Library materials are susceptible to fungal deterioration. The paper constituents of archival materials are subjected to harmful physical and chemical processes as they are slowly consumed by fungi. Remediation of fungal contamination can be costly and risk further damage to fragile or previously degraded materials. Early detection of fungal growth would permit the use of relatively noninvasive treatments to remediate fungal contamination of artifacts before visible or lasting damage has occurred. Current methods used for the detection of microbial biomass, such as colony counts, microscopic biovolume estimation, and ergosterol analysis are expensive, time consuming, or are inappropriate for use with fungi. Beta-N-acetylhexosaminidase (EC 3.2.1.52) activity provides a rapid and reliable means of fungal detection on a variety of cultural heritage materials. Adapted for use on archival materials, fluorogenic 4-Methylumbelliferyl (MUF) labeled substrate N-acetyl-Beta-D-glucosamine (NAG) was used to detect fungal beta-N-acetylhexosaminidase activity. The fluorescence generated by minute quantities of fungi was quickly detected at an early stage of growth. The sensitivity of the assay was comparable to other biochemical techniques. The fluorometric assay was well-suited for early detection of fungal biomass on paper and assessment of the effectiveness of common remediation practices.  相似文献

There is currently no validated model explaining the variability of sexual expression. This has created a scenario where sexuality, as a construct, is purely intuitive. Sexuality educators have frequently presented the Circles of Sexuality, a model that contends that sexuality is a combination of intimacy, sensuality, sexual health/behaviors, sexual identity, and sexualization. Adapting photovoice methodology and using the social media site Tumblr, the current analysis used this model to assess the social conceptualization of sexuality. Results indicate that the circles are unequally represented, with intimacy underrepresented and sexual identity overrepresented. Implications for sexuality educators, clinicians, and researchers are discussed.  相似文献

Representations are often used in instruction to highlight key mathematical ideas and support student learning. Despite their centrality in scaffolding teaching and learning, most of our understanding about the tasks involved with using representations in instruction and the knowledge requirements imposed on teachers when using these aids is theoretical. In this study, we examine the task and knowledge demands for teaching integer operations with representations by analyzing teaching practice. Teaching integer operations is used as an intensity case, as integer operations are challenging for students, and teachers are often required to employ several representations to teach this topic. Following a practice-based approach while also taking prior literature into consideration, we first generate a list of tasks entailed in teaching with representations and then discuss the knowledge demands imposed on teachers to successfully undertake this work. We highlight these tasks and knowledge demands by analyzing and discussing an integer addition and an integer subtraction episode for each of two teachers, Bonita and Karen. Based on our analysis, we organize the generated knowledge components using the Mathematical Knowledge for Teaching framework. We conclude by drawing implications for teacher educators and curriculum developers.  相似文献

Very little empirical research has been conducted about online career and technical education (CTE). Despite the continuing enrollment growth in online education (Lokken, 2016) and the national emphasis on CTE as a means of boosting the economy (D’Amico, Morgan, Katsinas, & Friedel, 2014), relatively little is understood about the scope and impact of online CTE at the community college. This study examined the dimensionality of morphological knowledge. The performance of 371 seventh‐ and eighth‐graders on seven morphological knowledge tasks was investigated using confirmatory factor analysis. Results suggested that morphological knowledge was best fit by a bifactor model with a general factor of morphological knowledge and seven specific factors, representing tasks that tap different facets of morphological knowledge. Next, structural equation modelling was used to explore links to literacy outcomes. Results indicated the general factor and the specific factor of morphological meaning processing showed significant positive associations with reading comprehension and vocabulary. Also, the specific factor of generating morphologically related words showed significant positive associations with vocabulary, while specific factors of morphological word reading and spelling processing showed small negative relationships to reading comprehension and vocabulary. Findings highlight the complexity of morphological knowledge and suggest the importance of being cognizant of the nature of morphology when designing and interpreting studies.  相似文献
