间歇运动激活心梗大鼠心肌NRG1-PI3K/Akt通路抑制心肌细胞凋亡

摘要：目的: 探讨心脏NRG1在间歇运动激活心梗(myocardial infarction,MI)大鼠心肌NRG1-PI3K/Akt通路抑制心肌细胞凋亡的作用。方法：雄性SD大鼠48只,随机分为假手术组(S),心梗安静组(MI),心梗+间歇运动组(ME),心梗+间歇运动+抑制剂组(MEA),每组12只。采用左冠脉前降支结扎法建立MI模型。S组手术仅穿线不结扎。ME组和MEA组在MI术后1wk进行跑台间歇运动。运动开始速度为10m/min运动10min后,速度逐渐增至25m/min×7 min,再以15 m/min×3min运动,之后依次交替进行。60min×1次/d,5d/1wk×8wk。训练结束后次日,腹腔麻醉,颈动脉插管测定LVSP、LVEDP及±dp/dtmax。之后开胸摘取心脏,进行组织学制片,Masson染色。荧光实时定量PCR测定心肌NRG1及其受体ErbB2/4表达。Western Blot法检测心肌NRG1、ErbB2/4、PI3K/Akt、Bcl-2和Bax蛋白含量。Caspase3表达及TUNEL检测观察分析心肌细胞凋亡情况。结果: MI组胶原容积百分比(CVF)和LVEDP较S组显著升高(P<0.01,P<0.01),LVSP和±dp/dtmax显著降低(P<0.01,P<0.01)。且MI后心肌ErbB4 mRNA表达显著降低(P<0.01),心肌NRG1、ErbB2和ErbB4蛋白表达显著降低(P<0.05,P<0.01,P<0.01),PI3K/Akt蛋白表达及Bcl-2/Bax比值显著降低(P<0.01,P<0.01),TUNEL阳性细胞数和Caspase3活力显著增加(P<0.01,P<0.01)。ME组CVF和LVEDP较MI组显著降低(P<0.01,P<0.01),LVSP和±dp/dtmax显著升高(P<0.01,P<0.01),心肌NRG1、ErbB2和ErbB4 mRNA及蛋白表达均显著增加(P<0.01,P<0.01,P<0.01),PI3K/Akt蛋白表达和Bcl-2/Bax比值显著增加(P<0.01,P<0.01),TUNEL阳性细胞数和Caspase3活力显著降低(P<0.01,P<0.01),且运动效应被抑制剂AG1478显著减弱。结论:间歇运动可通过激活心肌NRG1及其受体表达,上调PI3K/Akt蛋白表达,抑制心肌细胞凋亡和胶原过度增生,改善心功能。     

Injectable hybrid inorganic nanoscaffold as rapid stem cell assembly template for cartilage repair

Cartilage injuries are often devastating and most cannot be cured because of the intrinsically low regenerative capacity of cartilage tissues. Although stem-cell therapy has shown enormous potential for cartilage repair, the therapeutic outcome has been restricted by low survival rates and poor chondrocyte differentiation in vivo. Here, we report an injectable hybrid inorganic (IHI) nanoscaffold that facilitates fast assembly, enhances survival and regulates chondrogenic differentiation of stem cells. IHI nanoscaffolds that strongly bind to extracellular matrix (ECM) proteins assemble stem cells through synergistic 3D cell–cell and cell–matrix interactions, creating a favorable physical microenvironment for stem-cell survival and differentiation in vitro and in vivo. Additionally, chondrogenic factors can be loaded into nanoscaffolds with a high capacity, which allows deep, homogenous drug delivery into assembled 3D stem-cell-derived tissues for effective control over the soluble microenvironment of stem cells. The developed IHI nanoscaffolds that assemble with stem cells are injectable. They also scavenge reactive oxygen species and timely biodegrade for proper integration into injured cartilage tissues. Implantation of stem-cell-assembled IHI nanoscaffolds into injured cartilage results in accelerated tissue regeneration and functional recovery. By establishing our IHI nanoscaffold-templated 3D stem-cell assembly method, we provide a promising approach to better overcoming the inhibitory microenvironment associated with cartilage injuries and to advance current stem-cell-based tissue engineering.

Beyond the Blockbuster: Good Exhibitions in Small Packages

Many American and European art museums are now featuring small, highly-focused shows in their exhibition programs. In 1990, the Indianapolis Museum of Art organized an exhibition that reunited, for the first time in a century, the four landscape paintings created by neo-impressionist Georges Seurat during the last summer of his life. Using Seurat at Gravelines: The Last Landscapes as an example, this article addresses the advantages — for museums and their visitors — of the small temporary exhibition.     

Curriculum-based measures of written expression for learning-disabled and nondisabled students

This study examined differences in written expression between a sample of learning-disabled and nondisabled middle school students, matched by grade and sex, using eight curriculum-based measures. All of the learning-disabled students had been identified as having written language deficits as part of their handicapping conditions. The nondisabled students showed superior written expression skills, especially on the production-independent measures. Implications of these results are discussed in relation to psycho educational assessment, the use of CBM for ongoing assessment of written expression, and future studies.     

Creating information resources and trainings for farmworker-serving community health workers

Background:Farmworker-serving community health workers have limited access to farmworker health research findings, training, and education resources. With funding from the National Library of Medicine, we are working to improve the health information literacy of both community health workers and farmworkers. We conducted focus group discussions with community health workers to explore their experiences providing health education and information to farmworkers, their information-seeking behaviors, and their technology and information needs. Data from the focus groups provided insights into the main areas in which community health workers would like to receive professional development.Case Presentation:Our team, which includes health sciences librarians, developed a resource list of educational materials for farmworker health, videos to increase community health workers'' skills finding health information online, and webinars to introduce these resources to community health workers. Videos, available in Spanish and English, included instruction on finding and evaluating online health information, accessing reputable online consumer health information sources, and advanced searching tips for Google and PubMed. Through three webinars, we introduced the resource list, videos, and design software for creating handouts and infographics to community health workers.Conclusions:Community health workers have a critical role in providing health education and information to farmworkers, and our efforts represent a first step in addressing community health workers'' limited access to professional development. Health sciences librarians are well positioned to partner with interdisciplinary teams working to reduce health disparities and provide resources and training to community health workers, farmworkers, and other underserved communities.
