Background:The adhesion of monocytes to the endothelium following accumulation oflow-density lipoprotein(LDL) in subendothelial spaces is an important step in the development of intimal hyperplasia in arterially implanted vein grafts and atherosclerosis in both animals and humans.However.it iS not well known how serum factors affect the adhesion of monocytes.Methods:We have studied the efrect of fetal calf serum(FCS).which we considered a source of LDL.on the adhesion of monocytes to endothelial cells(Ecs)by using human monocytic THP-1 cells and both a monolayer of cultured bovine aortic endothelial cells(EC monoculture)and a co-culture with bovine aortic smooth muscle cells(EC-SMC co-culture).Results:It was found that the addition of FCS to the medium greatly affected the adhesion of THP-1 cells,and the higher the concentration of FCS in the medium,the greater the adhesion of THP-1 cells to endothelial cells.Adhesion of THP-1 cells to an EC-SMC co-culture Was approximately twofold greater than that to an EC monoculture,and after adhering to endothelial cells,many THP-1 cells transmigrated into the layer of smooth muscle cells.Conclusion:The results suggest that the elevation of the LDL(cholesterol)level in blood provides a favorable condition for the development of intimal hyperplasia and atherosclerosis by promoting the adhesion of monocytcs to the endothelium and their subsequent migration into subendothelial spaces.  相似文献

本文运用Panel Data模型对上海市城市居民不同收入水平下用于食品、交通通讯、文教娱乐、衣着、居住、医疗保健、家庭用品及服务等消费品的边际消费倾向进行了实证研究。研究结果表明:对于不同消费品,上海市城市居民不同收入家庭的边际消费倾向各不相同。总体而言,用于食品的边际消费倾向最高。交通通讯的边际消费倾向基本处于第二、第三位。文教娱乐的边际消费倾向总体上排在第三位及以前。衣着的边际消费倾向基本保持在第四和第五位。居住问题是上海市城市居民需要考虑的排序第四、第五位的消费问题,但比衣着的边际消费倾向稍微小一些。家庭用品及服务的边际消费倾向总体上排序在第六位。医疗保健的边际消费倾向是所有消费品中最低的。最后,依据研究结论给出几点关于相关行业发展的启示。  相似文献

Soybean (Glycine max (L.) Merrill) is a salt-sensitive crop, and its production is severely affected by saline soils. Therefore, the response of soybean seeds to salt stress during germination was investigated at both physiological and proteomic levels. The salt-tolerant cultivar Lee68 and salt-sensitive cultivar N2899 were exposed to 100 mmol/L NaCl until radicle protrusion from the seed coat. In both cultivars, the final germination percentage was not affected by salt, but the mean germination times of Lee68 and N2899 were delayed by 0.3 and 1.0 d, respectively, compared with controls. In response to salt stress, the abscisic acid content increased, and gibberellic acid (GA1+3) and isopentenyladenosine decreased. Indole-3-acetic acid increased in Lee68, but remained unchanged in N2899. The proteins extracted from germinated seeds were separated using two-dimensional gel electrophoresis (2-DE), followed by Coomassie brilliant blue G-250 staining. About 350 protein spots from 2-DE gels of pH range 3 to 10 and 650 spots from gels of pH range 4 to 7 were reproducibly resolved, of which 18 protein spots showed changes in abundance as a result of salt stress in both cultivars. After matrix-assisted laser desorption ionization-time of flight-mass spectroscopy (MALDI-TOF-MS) analysis of the differentially expressed proteins, the peptide mass fingerprint was searched against the soybean UniGene database and nine proteins were successfully identified. Ferritin and 20S proteasome subunit β-6 were up-regulated in both cultivars. Glyceraldehyde 3-phosphate dehydrogenase, glutathione S-transferase (GST) 9, GST 10, and seed maturation protein PM36 were down-regulated in Lee68 by salt, but still remained at a certain level. However, these proteins were present in lower levels in control N2899 and were up-regulated under salt stress. The results indicate that these proteins might have important roles in defense mechanisms against salt stress during soybean seed germination.  相似文献

To evaluate the therapeutic efficiency of tissue-engineered human corneal endothelia (TE-HCEs) on rabbit primary corneal endotheliopathy (PCEP), TE-HCEs reconstructed with monoclonal human corneal endothelial cells (mcHCECs) and modified denuded amniotic membranes (mdAMs) were transplanted into PCEP models of New Zealand white rabbits using penetrating keratoplasty. The TE-HCEs were examined using diverse techniques including slit-lamp biomicroscopy observation and pachymeter and tonometer measurements in vivo, and fluorescent microscopy, alizarin red staining, paraffin sectioning, scanning and transmission electron microscopy observations in vitro. The corneas of transplanted eyes maintained transparency for as long as 200 d without obvious edema or immune rejection. The corneal thickness of transplanted eyes decreased gradually after transplanting, reaching almost the thickness of normal eyes after 156 d, while the TE-HCE non-transplanted eyes were turbid and showed obvious corneal edema. The polygonal corneal endothelial cells in the transplanted area originated from the TE-HCE transplant. An intact monolayer corneal endothelium had been reconstructed with the morphology, cell density and structure similar to those of normal rabbit corneal endothelium. In conclusion, the transplanted TE-HCE can reconstruct the integrality of corneal endothelium and restore corneal transparency and thickness in PCEP rabbits. The TE-HCE functions normally as an endothelial barrier and pump and promises to be an equivalent of HCE for clinical therapy of human PCEP.  相似文献

Maintenance of cellular homeostasis and genome integrity is a critical responsibility of DNA double-strand break(DSB)signaling.P53-binding protein 1(53BP1)plays a critical role in coordinating the DSB repair pathway choice and promotes the non-homologous end-joining(NHEJ)-mediated DSB repair pathway that rejoins DSB ends.New insights have been gained into a basic molecular mechanism that is involved in 53BP1 recruitment to the DNA lesion and how 53BP1 then recruits the DNA break-responsive effectors that promote NHEJ-mediated DSB repair while inhibiting homologous recombination(HR)signaling.This review focuses on the up-and downstream pathways of 53BP1 and how 53BP1 promotes NHEJ-mediated DSB repair,which in turn promotes the sensitivity of poly(ADP-ribose)polymerase inhibitor(PARPi)in BRCA1-deficient cancers and consequently provides an avenue for improving cancer therapy strategies.  相似文献
