This study investigated the alleviating effects of hydrogen sulfide (H2S), derived from sodium hydrosulfide (NaHS), on inflammation induced by dextran sulfate sodium (DSS) in both in vivo and in vitro models. We found that NaHS injection markedly decreased rectal bleeding, diarrhea, and histological injury in DSS-challenged mice. NaHS (20 µmol/L) reversed DSS-induced inhibition in cell viability in Caco-2 cells and alleviated pro-inflammation cytokine expression in vivo and in vitro, indicating an anti-inflammatory function for H2S. It was also found that H2S may regulate cytokine expression by inhibiting the nuclear factor-κB (NF-κB) signaling pathway. In conclusion, our results demonstrated that H2S alleviated DSS-induced inflammation in vivo and in vitro and that the signal mechanism might be associated with the NF-κB signaling pathway. 相似文献

The aim of this study was to explore the protective effect of basic fibroblast growth factor (bFGF) on brain injury following global ischemia reperfusion and its mechanisms. Brain injury following global ischemia was induced by four vessels occlusion and systemic hypotension. Twenty-four rabbits were randomized into three groups: group A, only dissection of vessels; group B, intravenous infusion of normal saline after reperfusion for 6 h; group C, 30 μg/kg bFGF injected intravenously at the onset of reperfusion, then infused with 10 μg/(kg·h) for 6 h. Serum neuron specific enolase (NSE), S-100B, tumor necrosis factor-α(TNF-α), interleukin-1 (IL-1), interleukin-8 (IL-8) were measured before ischemia, 30 min after ischemia, 0.5, 1, 3, 6 h after reperfusion. Brain water content was determined and cerebral histopathological damages were compared. NSE and S-100B were increased 1 h after reperfusion and reached their peaks 6 h after reperfusion, but were much higher in group B than those in group C 3, 6 h after reperfusion. In groups B and C, TNF-a was increased after ischemia and IL-1 and IL-8 were increased significantly 0.5 h after reperfusion, then reached their peaks 6 h, 3 h, 6 h after reperfusion respectively. TNF-a and IL-8 at the time points of 1 h and 3 h and IL-1 at 3 h and 6 h in group C were correspondingly lower than those in group B. These indices in group A were nearly unchanged. There were less severe cerebral histopathological damages in group C compared with group B, but no difference in brain water content. It could be concluded that bFGF alleviates brain injury following global ischemia and reperfusion by down-regulating expression of inflammatory factors and inhibiting their activities. 相似文献

目的 :了解乙肝患者肝功能、乙肝五项及HBVDNA的变化及其相关性。方法 :收集门诊患者血清标本 ,同时检测肝功能、HBVELISA和HBVDNA三个不同水平。结果 :在收集的2 5 0 0例标本中 ,肝功能异常率为 6 2 .8% ,HBs -Ag阳性率为 71.2 % ,HBVDNA定量阳性率为 90 .4 %。当病毒拷贝数在 10 5~ 10 6之间时 ,肝功能异常率为 4 .2 85 % ,HBV拷贝数增加肝功能也不同程度加重 ;HBs -Ag阴性的标本中 ,FQ -PCR检出率为 30 .5 6 % ;单独HBs -Ab阳性的FQ -PCR检测的阳性率为 2 2 .35 %。结论 :HBVDNA的FQ -PCR是目前检测乙肝最为敏感的方法 相似文献

Use and effects of liver specific antigen in orthotopic liver transplantations were researched in this study. Group I: syngeneic
control (Wistar-to-Wistar); Group II: acute rejection (SD-to-Wistar); Group III: Thymic inoculation of SD rat LSA day 7 before
transplantation. The observation of common situation and survival time, rejection grades, NF-κB activity of splenocytes and
IL-2mRNA expression of grafted liver were used to analyze acute rejection severity and immune state of animals in different
groups. The common situation of group I was very well after transplantation and no signs of rejection were found. Recipients
of group II lost body weight progressively All dead within day 9 to day 13 posttransplantation; median survival time was 10.7
±0.51 days. It was an optimal acute rejection control. As for group III, 5 out of 6 recipients survived for a long time and
common situation was remarkably better than that of group II. Its rejection grades were significantly lower than that of group
II(P<0.05). NF-κB activity was only detected in group I at day 5 and day 7 after transplantation, whereas high activity of NF-κB
was detected at all time points in groupII and the low NF-κB activity detected in group III was significantly lower than that
of group II (P<0.05). No IL-2mRNA expression was detected at any time point in group I, whereas high level expression was detected at all
time points in group II and the low level expression only detected at day 3 in group III was significantly lower than that
of group II (P<0.05). Conclusion: LSA is an important transplantation antigen which is involved directly in the immunorejection of liver
transplantation. We report here for the first time that intrathymic inoculation of LSA can alleviate the rejection of liver
allotransplantation; and that grafts can survive for a long time thereby, thus leading to a novel way to achieve liver transplantation
immunotolerance.
