多路复用大型云动态数据完整性检测方法

对大型云动态数据的完整性检测可以避免云用户存储在其中数据被篡改或删除。传统方法中,采用代数结构标签加密方法进行数据完整性检测,不能保证足够的置信度,性能不好。在云储存环境下,提出一种基于可信第三方判定和动态标签信息更新的多路复用大型云动态数据完整性检测方法。假设被处理云采集数据的是可分类的,引入了一个管理因子,得到多路复用标签信息,基于可信第三方判定和动态标签信息更新进行,进行对用户存在云存储服务器上的文件块被篡改或删掉的判定,由此实现一次数据完整性验证,提高大型云动态数据检测的准确性。仿真结果表明,该算法具有较好的多路复用大型云动态数据完整性检测性能,系统使用率提高,计算开销较少,检测准确性提高。     

Serum thyroglobulin antibody levels within or near to the reference range may interfere with thyroglobulin measurement

High concentration of thyroglobulin antibodies (TgAb) is a major limiting factor of thyroglobulin measurements in patients with differentiated thyroid cancer. We investigated whether thyroglobulin antibody added to serum samples could interfere with the thyroglobulin assay. Thyroglobulin levels in serum samples with different concentrations of thyroglobulin were measured by electrochemiluminescence immunoassay before and after the addition of increasing concentrations of thyroglobulin antibody using the secondary calibrator solution of the thyroglobulin assay kit containing sheep thyroglobulin antibody to reach thyroglobulin antibody levels within or near to the reference range. Thyroglobulin and thyroglobulin antibody concentrations were also measured in 134 serum samples from 27 patients after thyroid ablation. There was a strong negative association (slope = −1.179) between thyroglobulin antibody and thyroglobulin concentrations in samples with added thyroglobulin antibody (beta = −0.86; P < 0.001). Changes in thyroglobulin concentrations were described mathematically as loss of thyroglobulin% = −0.2408 × Ln(thyroglobulin antibody IU/ml) + 0.1944. Thyroglobulin concentrations were significantly lower than those calculated from experiments with added thyroglobulin antibody in 26/134 samples from patients after thyroid ablation. We conclude that if the same TgAb interference exists in the presence of naturally occurring human TgAb, our observation may prove to be useful during follow-up of patients with differentiated thyroid cancer. However, further studies are needed to explore the clinical relevance of thyroglobulin antibody levels within or near to the reference range in monitoring these patients.     

荧光法测定脑组织单胺类递质和有关技术问题及其在运动医学中的应用

单胺类递质在大脑许多功能的控制调节中起重要作用,测定不同状态下脑内单胺类递质可了解这类递质在不同状态下脑内的合成、释放和转化,是探索运动性中枢疲劳和有关药物作用机制的重要手段。荧光测定法因其特异性高、操作简单、费用较低等特点,具有广阔的应用前景。但实验中许多值得注意的技术问题和环节影响测定的结果,通过查阅有关文献并结合实验中的影响因素,对其测定的原理、方法和值得注意的有关技术问题及其在医学和生物学中的应用进行梳理和归纳,旨在为应用领域和实验方法学提供理论指导和技术支持。

利用RAPD分子标记评价仲彬草属的种间关系

利用随机扩增多态性DNA(RAPD)技术分析了14种仲彬草属Kengyilia植物的种间关系。对34个OPRON公司十聚体随机引物进行多态性筛选,20个(58．8％)能产生多态性。14个引物产生的112个DNA片断,用于计算种间Jaccard遗传相似性系数分析,在NTSYS程序中利用UPGMA构建系统发育树状图。分析结果表明：(1)14个Kengyilia物种存在较大的遗传多样性;(2)青藏高原的物种与新疆的物种 的RAPD变异极大;(3)形态相似、地理分布一致的物种有一定的亲缘关系,聚类在一起;(4)RAPD结果与形态学和细胞学等分析结果一致。RAPD分析方法将为Kengyilia系统分类提供DNA水平上丰富的资料。     

Biochemical characterization of the recombinant schistosome tegumental protein SmALDH_312 produced in E. coli and baculovirus expression vector system

BackgroundThe heterologous expression of parasitic proteins is challenging because the sequence composition often differs significantly from host preferences. However, the production of such proteins is important because they are potential drug targets and can be screened for interactions with new lead compounds. Here we compared two expression systems for the production of an active recombinant aldehyde dehydrogenase (SmALDH_312) from Schistosoma mansoni, which causes the neglected tropical disease schistosomiasis.ResultsWe produced SmALDH_312 successfully in the bacterium Escherichia coli and in the baculovirus expression vector system (BEVS). Both versions of the recombinant protein were found to be active in vitro, but the BEVS-derived enzyme showed 3.7-fold higher specific activity and was selected for further characterization. We investigated the influence of Mg²⁺, Ca²⁺ and Mn²⁺, and found out that the specific activity of the enzyme increased 1.5-fold in the presence of 0.5 mM Mg²⁺. Finally, we characterized the kinetic properties of the enzyme using a design-of-experiment approach, revealing optimal activity at pH 7.6 and 41°C.ConclusionsAlthough, E. coli has many advantages, such as rapid expression, high yields and low costs, this system was outperformed by BEVS for the production of a schistosome ALDH. 张家口市SARS患者、疑似病例、留观及密切接触医护人员血清中IgG、IgM抗体测定

目的 :为临床诊断及排除严重急性呼吸综合征 (SARS)提供依据和对留观及密切接触医护人员有无感染作初步调查。方法 :用酶联免疫吸附测定 (ELISA) 5 3例临床诊断病例、2 9例疑似病例、6 3例留观及 114例密切接触医护人员的血清中SARS冠状病毒IgG、IgM抗体。结果 :SARS临床诊断病例IgG抗体阳性 2 1例 ,IgM抗体阳性 18例 ,其中有 17例IgG、IgM抗体均为阳性 ,总阳性率为 4 1.5 % ;留观人员IgG抗体阳性 1例 ;其他均为阴性。 结论 :ELISA测定血清抗SARS冠状病毒抗体阳性病例可确诊感染病毒 ,对于观察期后 (一般为 2 1d)人员 ,阴性结果可以排除感染。
