目的:探讨全自动生化分析参数设置对高酶活性样本测定结果质量控制的作用.方法:设吸光度窗口(Abs window)参数,以及既设Linrearity%、Abs window又设Flex(弹性速率)参数四种不同状态下测定4个不同浓度样本的AST活性.结果:未设参数、仅设线性率(Linearity%)参数、仅设吸光度窗口(Abs window)参数三种状态下测定4个不同浓度样本的AST活性与真实值差异明显;三种参数全设状态测定各浓度样本的AST活性,除极高浓度样本无结果有报警外,其它标本与真实值无明显差异.结论:全自动生化分析参数设置对高酶活性样本测定结果的质量控制具有重要作用.  相似文献

研究蛇足石杉生物碱的提取工艺.整个工艺是:用酸浸提、微波加热、蒸馏、萃取、反萃取、脱色等分离技术从蛇足石杉中提取石杉碱,最后定性定量检测.结果表明:(1)微波加热处理可提高石杉碱的提取率;(2)萃取和反萃取相结合,使用旋转蒸发仪蒸馏有助于分离石杉碱;(3)用碘化铋钾试液对提取的生物碱进行饱和滴定,然后烘干称量,再根据化学反应式来计算生物碱含量,是一种较好的近似测定方法;(4)根据标准样品的直线回归方程,利用紫外分析仪测定石杉碱甲含量是一种经济高效的方法;(5)洪江市塘湾镇的蛇足石杉的石杉碱甲的含量为0.085‰,粗提物中石杉碱甲的百分含量为26.1%.  相似文献

目的:研究血浆脂蛋白(a)[Lp(a)]水平与肾病综合征(NS)的关系,探讨Lp(a)在肾病综合征合并心脑血管疾病中的作用。方法:用酶联免疫吸附法分别测定了61例肾病综合征患者与55名健康对照者的Lp(a)浓度,并同时测定了BUN、Scr、TG、TC、LDL-C和HDL-C的含量。结果:肾病综合征患者Lp(a)、BUN、Scr、TC、LDL-C水平均明显高于正常对照组。而HDL-C显著低于对照组,NS组Lp(a)浓度与BUN和Scr呈显著正相关,Lp(a)水平变化与LDL-C、HDL-C、TG、TC含量无明显相关性。结论:肾病综合征患者体内存在着Lp(a)等脂代谢紊乱的情况,高血浆Lp(a)水平是肾病综合征合并心脑血管疾病的独立危险因素之一。  相似文献

目的:建立矮杨梅叶总黄酮的提取工艺和含量测定方法。方法:利用L9(34)进行矮杨梅叶总黄酮提取影响因素即乙醇浓度、提取时间、料液比的优化组合;应用分光光度计考察其含量测定方法。结果:矮杨梅叶总黄酮的提取工艺为提取时间为1.5h,乙醇浓度为50%,材液比为1:35,提取率为4.88%;总黄酮提取液在0-60min内测定,线性关系良好(0.9996),仪器较为稳定,测定结果准确、重现性好,回收率高。结论:矮杨梅叶总黄酮的提取工艺可行、测定方法可信。这一研究结果对于杨梅属植物中总黄酮的提取和测定具有可操作性和推广应用价值。  相似文献

以镇海林蛙(Rana zhenhaiensis)蝌蚪为研究对象,用碱性单细胞凝胶电泳方法(慧星试验,SCGE)检测不同质量浓度的甲氨基阿维菌素苯甲酸盐(甲维盐)对其的遗传毒性.结果表明,在实验室条件下,随着农药质量浓度的增加,蝌蚪红细胞的损伤率(拖尾率)及DNA损伤程度随之增加;在浓度为3×10-2 mg/L的甲维盐溶液中处理24 h后,与对照组相比,镇海林蛙蝌蚪红细胞的损伤率及DNA损伤程度有显著的提高;甲维盐的剂量与蝌蚪红细胞损伤率及DNA损伤程度呈显著的线性相关关系.研究表明,甲维盐对两栖动物蝌蚪具有一定的遗传毒性作用,在检测环境污染物对两栖类的遗传毒性方面,慧星试验是一种合适的方法.  相似文献

Cardiovascular autonomic neuropathy (CAN) is one of the most clinically significant complications of diabetes mellitus. Even though many ethological factors have been attributed for the pathogenesis of this disease no attempts were made to correlate DNA damage as a causative factor. Hence the present study was undertaken to asses the extent of somatic DNA damages by cytokinesis-block micronuclei assay (CBMN). An attempt is also being made to correlate the habits and/or risk factors and socioeconomic status with CAN. The CBMN frequency of 46 patients suffering from autonomic neuropathy was compared with that of 25 healthy age and sex matched controls. All the subjects were suffering from type 2 diabetes for at least 8 years and have varying degrees of coronary artery diseases. The mean CBMN frequency of the patients was statistically higher than that of the healthy control subjects (P < 0.05). The CBMN frequency was found to be significantly altered in CAN patients who where physical inactivity and smoking. A significant correlation could also be observed between CAN and smoking, diabetes mellitus, hypertension, dyslipidemia, abdominal obesity, and physical activity.  相似文献

随着网络多种业务的发展,人们对服务质量(QOS)提出了很高的要求,IP over ATM技术将成为未来网络的主流,但这加大了路由选择的复杂性.传统的蚂蚁算法虽然能较快地找到一条最优路径,但是容易造成该路径上的拥塞,而且无法解决这一状况.可对多路径改进的蚂蚁算法进行改进,当最优路径处于拥塞状态时,该算法将绕过处于拥塞的节点重新搜索新的"次优"路径对原最优路径分流,这样既保证了服务质量,又达到优化路由选择的目的.  相似文献

The objective of the present study was to investigate the antioxidant status and the extent of oxidative DNA damage in lymphocytes and their relation with essential hypertension (EHT). A total of 100 South Indian subjects aged 30–65 were included for the study. Of these 50 were normotensive controls (group-1) with blood pressure ≥120/80 mm Hg, 50 were newly diagnosed (group-2) and were not on any antihypertensive drugs, but had systolic blood pressure ranging between 140 and 160 mmHg and diastolic blood pressure 95–100 mmHg and 50 newly diagnosed essential hypertensive patients underwent drug therapy for 1 year was considered as group-3. Enzymatic and non-enzymatic antioxidants significantly decreased and lymphocyte DNA damage was significantly increased in newly diagnosed hypertensive patients compared with control group. The major decrease in DNA damage and significant improvement in enzymatic and non-enzymatic antioxidants were observed after 1 year of antihypertensive therapy in treated group compared with newly diagnosed hypertensive patients. Total antioxidant status and lymphocyte DNA damage showed a strong negative correlation in all the three groups. Essential hypertension associated with oxidative stress which in turn causes genotoxic susceptibility to variety of disease including cancer. In the absence of DNA repair process and DNA checkpoint mechanisms, the genomic integrity is susceptible to extensive damage. In our study, increased oxidative DNA damage and decreased antioxidant levels were frequently observed in the newly diagnosed essential hypertensive patients, suggesting that oxidative stress is important in the pathogenesis of EHT. Therefore, the present study has additional clinical implication. Further investigations with large number of patients along with antioxidant supplement are highly warranted.  相似文献

Development and evaluation of immunoassay for zeranol in bovine urine   总被引:1,自引:0,他引:1  
A high affinity polyclonal antibody-based enzyme linked immunosorbent assay (ELISA) was developed for the quantification of zeranol in bovine urine. On the basis of urine matrix studies, the optimized dilution factors producing insignificant matrix interference were selected as 1:5 in pretreatment. In the improved ELISA, the linear response range was between 0.02 and 1 μg/ml, and the detection limit was 0.02 μg/ml for the assay. The overall recoveries and the coefficients of variation (CVs) were in the range of 82%-127% and 3.5%-8.8%, respectively. Thirty-six bovine urine samples spiked with zeranol (ranging from 0.2 to 10 μg/ml) were detected by the ELISA and liquid chromatography (LC) method, and good correlations were obtained between the two methods (R^2=0.9643). We conclude that this improved ELISA is suitable tool for a mass zeranol screening and can be an altemative for the conventional LC method for zeranol in bovine urine.  相似文献
