TIRF microscopy has provided a means to view mobile granules within 100 nm in size in two dimensions. However quantitative analysis of the position and motion of those granules requires an appropriate tracking method. In this paper, we present a new tracking algorithm combined with the unique features of TIRF. Firstly a fluorescence correction procedure was processed to solve the problem of fluorescence bleaching over time. Mobile granules were then segmented from a time-lapse image stack by an adaptive background subtraction method. Kalman filter was introduced to estimate and track the granules that allowed reducing searching range and hence greater reliability in tracking process. After the tracked granules were located in x-y plane, the z-position was indirectly inferred from the changes in their intensities. In the experiments the algorithm was applied in tracking GLUT4 vesicles in living adipose cells. The results indicate that the algorithm has achieved robust estimation and tracking of the vesicles in three dimensions.  相似文献

Mesoscopic characteristics of a clayey soil specimen subjected to macroscopic loading are examined using a medical-use computerized tomography (CT) instrument. Disturbed state concept (DSC) theory is based on the utilization of the hardening model. DSC indirectly describes material behavior by claiming that the actual response of the material is expressed in terms of the relative intact (RI) response and the fully adjusted (FA) response. The occurrence of mesoscopic structural changes of material has similarities with the occurrence of a macroscopic response of the material under loadings. In general, the relative changing value of a softening material is three to five times more than that of a hardening material. Whether special zones exist or not in a specimen cross section does not affect the following conclusion: hardening material and softening material show mechanical differences with CT statistical indices values prominently changing, and the change is related to the superposing of a disturbance factor. A new disturbance factor evolution function is proposed. Thus, mesoscopic statistical indices are introduced to describe macroscopic behavior through the new evolution function. An application of the new evolution function proves the effectiveness of the amalgamation of a macroscopic and a mesoscopic experimental phenomenon measurement methods.  相似文献

As the idea of simulated annealing (SA) is introduced into the fitness function, an improved genetic algorithm (GA) is proposed to perform the optimal design of a pressure vessel which aims to attain the minimum weight under burst pressure constraint. The actual burst pressure is calculated using the arc-length and restart analysis in finite element analysis (FEA). A penalty function in the fitness function is proposed to deal with the constrained problem. The effects of the population size and the number of generations in the GA on the weight and burst pressure of the vessel are explored. The optimization results using the proposed GA are also compared with those using the simple GA and the conventional Monte Carlo method.  相似文献

We present a semi-custom design methodology based on transistor tuning to optimize the design performance Compared with other transistor tuning approaches, our tuning process takes the cross-talk effect into account and prominently reduces the complexity for circuit simulation and analysis by decomposing the circuit network utilizing graph theory. Furthermore, the incremental placement and routing for the corresponding transistor tuning in conventional approaches is not required in our methodology, which might induce timing graph variation and additional iterations for design convergence. This methodology combines the flexible automated circuit tuning and physical design tools to provide more opportunities for design optimization throughout the design cycle.  相似文献

Hemin can improve the stress resistance of plants through the heme oxygenase system. Additionally, substances contained in plants, such as secondary metabolites, can improve stress resistance. However, few studies have explored the effects of hemin on secondary metabolite content. Therefore, the effects of hemin on saponin synthesis and the mechanism of plant injury relief by hemin in Conyza blinii were investigated in this study. Hemin treatment promoted plant growth and increased the antioxidant enzyme activity and saponin content of C. blinii under osmotic stress and cold stress. Further study showed that hemin could provide sufficient precursors for saponin synthesis by improving the photosynthetic capacity of C. blinii and increasing the gene expression of key enzymes in the saponin synthesis pathway, thus increasing the saponin content. Moreover, the promotion effect of hemin on saponin synthesis is dependent on heme oxygenase-1 and can be reversed by the inhibitor Zn-protoporphyrin-IX(ZnPPIX). This study revealed that hemin can increase the saponin content of C. blinii and alleviate the damage caused by abiotic stress, and it also broadened the understanding of the relationship between hemin and secondary metabolites in plant abiotic stress relief.  相似文献
