MoFLP1, encoding a novel fungal fasciclin-like protein, is involved in conidiation and pathogenicity in Magnaporthe oryzae

Fasciclin family proteins have been identified as cell adhesion molecules in various organisms. In this study, a novel Magnaporthe oryzae fasciclin-like protein encoding gene, named MoFLP1, was isolated from a subtractive suppressive cDNA library and functionally analyzed. Sequence analysis showed that the MoFLP1 gene contains an open reading frame (ORF) of 1050 nucleotides encoding 349 amino acids with a calculated molecular weight of 35.85 kDa and a pI of 7.76. The deduced MoFLP1 protein contains a 17-amino acid secretion signal sequence and an 18-amino acid sequence with the characteristics of a glycosylphosphotidylinositol (GPI) anchor additional signal at its N- and C-terminuses, respectively. Potential N-glycosylation sites and domains involving cell adhesion were also identified in MoFLP1. Sequence analysis and subcellular localization by the expression of MoFLP1-GFP fusion construct in M. oryzae indicated that the MoFLP1 protein is probably localized on the vacuole membrane. Two MoFLP1 null mutants generated by targeted gene disruption exhibited marked reduction of conidiation, conidial adhesion, appressorium turgor, and pathogenicity. Our results indicate that fasciclin proteins play important roles in fungal development and pathogenicity in M. oryzae. Magnetic cell sorting and flow cytometry sorting methods for the isolation and function analysis of mouse CD4＋ CD25＋ Treg cells

Objective: In this paper we compared the two methods of cell sorting (magnetic cell sorting and flow cytometry sorting) for the isolation and function analysis of mouse CD4~+ CD25~+ regulatory T (Treg) cells, in order to inform further studies in Treg cell function. Methods: We separately used magnetic cell sorting and flow cytometry sorting to identify CD4~+ CD25~+ Treg cells. After magnetic cell separation, we further used flow cytometry to analyze the purity of CD4~+ CD25~+ Treg cells, trypan blue staining to detect cell viability, and propidium iodide (PI) staining to assess the cell viability. We detected the immune inhibition of CD4~+ CD25~+ Treg cells in the in vitro proliferation experiments. Results: The results showed that compared to flow cytometry sorting, magnetic cell sorting took more time and effort, but fewer live cells were obtained than with flow cytometry sorting. The CD4~+ CD25~+ Treg cells, however, obtained with both methods have similar immunosuppressive capacities. Conclusion: The result suggests that both methods can be used in isolating CD4~+ CD25~+ Treg cells, and one can select the best method according to specific needs and availability of the methodologies.

一种传感器网络目标定位的建模方法

本文提出一种传感器网络目标定位系统的系统模型,然后利用PtolemyⅡ建模仿真平台完成该传感器网络目标定位系统的系统级行为描述,并根据系统级建模方法学对该目标定位系统进行建模与仿真,分析了传感器网络目标定位系统的诸多影响因素.测试结果表明:传感器网络目标定位系统对于传感器节点的功耗及信息时间同步窗口具有敏感性,符合传感器网络目标定位系统应用特性,验证了Ptolemy嵌入式系统级设计方法学对于指导传感器网络系统设计的正确性和可行性.     

精品课网络教学系统建设的研究与实践

精品课程建设是提高教学质量的重要手段.而基于网络的多媒体教学系统又是精品课程建设非常重要的环节.借助网络进行教学资源建设、辅助教学,是现代教学方式区别于传统教学方式的重要方面.本文提出了面向精品课程资源整合与信息发布的网络教学系统的研究方案.用以辅助和延伸课堂教学,提高教学质量,从而达到现代化精品课程建设的要求.     

基于粗集理论的教师教学水平分析

借助粗集理论,以学生成绩为指标,分别以教师的普通话水平、课前备课情况、教师的年龄、学历、性别以及教师上课的仪表为条件属性,对四川某高校的在校大学生的学习成绩进行问卷调查,并加以归一化处理和仔细分析.研究结果表明,各条件属性对结果的重要性有着显著的差别.     

激光与等离子体相互作用中的孤立波简化模型研究

强激光在等离子体中传播时,从麦克斯韦方程组出发,考虑相对论效应后,得到了圆偏振电磁横波与尾波相互作用的耦合方程组,并采用一个物理上合理的假设来求解这个耦合方程组,分析了两种简化的模型,分别进行数值模拟,得出了在以激光传播速度运动的结构中,若选取合适的系统参数,亚浓密等离子体和过浓密等离子体中都存在孤立波或孤立子.

论嫖宿幼女罪的立法缺陷及完善——以贵州“习水案”为线索的分析

嫖宿幼女罪在立法上存在缺陷.从犯罪构成上来讲,嫖宿幼女罪与奸淫幼女罪(现为强奸罪)、猥裹儿童罪客观要件完全或部分重叠,客观上赋予了幼女的性自主权,与保护幼女权益的立法目的相背离;从犯罪的停止形态上来看,未遂和既遂难以区分;从罪数来讲,想象竞合和法条竞合处理原则相冲突;从刑罚上看,配刑不科学,违反罪责刑相适应的刑法原则;在司法实践中也易放纵犯罪分子.应该通过刑法修正案的形式废止嫖宿幼女罪,将其纳入奸淫幼女罪和猥亵儿童罪中.
