利用UAS/hand-gal4系统构建果蝇心脏Nmnat基因特异性过表达,研究心脏Nmnat基因过表达联合耐力运动对高脂膳食诱发脂毒性心肌症的影响,并分析其分子机制。方法:雄性w^1118和NmnatUAS系果蝇分别与雌性hand-Gal4杂交,收集F1代心脏Nmnat正常表达(hand>w^1118)和过表达(hand>Nmnat)处女蝇,饲养至15 d后开始进行耐力运动(E)和高脂膳食(HFD),并分为hand>w^1118、hand>w^1118+E、hand>w^1118+HFD、hand>w^1118+HFD+E、hand>Nmnat、hand>Nmnat+E、hand>Nmnat+HFD、hand>Nmnat+HFD+E共8组,每组410只。ELISA检测心脏TAG、NAD+、SIR2蛋白去乙酰化、SOD活力和MDA水平,qRT-PCR检测心脏相关基因mRNA表达,M-mode检测心脏功能情况。结果:hand>w^1118+HFD+E果蝇心脏的Nmnat、dFoxo、bmm表达、NAD+水平、SIR2去乙酰化、SOD活力、缩短分数高于hand>w^1118+HFD(P<0.05或P<0.01),且d FAS和Col4al表达、TAG和MDA水平、心率和纤维性振颤低于hand>w^1118+HFD(P<0.05或P<0.01);hand>Nmnat果蝇心脏各指标与hand>Nmnat+HFD相比较未见显著差异(P>0.05);hand>Nmnat+HFD+E果蝇心脏的Nmnat、dFoxo、bmm表达、心脏NAD+水平、SIR2去乙酰化、SOD活力、心脏缩短分数高于hand>Nmnat+HFD(P<0.05或P<0.01),且dFAS和Col4al表达\TAG和MDA水平、心率和纤维性振颤低于hand>Nmnat+HFD(P<0.05或P<0.01)。结论:心脏Nmnat过表达和耐力运动均能抵抗果蝇高脂膳食诱发的心脏脂质过度堆积、氧化损伤、减弱心脏收缩能力和增加纤维性振颤,其分子机制与心脏Nmnat/NAD+/SIR2/dFoxo通路活性增强有关。心脏Nmnat基因过表达联合耐力运动能更好地降低脂毒性心肌症的发生概率,其机制与二者联合对心脏Nmnat/NAD+/SIR2/dFoxo通路活性上调的叠加作用有关。 相似文献

Classification is a defining feature of Para-sport, and sports-specific classification systems determined through multidisciplinary scientific research are required, i.e., evidence-based and focused on the relationship between the impairment and the key performance determinants. Data envelopment analysis (DEA) was applied as a classification tool using a directional distance function (DDF) model. The aim of the study was to test the DEA as a possible classification tool in cerebral palsy football. We analyse the performance of 56 international para-footballers with hypertonia, ataxia or athetosis, who completed a 20-test battery with DEA models. Five of the tests are included in the model (change of direction: Illinois agility test; jumping: standing broad jump, four bounds for distance, and triple hop with the non-dominant leg; 10-m sprint/acceleration; and ball dribbling, both in a straight line and following a trajectory), showing that players with less impairment exhibit the highest efficiency. This outcome suggests that DEA models might be feasible for detecting and discriminating the performance and magnitude of impairment in cerebral palsy football, with an objective ranking of the athletes in relation to different physical performance tests. This study also provides reference scores for decision-making during classification and guidance for further research in team Paralympic sports. 相似文献

Aim was to identify critical load (CL) in young and elderly apparently healthy male cohorts. To contrast the metabolic, cardiovascular and perceptual responses on CL according to age. We evaluated 12 young (23 ± 3 years) and 10 elderly (70 ± 2 years) apparently healthy active males, who underwent: (1) 1 repetition maximum (1RM) test on a 45° Leg Press; (2) on different days, three high-intensity resistance exercise constant load tests (60%, 75% and 90% 1RM) until fatigue (Tlim). Absolute values of both the CL asymptote and curvature constant (kg) were significantly lower in elderly subjects (P < 0.05). In contrast, elderly subjects demonstrated a significantly higher number of repetitions at CL when compared with young subjects (P < 0.05). As expected, oxygen uptake (VO2) and heart rate (HR) during maximal aerobic exercise testing were significantly reduced in older subjects. However, percent-predicted aerobic capacity were higher in older subjects (P < 0.05). In addition, blood lactate ([La?]) corrected to Tlim and rating of perceived exertion values were greater in younger subjects at all intensities (P < 0.05). These findings, despite reduced force production in older subjects, endurance-related parameters are well preserved according to age-adjusted percent-predicted values in apparently healthy males. 相似文献

The purpose of this study was to identify which averaging methods most accurately measures peak cardiorespiratory fitness (CRF) parameters [peak O2 uptake (VO2), peak O2pulse and peak respiratory exchange ratio (RER)] in a sample of healthy children and adolescents. In this cross-sectional multicenter study, we recruited 278 healthy children aged 12–17 years. We compared the mean peak value of three CRF parameters using the recommended averaging methods (30-second block average) with alternative averaging methods such as moving averages or shorter smoothing periods. We also assessed averaging methods for accuracy by individually reviewing breath-by-breath scatter plots. The 30-second block average method resulted in a lower mean peak VO2 and in an increased proportion of underestimated peak values. Using a 30-second moving average significantly increased mean peak values which increased accuracy. Similar results were found for peak RER and peak O2pulse. In conclusion, the currently recommended averaging method (30-second block average) increased the risk of misinterpretation of peak CRF values in children. According to Allport's (1954) model of socialization of prejudice the level of prejudice of parents and their offspring should co-vary due to the offspring's adaptation to the parents’ attitudes. Available empirical studies and literature reviews support Allport's assumption. Modern models of socialization give reason to extend Allport's unidirectional influence model to a bidirectional perspective which assumes that parents influence their offspring's attitudes and are simultaneously influenced by them. In a cross-sectional study, 408 parent–offspring dyads (64% female parents, 55% female offspring, mean age of children = 16.47 years, SD 1.87, parents = 42.08 years, SD 7.08) from the ethnic majority population in Costa Rica were asked about their prejudice towards immigrants and ethnic minorities in Costa Rica. Using structural equation modelling, unidirectional and bidirectional models of prejudice determination were tested. The data clearly support the unidirectional model of prejudice transmission from parents to offspring, even if moderator effects of sex, age, and importance of contact are taken into account. 相似文献

Near infrared reflectance (N1R) spectroscopy is as a rapid, convenient and simple nondestructive technique useful for quantifying several soil properties. This method was used to estimate nitrogen (N) and organic matter (OM) content in a soil of Zhejiang Province, Hangzhou County. A total of 125 soil samples were taken from the field. Ninety-five samples spectra were used during the calibration and cross validation stage. Thirty samples spectra were used to predict N and OM concentration. NIR spectra of these samples were correlated using partial least square regression. The regression coefficients between measured and predicted values of N and OM was 0.92 and 0.93, and SEP (standard error of prediction) were 3.28 and 0.06, respectively, which showed that NIR method had potential to accurately predict these constituents in this soil. The results showed that NIR spectroscopy could be a good tool for precision farming application. 相似文献
