Imaging techniques provide ways of knowing structure and function in biology at different scales. The multidisciplinary nature and rapid advancement of imaging sciences requires imaging education to begin early in the biology curriculum. Guided by the National Institutes of Health (NIH) Roadmap initiatives, we incorporated a nanoimaging, molecular imaging, and medical imaging teaching unit into three 1-h class periods of an introductory course on ways of knowing biology. Activities were derived from NIH Roadmap initiatives in nanomedicine, regenerative medicine, and nuclear medicine. The course materials we describe contributed positively to student learning gains in quantifying and interpreting images, in characterizing imaging methods that provide ways of knowing biological structure and function, and in understanding scale in biology and imaging. The NIH Roadmap provides a useful context to educate students about the multidisciplinary imaging continuum.  相似文献

Sonoporation is a useful biophysical mechanism for facilitating the transmembrane delivery of therapeutic agents from the extracellular to the intracellular milieu. Conventionally, sonoporation is carried out in the presence of ultrasound contrast agents, which are known to greatly enhance transient poration of biological cell membranes. However, in vivo contrast agents have been observed to induce capillary rupture and haemorrhage due to endothelial cell damage and to greatly increase the potential for cell lysis in vitro. Here, we demonstrate sonoporation of cardiac myoblasts in the absence of contrast agent (CA-free sonoporation) using a low-cost ultrasound-microfluidic device. Within this device an ultrasonic standing wave was generated, allowing control over the position of the cells and the strength of the acoustic radiation forces. Real-time single-cell analysis and retrospective post-sonication analysis of insonated cardiac myoblasts showed that CA-free sonoporation induced transmembrane transfer of fluorescent probes (CMFDA and FITC-dextran) and that different mechanisms potentially contribute to membrane poration in the presence of an ultrasonic wave. Additionally, to the best of our knowledge, we have shown for the first time that sonoporation induces increased cell cytotoxicity as a consequence of CA-free ultrasound-facilitated uptake of pharmaceutical agents (doxorubicin, luteolin, and apigenin). The US-microfluidic device designed here provides an in vitro alternative to expensive and controversial in vivo models used for early stage drug discovery, and drug delivery programs and toxicity measurements.  相似文献

In the absence of any food or fluid intake during the hours of daylight during the month of Ramadan, a progressive loss of body water will occur over the course of each day, though these losses can be completely replaced each night. Large body water deficits will impair both physical and cognitive performance. The point at which water loss will begin to affect performance is not well defined, but it may be as little as 1-2% of body mass. For resting individuals in a temperate environment, the water loss that occurs during a day without food or fluid will typically amount to about 1% of body mass by the time of sunset. This small loss of body water is unlikely to have a major adverse effect on any aspect of physical or cognitive performance. Larger body water losses will occur, however, in hot weather or if exercise is undertaken. Performance in events lasting about 1 hour or longer may be impaired in the absence of fluid intake during the event. In weight-category sports, there may be difficulties due to the impossibility of restoring body water content between the weigh-in and competition, and athletes will require alternative strategies. Where more than one competition or training session takes place in a single day and where substantial fluid losses are incurred, recovery will be impaired by the absence of fluid intake.  相似文献
