A method of in situ chromosome immobilisation and DNA extraction in a microfluidic polymer chip was presented. Light-induced local heating was used to induce poly(N-isopropylacrylamide) phase transition in order to create a hydrogel and embed a single chromosome such that it was immobilised. This was achieved with the use of a near-infrared laser focused on an absorption layer integrated in the polymer chip in close proximity to the microchannel. It was possible to proceed to DNA extraction while holding on the chromosome at an arbitrary location by introducing protease K into the microchannel. 相似文献

This paper provides a theoretical review of self-rated competences questionnaires. This topic is influenced by the ongoing world-wide reform of higher education, which has led to a focus on the learner outcomes of higher education. Consequently, questionnaires on self-rated competences have increasingly been employed. However, self-ratings are often criticised for their lack of validity. Our intention is to outline some principles of good questionnaire design and to use these principles to contrast questionnaires on self-rated competences. We begin with an overview of research about questionnaire design. Then we introduce seven questionnaires and portray them in terms of their design characteristics. A comparison reveals some significant points: biographical data need to be handled more carefully, and there is an overuse of vague and abstract language. On the positive side, all of the questionnaires that were examined provide reliable sub-scales covering important facets of competences. 相似文献

The expectation for extended library hours including 24 h access presents many questions about late night library users and the value of this service to the library and their corresponding institutions. Using data gathered about patrons who use the library after 11 pm and before 7 am, a study was conducted to discover who the library users were, what services and facilities they made use of, and what, if any, concerns they have regarding the late night library. The usage patterns provide evidence that that while more usage occurs during traditional hours, users who prefer late night do so consistently. The researchers found the late night library was serving patrons who were cross sections of university community with higher than average grade point averages and retention rates when compared to university averages. These figures can have a significant effect on institutional recruitment and retention presenting the library as an indispensable resource to the community. 相似文献

A number of strategies relating to the evaluation of preschool programs have been changing. One change has been from evaluating service delivery systems and child development outcomes independent of one another. A second area of change has been from evaluating the impact of a particular intervention on an individual child to evaluating the impact of an intervention which is part of a dynamic (ecological) setting, on a child who is within that dynamic (ecological) setting. A third change has been from evaluating simply in terms of child development outcomes to evaluating both outcomes as well as those aspects of a child development program which can be manipulated or controlled by policy and management decisions. These are not mutually exclusive changes, and, indeed, a good deal of overlap will be evident. However, there are sufficient differences in approach to warrant separate treatment. The present study examined faculty commitment to their university across hard and soft sciences and across pure and applied fields. Within each field, faculty commitment was separately examined for each career stage and for each level of research productivity. Furthermore, this study assessed (separately for each field) the relative powers of rewards and support variables, derived from the exchange framework, in explaining faculty commitment. A stratified random sample of 40 U.S. research university departments, 10 each in physics, sociology, electrical engineering, and education, was surveyed by means of a questionnaire. The major findings of the study suggest that the reward-support framework plays a meaningful role, although distinctly different for hard and soft sciences, in determining faculty commitment to their university in each of the four fields. The relative power of rewards in explaining faculty commitment is stronger in hard sciences and weaker in soft sciences whereas a reversed pattern is found for support indicators. In addition, faculty commitment to their university varies across fields, career stages, and levels of research productivity (only for hard sciences). 相似文献
