Objective: To investigate the mechanism of carbapenem resistance and the occurrence of plasmid-mediated quinolone resistance determinants qnr and aac(6')-Ib-cr in a clinical isolate of Enterobacter cloacae. Methods: An ertapenem-resistant E. cloacae ZY106, which was isolated from liquor puris of a female gastric cancer patient in a Chinese hospital, was investigated. Antibiotic susceptibilities were determined by agar dilution method. Conjugation experiments, isoelectric focusing, polymerase chain reaction (PCR), and DNA sequence analyses of plasmid-mediated carbapenemases and quinolone resistance determinants were preformed to confirm the genotype. Outer membrane proteins (OMPs) were examined by urea-sodium dodecyl sulfate-polyacrylamide gel electrophoresis (Urea-SDS-PAGE). Results: Minimum inhibitory concentrations (MCs) of imipenem, mer-openem, and ertapenem for ZY106 were 2,4, and 16 ug/ml, respectively. Conjugation studies with Escherichia coli resulted in the transfer of significantly reduced carbapenem susceptibility. ZY106 produced IMP-1 metallo-p-lactamase and CTX-M-3 extended-spectrum P-lactamase, and E. coli transconjugant produced IMP-1. Plasmid-mediated quinolone resistance determinant qnrSI was detected in ZY106. Transfer of the qnrSI-encoding-plasmid into E. coli by conjugation resulted in intermediate resistance to ciprofloxacin in E. coli transconjugant. Urea-SDS-PAGE analysis of OMPs showed that ZY106 lacked an OMP of approximately 38 KDa. Conclusion: It is the first IMP-1-producing Enterobacteriaceae in China and the first report of a clinical isolate that harbors both blaIMP and qnrS genes as well. The blaIMP-1, blaCTX-M-3, and qnrSl are encoded at three different plasmids. IMP-1 combined with the loss of an OMP possibly resulted in ertapenem resistance and reduced imipenem and mero-penem susceptibility in E. cloacae.  相似文献

Abstract

Several nutritional strategies can optimize muscle bulk and strength adaptations and enhance recovery from heavy training sessions. Adequate energy intake to meet the needs of training and carbohydrate intake sufficient to maintain glycogen stores (>7 g carbohydrate·kg?1·day?1 for women; >8 g carbohydrate·kg?1·day?1 for men) are important. Dietary protein intake for top sport athletes should include some foods with high biological value, with a maximum requirement of approximately 1.7 g·kg?1·day?1 being easily met with an energy sufficient diet. The early provision of carbohydrate (>1 g·kg?1) and protein (>10 g) early after an exercise session will enhance protein balance and optimize glycogen repletion. Creatine monohydrate supplementation over several days increases body mass through water retention and can increase high-intensity repetitive ergometer performance. Creatine supplementation can enhance total body and lean fat free mass gains during resistance exercise training; however, strength gains do not appear to be enhanced versus an optimal nutritional strategy (immediate post-exercise protein and carbohydrate). Some studies have suggested that β-OH-methyl butyric acid (β-HMB) can enhance gains made through resistance exercise training; however, it has not been compared “head to head” with optimal nutritional practices. Overall, the most effective way to increase strength and bulk is to perform sport-specific resistance exercise training with the provision of adequate energy, carbohydrate, and protein. Creatine monohydrate and β-HMB supplementation may enhance the strength gains made through training by a small margin but the trade-off is likely to be greater bulk, which may be ergolytic for any athlete participating in a weight-supported activity.  相似文献

Abstract

It is currently unknown whether hypoxia training can effectively suppress overweight and hyperinsulinemia in genetically obese animals. In this study, both lean and obese Zucker rats were randomly assigned into the following groups: control (CON, n=7), exercise training (EX, n=7), hypoxia (HYP, n=7) and exercise training with hypoxia recovery (EX+HYP, n=7). During a 6-week training period, rats performed swimming exercise progressively from 30 to 180 min·day?1, and recovered under hypoxia (14% oxygen for 8 h·day?1). Obese Zucker rats exhibited substantially greater fasting insulin levels, and exaggerated glucose and insulin responses following an oral glucose challenge compared with lean rats. At the beginning of week 6, body weight, fasting glucose, fasting insulin, area under curve of glucose (GAUC) and insulin (IAUC) in the EX+HYP group were significantly lower than CON group among the obese rats. Meanwhile, only GAUC was significantly lower in the EX group compared to the CON group. At the end of week 6, capillaries to fibre ratio (C/F), capillary density (CD) and type IIa fibre proportion of the plantaris muscle in the EX group were significantly greater than the CON group (P<0.05), but no additive effect of hypoxia on exercise training was observed. Our data demonstrate that exercise training with prolonged hypoxia recovery offers better metabolic benefits than exercise training alone for the obese Zucker rats. This advantage was closely associated with effective weight reduction.  相似文献

Technical change is one of many factors underpinning success in elite, fixation/diversification stage performers. Surprisingly, however, there is a dearth of research pertaining to this process or the most efficacious methods used to bring about such a change. In this paper we highlight the emergent processes, yet also the lack in mechanistic comprehension surrounding technical change, addressing issues within the motor control, sport psychology, coaching and choking literature. More importantly, we seek an understanding of how these changes can be made more secure to competitive pressure, and how this can be embedded within the process of technical change. Following this review, we propose the Five-A Model based on successful coaching techniques, psychosocial concomitants, the avoidance of choking and principles of effective behaviour change. Specific mechanisms for each stage are discussed, with a focus on the use of holistic rhythm-based cues as a possible way of internalising changes. Finally, we suggest the need for further research to examine these five stages, to aid a more comprehensive construction of the content and delivery of such a programme within the applied setting.  相似文献

Abstract

Although caffeine is a widely used ergogenic resource, some information regarding its effects on resistance exercises is still lacking. The objective of the present study was to verify the acute effect of the ingestion of two different doses of caffeine on performance during a session of resistance exercises and to analyze the perception of the subjects in relation to the intake of caffeine. Following a double-blind, randomised, cross-over, controlled, and non-placebo design, 14 trained and healthy men (24.7?±?6.8 years; 79.8?±?9.8?kg; 177.3?±?8.5?cm) performed a training session in chest-press, shoulder-press, and biceps curl exercises (3 sets until exhaustion; 70% 1RM; 3 min rest interval; 2?s for each concentric and eccentric phase) on three non-consecutive days after ingestion of 3?mg.kg?1 caffeine (CAF3), 6?mg.kg?1 caffeine (CAF6), or no substance (CON). Subjects were informed that one of the caffeine doses would be placebo. The total number of repetitions performed in CON (93.6?±?22.4) was significantly lower than in CAF3 (108.0?±?19.9, P?=?0.02) and in CAF6 (109.3?±?19.8, P?=?0.03) and there were no differences between caffeine doses. Eight subjects noticed that caffeine was in CAF3 and six in CAF6 and there were no differences in the number of repetitions between sessions in which the subjects perceived and did not perceive caffeine. In conclusion, caffeine doses of 3 or 6?mg.kg?1 similarly increased performance in resistance upper limb exercises, independent of the subject's perception of substance ingestion.  相似文献

Strength training with isometric contractions produces large but highly angle-specific adaptations. To contrast the contractile mode of isometric versus dynamic training, but diminish the strong angle specificity effect, we compared the strength gains produced by isometric training at four joint angles with conventional dynamic training. Thirty-three recreationally active healthy males aged 18?–?30 years completed 9 weeks of strength training of the quadriceps muscle group three times per week. An intra-individual design was adopted: one leg performed purely isometric training at each of four joint angles (isometrically trained leg); the other leg performed conventional dynamic training, lifting and lowering (dynamically trained leg). Both legs trained at similar relative loads for the same duration. The quadriceps strength of each leg was measured isometrically (at four angles) and isokinetically (at three velocities) pre and post training. After 9 weeks of training, the increase in isokinetic strength was similar in both legs (pooled data from three velocities: dynamically trained leg, 10.7%; isometrically trained leg, 10.5%). Isometric strength increases were significantly greater for the isometrically trained leg (pooled data from four angles: dynamically trained leg, 13.1%; isometrically trained leg, 18.0%). This may have been due to the greater absolute torque involved with isometric training or a residual angle specificity effect despite the isometric training being divided over four angles.  相似文献

The aim of this study was to compare sprint performance over 10 and 20?m when participants ran while towing resistances, weighing between 0 and 30% of body mass. The sample of 33 participants consisted of male rugby and soccer players (age 21.1?±?1.8 years, body mass 83.6?±?13.1?kg, height 1.82?±?0.1?m; mean?±?s). Each participant performed two sets of seven sprints over 20?m using a Latin rectangular design. The times were recorded at 10 and 20?m using electronic speed gates. The sprints of 13 players were video-recorded to allow calculation of stride length and frequency. For both sprints, a quadratic relationship was observed between sprint time and resistance as sprint time increased from 2.94?s to 3.80?s from 0 to 30% resistance. This relationship was statistically significant but considered not to be meaningful for performance because, over the range of resistances used in this study, the quadratic model was never more than 1% (in terms of sprint time) from the linear model. As resistance increased, the stride length shortened, with mean values of 1.63?±?0.13?m at 0% body mass and 1.33?±?0.13?m at 30% of body mass. There was no significant change in stride frequency with increasing resistance. The results show that in general there is an increase in sprint time with an increase in resistance. No particular resistance in the range tested (0?–?30%) can be recommended for practice.  相似文献
