Synaptic associativity, a feature of Hebbian plasticity wherein coactivation of two inputs onto the same neuron produces synergistic actions on postsynaptic activity, is a primary cellular correlate of associative learning. However, whether and how synaptic associativity are implemented into context-dependent relapse of extinguished memory (i.e. fear renewal) is unknown. Here, using an auditory fear conditioning paradigm in mice, we show that fear renewal is determined by the associativity between convergent inputs from the auditory cortex (ACx) and ventral hippocampus (vHPC) onto the lateral amygdala (LA) that reactivate ensembles engaged during learning. Fear renewal enhances synaptic strengths of both ACx to LA and the previously unknown vHPC to LA monosynaptic inputs. While inactivating either of the afferents abolishes fear renewal, optogenetic activation of their input associativity in the LA recapitulates fear renewal. Thus, input associativity underlies fear memory renewal.  相似文献

BackgroundPhysical activity has been hypothesized to play a protective role in neurodegenerative diseases. However, effect estimates previously derived from observational studies were prone to confounding or reverse causation.MethodsWe performed a two-sample Mendelian randomization (MR) analysis to explore the causal association of accelerometer-measured physical activity with 3 common neurodegenerative diseases: Alzheimer''s disease (AD), Parkinson''s disease (PD), and amyotrophic lateral sclerosis (ALS). We selected genetic instrumental variants reaching genome-wide significance (p < 5 × 10−8) from 2 largest meta-analyses of about 91,100 UK Biobank participants. Summary statistics for AD, PD, and ALS were retrieved from the up-to-date studies in European ancestry led by the international consortia. The random-effect, inverse-variance weighted MR was employed as the primary method, while MR pleiotropy residual sum and outlier (MR-PRESSO), weighted median, and MR-Egger were implemented as sensitivity tests. All statistical analyses were performed using the R programming language (Version 3.6.1; R Foundation for Statistical Computing, Vienna, Austria).ResultsPrimary MR analysis and replication analysis utilized 5 and 8 instrumental variables, which explained 0.2% and 0.4% variance in physical activity, respectively. In each set, one variant at 17q21 was significantly associated with PD, and MR sensitivity analyses indicated them it as an outlier and source of heterogeneity and pleiotropy. Primary results with the removal of outlier variants suggested odds ratios (ORs) of neurodegenerative diseases per unit increase in objectively measured physical activity were 1.52 for AD (95% confidence interval (95%CI): 0.88–2.63, p = 0.13) and 3.35 for PD (95%CI: 1.32–8.48, p = 0.01), while inconsistent results were shown in the replication set for AD (OR = 1.06, 95%CI: 1.01–1.12, p = 0.02) and PD (OR = 0.99, 95%CI: 0.88–0.12, p = 0.97). Similarly, the beneficial effect of physical activity on ALS (OR = 0.51, 95%CI: 0.29–0.91, p = 0.02) was not confirmed in the replication analysis (OR = 0.96, 95%CI: 0.91–1.02, p = 0.22).ConclusionGenetically predicted physical activity was not robustly associated with risk of neurodegenerative disorders. Triangulating evidence across other studies is necessary in order to elucidate whether enhancing physical activity is an effective approach in preventing the onset of AD, PD, or ALS.  相似文献

目的:通过研究运动及多巴胺II型受体(dopamine type II receptor,D2R)激动剂干预对PD模型小鼠纹状体D2MSN-D1MSN侧抑制效应的影响,揭示运动在改善PD小鼠基底神经节信息输出中的作用及机制。方法:选用4周龄雄性D2-Cre小鼠,右侧纹状体注射兴奋性光敏感蛋白病毒(rAAV-Ef1α-DIO-ChR2-EYFP-WPRE-pA),通过光遗传技术精准操控D2MSN。小鼠随机分为对照组(D2-Cre)和模型组,纹状体分别注射生理盐水和神经毒素6-羟基多巴胺(6-Hydroxydopamine hydrobromide,6-OHDA),经鉴定符合PD标准的模型组小鼠再分为PD组(PDD2-Cre)和PD运动组(PD+Ex D2-Cre)。采用4周匀速跑台运动(18 m/min,40 min/天,连续5天/周)和D2R激动剂分别对各组小鼠进行干预。实验结束后,制备离体脑片,利用全细胞膜片钳结合光遗传技术检测各组小鼠纹状体D2MSN-D1MSN侧抑制效应及黑质网状部的信息输出,并验证侧抑制效应的变化是否由D2R介导。结果:光刺激D2-MSN后,D1-MSN动作电位发放个数PDD2-Cre组显著低于D2-Cre组(P<0.05);PD+ExD2-Cre组显著高于PDD2-Cre组,但仍低于D2-Cre组(P<0.05)。光刺激D2-MSN诱发D1-MSN抑制性突触后电流(Optogenetic stimulation evoked inhibitory postsynaptic currents,oIPSC)的幅值与基线比值,PDD2-Cre组显著高于D2-Cre组(P<0.05),PD+ExD2-Cre组显著低于PDD2-Cre组,但高于D2-Cre组(P<0.05);光刺激D2-MSN诱发黑质网状部(SNr)抑制性突触后场电位(field inhibitory post synaptic potential,fIPSP)最大幅值,PDD2-Cre组显著低于D2-Cre组(P<0.05),PD+ExD2-Cre组显著高于PDD2-Cre组,但低于D2-Cre组(P<0.05)。结论:PD模型小鼠纹状体D2MSN-D1MSN侧抑制效应异常,并影响了SNr的信息输出,这可能是导致PD基底神经节功能紊乱的重要原因之一。4周运动干预通过改善PD模型小鼠纹状体D2MSN-D1MSN侧抑制效应,调节了SNr的信息输出。D2R在运动改善PD小鼠纹状体D2MSN-D1MSN侧抑制效应和调节基底神经节信息输出中起重要作用。  相似文献

It is of importance to study and predict the possible buckling of submarine pipeline under thermal stress in pipeline design.Since soil resistance is not strong enough to restrain the large deformation of pipeline,high-order buckling modes occur very easily.Analytical solutions to high-order buckling modes were obtained in this paper.The relationships between buckling temperature and the amplitude or the wavelength of buckling modes were established.Analytical solutions were obtained to predict the occurrence and consequence of in-service buckling of a heated pipeline in an oil field.The effects of temperature difference and properties of subsoil on buckling modes were investigated.The results show that buckling will occur once temperature difference exceeds safe temperature;high-order pipeline buckling occurs very easily;the larger the friction coefficients are,the safer the submarine pipeline will be.  相似文献
