨木霉的条带。对该条带进行克隆测序,并设计5个20-23聚体引物。成功利用引物对2F2/2R2和2F2/2R3278分别特异性地扩增出哈茨木霉BpTloa菌株278bp和448bp的DNA片段。同时,用这两个引物对14个哈茨木霉菌株和几种不同的真菌菌株进行特异性对照试验,也只成功扩增出哈茨木霉菌株。此外,使用真菌DNA混合物和试验真菌群的DNA为模板,采用实时聚合酶链式反应(PCR)对引物对2F2/2R2进行评估。当仅使用100份SCAR标记物或哈茨木霉仅占整个菌群的0.1%时,仍能检测出哈茨木霉。 重要结论:本研究所建立的SCAR分子标记能有效监测菌群中的哈茨木霉的定殖和生长,具有较高特异性、灵敏度和准确度。  相似文献

In this article, we examine key dilemmas childbirth educators experienced as they made crucial decisions about the content and format of their classes in the current U.S. maternity-care context. This ethnographic study demonstrates that childbirth education is a cultural phenomenon with deeply embedded values regarding the nature and importance of information, scientific evidence, and consumer choice. Articulating how culture shapes the presentation, content, and format of childbirth classes is an important step in understanding and increasing the relevance of this experience for birthing women.  相似文献

Learning from curriculum materials: Scaffolds for new teachers?   总被引:1,自引:0,他引:1  
This article explores how beginning teachers use and learn from curriculum materials. As part of a longitudinal study of beginning English teachers who teach in the Pacific Northwest of the United States, the researchers tracked teachers’ responses to and use of materials over time, and how these materials shaped their classroom practice. The authors found that the teachers spent an enormous amount of time searching out curriculum materials for their classes and that the curriculum materials they encountered did, indeed, powerfully shape their ideas about teaching language arts as well as their classroom practice. Based on their findings, the authors propose a trajectory for the teachers’ use of the curriculum materials. New teachers begin by sticking close to the materials they have at hand. Then, over time, as they learn more about both students and curriculum, they adapt and adjust their use of the materials. The authors argue that new and aspiring teachers need opportunities to analyze and critique curriculum materials, beginning during teacher education and continuing in the company of their more experienced colleagues.  相似文献

Health-seeking encompasses the activities individuals perform in order to restore wellness when they perceive themselves as ill. Alarmingly, it is apparent that there is a decrease in frequency of health-seeking as one ages. While there have been numerous studies about elderly health-seeking behavior across the globe, the need to know about their preferences remains a research imperative, hence this conjoint analysis investigation. The objective of this study is to identify the health-seeking preferences of a select group of Filipino elderly in the community. To measure preferences for health-seeking, a conjoint analysis survey of 304 Filipino elderly aged 60 years and above was conducted from June to July 2012 using the balanced incomplete block design. Results indicate that the most important attribute in terms of health-seeking behavior is the physician's experience. Elderly clients tend to seek health care from expert private practitioners who provide full information regarding illness and charge professional fees of less than 500 pesos (8–9 British pounds or 12–13 U.S. dollars). Results suggest that there are factors that may impede or promote health care seeking in elderly clients, of which physicians’ experience is deemed most important. These preferences, if integrated into the current practice, can help health care practitioners in improving the quality of care provided.  相似文献

Forty-seven talented fourth graders were assessed on different WISC-R short-form combinations in order to determine which short-form was the most effective predictor of high Full Scale WISC-R IQ. The short-form combinations of Similarities + Object Assembly + Vocabulary subtests (SI-OA-VO) or Similarities + Object Assembly subtests (SI-OA) were more effective in this regard than eight other WISC-R short-form combinations, the Slosson Intelligence Test, and school grades in Math, Language, and Reading combined. This meta-analysis examined concurrent associations between aggression, withdrawal, assertion, and prosocial behavior and each of positive and negative friendship quality across studies with 22,657 children and adolescents (Mage = 11.71 years; 51.7% girls; 67.7% White). Studies were published between 1995 to 2021 and 32.4% were conducted outside of North America. Aggression was linked to more negative,  = .19, 95% CI [.14, .24], and less positive,  = −.05 [−.08, −.01], friendships. Withdrawal was associated with less positive friendships,  = −.13 [−.18, −.08], whereas prosocial behavior was related to more positive,  = .29 [.22, .37], as well as less negative,  = −.16 [−.20, −.12], friendships. A 4‐week mini‐project is proposed for an experimental Physical Chemistry course applied in Food Science training. Activities include preparation of beet extract for the extraction of betanins and analysis of the kinetics of betanins in thermal degradation at three temperatures (60, 70, and 80 ºC). In addition to developing common laboratory routines (for example, dilution and use of the UV‐Vis spectrophotometer), students will have contact with the usual procedure for evaluating use of natural dyes in the food industry.  相似文献

Just like adults, children of all ages need time and understanding in order to process the concept of death and dying. This process is much different for children than it is for adults. There are 4 components relative to children's understanding of death: (a) the irreversibility factor, (b) finality, (c) inevitability, and (d) causality. These 4 components relate directly to the developmental level of the child at the time the death occurs. Knowing how children's concept of death is constructed provides parents and caregivers important information and helps them respond more sensitively to what children might feel and experience. This article provides an overview of how children understand death, concrete strategies for talking to children about death, and suggestions for teachers about how to help children through grief and mourning.  相似文献
