As a non-invasive therapeutic method without penetration-depth limitation, magnetic hyperthermia therapy (MHT) under alternating magnetic field (AMF) is a clinically promising thermal therapy. However, the poor heating conversion efficiency and lack of stimulus–response obstruct the clinical application of magnetofluid-mediated MHT. Here, we develop a ferrimagnetic polyethylene glycol-poly(2-hexoxy-2-oxo-1,3,2-dioxaphospholane) (mPEG-b-PHEP) copolymer micelle loaded with hydrophobic iron oxide nanocubes and emodin (denoted as EMM). Besides an enhanced magnetic resonance (MR) contrast ability (r2 = 271 mM−1 s−1) due to the high magnetization, the specific absorption rate (2518 W/g at 35 kA/m) and intrinsic loss power (6.5 nHm2/kg) of EMM are dozens of times higher than the clinically available iron oxide nanoagents (Feridex and Resovist), indicating the high heating conversion efficiency. Furthermore, this composite micelle with a flowable core exhibits a rapid response to magnetic hyperthermia, leading to an AMF-activated supersensitive drug release. With the high magnetic response, thermal sensitivity and magnetic targeting, this supersensitive ferrimagnetic nanocomposite realizes an above 70% tumor cell killing effect at an extremely low dosage (10 μg Fe/mL), and the tumors on mice are completely eliminated after the combined MHT–chemotherapy. 相似文献

Streptomyces produces many valuable and important biomolecules with clinical and pharmaceutical applications. The development of simple and highly efficient gene editing tools for genetic modification of Streptomyces is highly desirable. In this study, we developed a screening system for targeted gene knockout using a uracil auxotrophic host(Δpyr F) resistant to the highly toxic uracil analog of 5-fluoroorotic acid(5-FOA) converted by Pyr F, and a non-replicative vector p KC1132-pyr F carrying the complemented pyr F gene coding for orotidine-5'-phosphate decarboxylase. The pyr F gene acts as a positive selection and counterselection marker for recombinants during genetic modifications. Single-crossover homologous integration mutants were selected on minimal medium without uracil by reintroducing pyr F along with p KC1132-pyr F into the genome of the mutant Δpyr F at the targeted locus. Double-crossover recombinants were generated, from which the pyr F gene, plasmid backbone, and targeted gene were excised through homologous recombination exchange. These recombinants were rapidly screened by the counterselection agent, 5-FOA. We demonstrated the feasibility and advantage of using this pyr F-based screening system through deleting the otc R gene, which encodes the cluster-situated regulator that directly activates oxytetracycline biosynthesis in Streptomyces rimosus M4018. This system provides a new genetic tool for investigating the genetic characteristics of Streptomyces species. 相似文献

The impact of Sensory Processing Difficulties across a range of Special Educational Needs is well researched. More recently, the impact on children’s mental health and well-being, linked with anxiety, depression and self-injurious behaviours, have redirected international research to consider the holistic benefits of sensory provision, for those with special educational provision and the wider classroom population. A SMART SURVEY was designed to collate empirical evidence regarding current sensory awareness and provision in mainstream primary schools throughout Northern Ireland, in a time efficient manner. Out of 809 schools, 164 (20.27%) fully completed the online survey. Findings indicate that Special Educational Needs Coordinator confidence in developing practice, supporting staff and children was limited and impaired by insufficient and inconsistent training opportunities. While healthcare research recommends provision by trained professionals, Occupational Therapy input was limited (n = 34; 24.8%), resulting in practice that could be detrimental rather than beneficial to children’s progress. Respondents identified a need for sensory training for teachers (n = 124; 93.94%) to ensure that the impact of Sensory Processing Difficulties on the holistic development of all children is understood. Pupil voice was undervalued (n = 5; 3.6%). Active involvement of children in the process of evaluation and intervention is recommended to enhance pupil autonomy and well-being. 相似文献

The current status of play therapy in the elementary schools of Taiwan was investigated in relation to its compatibility with Chinese culture and its potential for wider societal adoption. Seven participants, four counselor educators and three elementary school counselors, in West Taiwan participated in this phenomenological qualitative inquiry. Seven assertions emerged from the interviewing data, implying that the potential for adopting play therapy in Taiwanese school counseling is embedded in the society. However, there are insufficient facilities and a lack of play therapy resources. Further investigation of the infrastructure in school counseling is necessary. Suggestions for practice and research regarding the development of play therapy and child counseling are presented. 相似文献

目的:分析研究蓝光治疗照射对于新生儿ABO血溶性黄疸的治疗效果。方法:针对在某医院2012-2013年间治疗的100例患儿的临床试验进行研究分析,根据住院时间的先后顺序将这些患儿随机地分成两个小组,对照组和观察组。每一个组别中有50个患儿,其中对照组中的患儿分别给予口服苯巴比妥、皮质激素、尼可刹米、碳酸氢钠等常规基础治疗;观察组在对照组治疗的基础上利用黄疸治疗箱行间断蓝光照射进行治疗。结果:试验发现观察组临床实验效率远远高于对照组,由于两个组别之间的差异存在一定的研究意义,实验表明对于新生儿ABO血溶性黄疸患儿,蓝光间断治疗是一种比较安全并且可靠的治疗手段,具有一定的现实意义。 相似文献

Scapular dyskinesis is the term used to describe changes in the positioning or movement of the scapula. Such dysfunction is associated with changes in the activation of the scapular muscles. However, the influence of the axial muscles on the scapular muscles activity of subjects with scapular dyskinesis is unknown. This study aimed to compare the electromyography (EMG) activity of periscapular muscles and its correlation with the external oblique muscle during the execution of push-up performed in different surfaces, in volunteers with and without scapular dyskinesis. Thirty-six men, divided in two groups (control and dyskinesis), performed push-up on stable and unstable surface. The EMG activity of serratus anterior (SA_5th and SA_7th fibers), upper (UT) and lower (LT) trapezius, external oblique (EO) was recorded during execution of each task condition. Statistical analyzes were performed using two way ANOVA repeated measures and Pearson correlation. It was observed effect of interaction between factors, being evidenced increased activity of UT, SA_7th and OE for the control group and decreased activity of SA_5th, SA_7th and EO for dyskinesis group during execution of push-up on unstable surface. In both groups positive correlations (r > 0.47) were observed between EMG activity of SA and EO. In the exercises tested, there seems to be an anatomical and functional relationship between the SA and EO muscles. The use of the unstable surface promotes increased neuromuscular demand, but the neuromuscular strategies appear to differ between groups. 相似文献

Benign prostate hyperplasia (BPH) is a common condition in aged men and result from prolong chronic inflammation in prostate
gland. Cytokines are important molecules responsible for inflammation. Single nucleotide polymorphisms (SNPs) in promoter
region of cytokine genes have been shown to alter the level of cytokines. Hence we evaluated the association of pro-inflammatory
and anti-inflammatory cytokine SNPs in a North Indian cohort of BPH patients. We observed that IL-1B −511 CT + TT genotypes conferred protective effect for susceptibility to BPH (OR 0.39, P 0.001). Our results also demonstrated that TNF-A −1031 C allele to be associated with risk for BPH (OR 1.89, P < 0.0001). Moreover, we also observed twofold risk for IL-10 −1082 cytokine gene polymorphism (OR 1.96, P 0.048). No association was observed with risk of BPH for IFN-G +874, IL-1 RN VNTR, IL-6 −174, IL-10 −819 and TGF-B +28. Our findings of IL-1B −511, TNF-A −1031 and IL-10 −1082 suggested that these variants play important role in susceptibility to BPH. Future studies in large cohort of different
ethnicity BPH groups are warranted to establish definite associations with other cytokine gene polymorphisms as well. 相似文献
