Based on bioinformatic analysis, we selected two novel microRNAs (miRNAs), bmo-miR-0001 and bmomiR-0015, from high-throughput sequencing of the Bombyx mori larval posterior silk gland (PSG). Firstly, we examined the expression of bmo-miR-0001 and bmo-miR-0015 in 12 different tissues of the 5th instar Day-3 larvae of the silkworm. The results showed that the expression levels of both bmo-miR-0001 and bmo-miR-0015 were obviously higher in the PSG than in other tissues, implying there is a spatio-temporal condition for bmo-miR-0001 and bmo-miR-0015 to regulate the expression of BmFib-L. To test this hypothesis, we constructed pri-bmo-miR-0001 expressing the plasmid pcDNA3.0 [ie1-egfp-pri-bmo-miR-0001-SV40] and pri-bmo-miR-0015 expressing the plasmid pcDNA3.0 [ie1-egfp-pribmo-miR-0015-SV40]. Finally, the BmN cells were harvested and luciferase activity was detected. The results showed that luciferase activity was reduced significantly (P<0.05) in BmN cells co-transfected by pcDNA3.0 [ie1-egfp-pri-bmomiR-0001-SV40] or pcDNA3.0 [ie1-egfp-pri-bmo-miR-0015-SV40] with pGL3.0 [A3-luc-Fib-L-3′UTR-SV40], suggesting that both bmo-miR-0001 and bmo-miR-0015 can down-regulate the expression of BmFib-L in vitro. 相似文献

Although there are a number of medical schools in the Caribbean islands, very few reports have come out so far in the literature regarding the efficacy of small‐group teaching in them. The introduction of small‐group teaching in the gross anatomy laboratory one and a half years ago at St. Matthew's University (SMU) on Grand Cayman appears to have had a significant positive impact on the academic achievement of students in anatomy. This study surveyed the responses of the students to the small‐group learning method in gross anatomy at SMU using a structured questionnaire. The results show that our students prefer this small‐group learning method over a completely self‐directed method in the gross anatomy lab because the study materials were carefully chosen and the study objectives were demonstrated by the resource person. However, teacher‐centered teaching was deliberately avoided by fostering problem‐solving skills in the anatomy lab sessions. Another aim of the small‐group teaching at SMU was to develop the interpersonal and communication skills of the students, which are important in their later education and career. Body donation is important for medical education and academic research. However, it is relatively rare in Hong Kong when compared with many Western countries. Comprehensive research has been performed on the motivation for body donation in Western countries; however, there is still insufficient research on body donation in Hong Kong to provide information on how to increase the body‐donation rate. To understand the factors involved in the decision to donate one's body, the authors interviewed a registered donor and the daughter of another donor in Hong Kong. The authors interpreted the information collected in light of the available published reports, which mostly focus on body donation in Western countries. Despite the consistency of some demographic factors and motivations between the participants in our study and those investigated in the published reports from Western countries, there are differences in education level and socioeconomic status between the donors in our study and those from Western studies. The authors also suggest that Confucianism and Buddhism in Chinese culture may motivate potential body donors in Hong Kong. Other important factors that influence the body‐donation decision may include family members' body donation, registration as organ donors, and good doctor–patient relationships. Although case report studies have their limitations, this study allows us to explore the complexity of events and establish the interconnectivity of factors involved in body donation, which could not be achieved in previous survey‐based studies. © 2012 American Association of Anatomists. 相似文献

Anatomical corrosion casts of human specimens are useful teaching aids. However, their use is limited due to ethical dilemmas associated with their production, their lack of perfect reproducibility, and their consumption of original specimens in the process of casting. In this study, new approaches with modern distribution of complex anatomical spatial information were explored to overcome these limitations through the digitalization of anatomical casts of human specimens through three‐dimensional (3D) reconstruction, rapid prototype production, and Web‐based 3D atlas construction. The corrosion cast of a lung, along with its associated arteries, veins, trachea, and bronchial tree was CT‐scanned, and the data was then processed by Mimics software. Data from the lung casts were then reconstructed into 3D models using a hybrid method, utilizing both “image threshold” and “region growing.” The fine structures of the bronchial tree, arterial, and venous network of the lung were clearly displayed and demonstrated their distinct relationships. The multiple divisions of bronchi and bronchopulmonary segments were identified. The 3D models were then uploaded into a rapid prototype 3D printer to physically duplicate the cast. The physically duplicated model of the lung was rescanned by CT and reconstructed to detect its production accuracy. Gross observation and accuracy detection were used to evaluate the duplication and few differences were found. Finally, Virtual Reality Modeling Language (VRML) was used to edit the 3D casting models to construct a Web‐based 3D atlas accessible through Internet Explorer with 3D display and annotation functions.
