v-Src is a non-receptor protein tyrosine kinase involved in many signal transduction pathways and closely related to the activation and development of cancers. We present here the expression, purification, and bioactivity of a GST (glutathione S-transferase)-fused v-Src from a bacterial expression system. Different culture conditions were examined in an isopropyl ,B-D-thiogalactopyranoside (IPTG)-regulated expression, and the fused protein was purified using GSH (glutathione) affinity chromatography. ELISA (enzyme-linked immunosorbent assay) was employed to determine the phosphorylation kinase activity of the GST-fused v-Src. This strategy seems to be more promising than the insect cell system or other eukaryotic systems employed in earlier Src expression. 相似文献

This article is to summarize the molecular and functional analysis of the gene “suppression of tumorigenicity 13” (ST13). ST13 is in fact the gene encoding Hsp70 interacting protein (Hip), a co-factor (co-chaperone) of the 70-kDa heat shock proteins (Hsc/Hsp70). By collaborating with other positive co-factors such as Hsp40 and the Hsp70-Hsp90 organizing protein (Hop), or competing with negative co-factors such as Bcl2-associated athanogen 1 (Bag1), Hip facilitates may facilitate the chaperone function of Hsc/Hsp70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and regulators of proliferation or apoptosis. Although the nomenclature of ST13 implies a role in the suppression of tumorigenicity (ST), to date available experimental data are not sufficient to support its role in cancer development, except for the possible down-regulation of ST13 in gastric and colorectal cancers. Further investigation of this gene at the physiological level would benefit our understanding of diseases such as endocrinological disorders, cancer, and neurodegeneration commonly associated with protein misfolding. 相似文献

研究目的:探讨组织微阵列(tissuemicroarrays,TMAs或tissuechip)技术在体育生物科学领域中的应用前景。研究方法:采用历史回顾与前瞻性分析及逻辑推理相结合的方法,跟踪TMAs技术的发展及其在体育生物科学领域中的应用。研究结果与结论:TMAs技术是近年来基因芯片(DNA芯片)为代表的生物芯片技术的发展和延伸,它与细胞芯片、蛋白质芯片、抗体芯片一样,属于一种特殊的生物芯片技术。它将成百上千个不同个体或/和同一个体不同器官的组织标本排列在一张载玻片上所形成的组织微阵列生物芯片。TMAs是一种高通量、并行性、多样本研究mRNA、DNA、基因以及蛋白质定位、定性和相对定量的分析工具,在体育生物科学领域中具有广阔的应用前景。 相似文献

Protein hydrolysates are good nutritional supplements as their bioactive ingredients can be easily absorbed and utilized for
various metabolic activities. A fish protein hydrolysate (Amizate), prepared by a unique process of hydrolysis has the advantage
of high di/tri peptide content (<10 kDa) along with essential and non essential amino acids, micronutrients and vitamins.
The effect of Amizate on malnourished children (6–8 years, a total of 438) of Grade I and II (Gomez’s classification) with
respect to immunoglobulins, CD4/CD8 ratios and hemoglobin was examined. Measurement of these parameters during the user trial
study (at the beginning and the end after 4 months) indicated that the levels of the immunological parameters were not significantly
altered by the Amizate treatment. The values of immunoglobulins and CD4/CD8 ratios of malnourished children (India) are in
the normal range and are in accordance with the reported values of various ethnic groups. 相似文献
