Cloning and efficient expression of <Emphasis Type="Italic">Bacillus sp.</Emphasis> BH072 <Emphasis Type="Italic">tasA</Emphasis> gene in <Emphasis Type="Italic">Escherichia coli</Emphasis>

The Bacillus strain BH072 isolated from a honey sample showed strong antifungal activity against phytopathogen. Gene cloning test demonstrated that the strain had a tasA gene encoding an antifungal TasA protein. Although the wild strain simultaneously produced various antifungal substances, only the physicochemical property and antifungal activity of TasA protein were unclear due to the difficulty in extraction. In this study, tasA gene encoding the protein from Bacillus sp. BH072 was amplified by using the polymerase chain reaction (PCR) method and cloned into pET 28a (+) vector, and then expressed in host cells Escherichia coli BL21 (DE3). The expressed proteins were collected by centrifugation and ultrasonic treatment, and then purified by using nickel-nitrilotriacetic acid (Ni-NTA) metal affinity column and dialysis methods. The result of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) test showed that an expected protein band appeared with a size of 31 kDa. The expressed products possessed antifungal activity against the phytopathogenic indicator strain Botrytis cinerea. A genetically engineered strain tasA of E. coli was established in this study which can efficiently express Tas A protein.

研究WLED用红色荧光粉体材料Eu3+'Bi3+∶NaLu(MoO4)2.采用燃烧法制备一系列NaLu(0.95-x)Eu0.05Bix(MoO4)2(x=0,0.01,0.05,0.10,0.15,0.20,0.25)样品,通过多种表征手段来确定Bi3+的最佳含量并分析了激活离子发光性能的变化规律和机制.结果表明:在Eu3+的掺杂量为5a%条件下,当Bi3+的含量达到20a%左右、粒径控制在40-55μm之间时,材料的发光性能趋于最佳.     

AtHsfAla 对拟南芥幼苗在干旱中生理指标的影响

以过量表达 AtHsfAla 及 AtHsfAla 基因沉默的转基因植株为材料,通过控制土壤水分进行干旱处理,结果显示,过量表达型干旱处理后的存活率最大,而基因沉默型最小,表明 AtHsfA1a 能增强植物的抗旱性。为了从生理水平研究 AtHsfA1a 提高抗旱性的机制,研究发现过量表达型的叶片相对含水量和叶绿素含量最高,而沉默型最低。不同基因型植株的 H2 O2和 MDA 的含量正好相反,说明 AtHsfA1a 通过保护细胞膜而减小细胞所受氧化伤害来提高抗旱能力。

General University Requirements at Hong Kong Polytechnic University: evaluation findings based on student focus groups

Under the new four-year undergraduate programme, a general education framework titled ‘General University Requirements’ (GUR) has been developed and implemented since 2012/2013 at Hong Kong Polytechnic University (PolyU). To evaluate the implementation and effectiveness of the GUR in its first year, focus group interviews with students were carried out. In 13 focus groups, 62 first-year students from eight faculties/schools who took GUR subjects in 2012/2013 academic year were interviewed by trained researchers. Results showed that students generally had good perceptions of the GUR in terms of the subject content, teaching methods and the overall effectiveness of the subjects. The interactive teaching and learning initiatives adopted in GUR subjects were particularly appreciated, although students expressed minor concerns on the administration of GUR subjects. The results suggest that the first-year implementation of the GUR at PolyU was generally smooth and successful from the perspectives of the students.     

Changes in children’s anger,sadness, and persistence across blocked goals: Implications for self-regulation

The current study took a person-centered approach to examine the heterogeneity of changes in children’s emotions and persistence during a goal-blocking task and examined how different profiles of emotions and persistence related to children’s self-regulation. Children’s anger, sadness, and persistence were rated in a goal-blocking task in toddlerhood (T1; N = 140, 72 boys, M_age = 2.67 years, 90.7% White) and preschool (T2). Children’s self-regulation, specifically sustained attention and engagement, was assessed at T1, T2, and early school-age (T3) from 2005 to 2012. Growth mixture modeling revealed two classes of children at T1 and three classes at T2 with different patterns of anger, sadness, and persistence. Children’s classification at T2, but not T1, significantly predicted their sustained attention and engagement both concurrently and longitudinally.     

LD夜视成像探测中基于PSF的超分辨率重建技术研究

夜视目标识别在视频监控中起着非常重要的作用。从早期的红外热成像发展至今,激光夜视成像系统被广泛的应用。它不仅受到大气吸收和散射特性的影响,还与光学系统中的透镜及传感器的衍射极限有关。在既有成本下,通过图像复原和超分辨率重建,可以尽可能的提高夜视成像的成像质量。很多学者的研究结果表明,成像系统的点扩散函数作为先验知识,可以提高图像复原的质量。因此,本文在最大后验概率框架下,实现点扩散函数与凸集投影超分辨率重建相结合,并应用到半导体激光夜视成像探测系统中,客观的图像质量评价标准被用来决定迭代次数。目标识别的实验结果表明,用本文提出的重建方法可以有效提高半导体激光夜视成像的范围和成像质量。

在高职食品加工技术专业中探索出“生产教学化,教学生产化”的工学结合人才培养模式,即以“食品加工高新技术中试工程中心”为依托建立了几条柔性中试生产线,模拟企业提供不同的岗位,供学生轮训。教学内容围绕企业委托的中试项目（产品）等现实任务展开从而实现生产教学化,教学生产化。教学实践表明,此模式有利于提高学生的综合素质能力。
