藻细胞计数及死／活分析的流式细胞仪方法

以我国原水典型藻类铜绿微囊藻和椭圆小球藻为例,建立了采用双组分绝对计数微球进行藻细胞计数的流式细胞仪分析方法,以及采用染料SYBR green I和碘化丙啶（PI）双染色判定细胞死／活的流式细胞仪分析方法,运用所建立方法开展了0-200 mJ·cm^-2紫外消毒处理后铜绿微囊藻的细胞计数及死／活分析.研究结果表明,流式细胞方法计数结果与显微计数结果具有较好的线性相关性（R0=0．980）,采用流式细胞仪使单样品操作时间缩短至5min以内．流式细胞方法能灵敏检测细胞死／活状态,测定梯度配比的热处理致死细胞的结果与配比值良好吻合,铜绿微囊藻拟合曲线尺。值达0．999,椭圆小球藻拟合曲线彤值0．995．流式细胞方法用于紫外消毒处理水样中藻细胞计数及死／活分析灵敏有效．     

吸烟对体育专业学生外周血淋巴细胞DNA损伤的研究

目的：用慧星试验检测体育专业学生吸烟所诱发的DNA损伤。方法：选体育专业15名吸烟学生（平均烟龄10．3支／天，1．4年）和8名不吸烟的对照组学生。用彗星试验检测了两组对象外周血淋巴细胞DNA的迁移。结果：实验结果表明吸烟组的平均彗星尾长为2．75μm，明显高于对照组（1．2μm），差异有显著性（P＜0．01）。平均彗星尾相为3．66，明显高于对照组（2．14），差异有显著性（P＜0．01）。吸烟组平均长尾核的百分率为27．74％，明显高于对照组的5．55％，差异有显著性（P＜0．01）。结论：体育专业吸烟学生外周血淋巴细胞的DNA有不同程度的损伤。     

微量全血单细胞凝胶电泳技术在身体机能评定中的应用

单细胞凝胶电泳技术是一种在单细胞水平上检测有核细胞DNA损伤与修复的方法.本文总结了该实验技术的基本原理、方法及应用现状.改进的微量全血单细胞凝胶电泳技术具有敏感性高、取材方便、费用低、实验周期短等优点,对于运动中身体机能的评定具有重要的意义。     

快速优化培养基检测技术在实验教学中的应用

以紫外分光光度法作为庆大霉素含量测定手段,采用正交试验方法,对紫色小单孢菌产庆大霉素的发酵培养基及发酵条件进行优化,通过统计学分析,确定发酵培养基最优组合为:黄豆饼粉3.5%,CaCO30.5%,淀粉5.5%,CoCl20.0006%。优化的发酵条件为:种龄60 h,接种量12 mL,装液量50 mL/500 mL摇瓶。在此优化条件下添加一定量的氨基酸培养6 d,庆大霉素效价较优化前提高了85.5%。用紫外分光光度法与微生物法测定庆大霉素发酵液效价产生的误差小于4%,该方法可以用于教学实验中庆大霉素液体发酵培养基优化的快速筛选。     

Molecular Studies on Coronary Artery Disease—A Review

Coronary artery disease (CAD) remains the major cause of mortality and morbidity in the entire world population. The conventional risk factors of CAD include hypertension, hyperlipidemia, diabetes mellitus, family history, smoking etc. These factors contribute only 50 % of the total risk of CAD. For providing a complete risk assessment in CAD, it is mandatory to have well-planned clinical, biochemical and genetic studies in patients with CAD and subjects who are at risk of developing CAD. In this review an attempt is made to critically evaluate the conventional and emerging risk factors which predispose the individual to CAD. Specifically, the molecular basis of CAD including high oxidative stress, low antioxidant status and increased DNA damage are covered. A comprehensive and multifactorial approach to the problem is the better way to reduce the morbidity and mortality of the disease.     

龙竹和凤尾竹PpMADS1序列的扩增与序列分析

以龙竹和凤尾竹基因组DNA为模板,通过试验不同的退火温度、循环数、Mg2＋浓度及DMSO浓度,获得了16条龙竹和凤尾竹PpMADS1相似序列片段,并得出稳定扩增PpMADS1相似序列的条件.对获得的龙竹和凤尾竹PpMADS1相似序列进行序列分析,发现龙竹和凤尾竹PpMADS1相似序列可根据序列长度差异分成组I和组II,并对其推出的氨基酸序列进行分析,找到了保守的paleoAP1基序和KIII区保守的氨基酸残基,并据此认为龙竹和凤尾竹PpMADS1相似序列是AP1/SQUA亚家族成员.     

变压器多路温度监控系统设计

介绍了一种基于平衡电桥和电子开关矩阵实现的多路温度硬件测量系统以及软件实现监控的系统设计方案,详细介绍了应用电子开关矩阵MT8816组成的多点温度测量技术、平衡电桥实现的温度测量方式以及单片机程序和上位机软件程序的设计,并对该设计方案中的误差消除办法进行了分析。对于检测点较多的系统,采用本方案可以以最低的成本实现高精度的检测。     

Administering the Optimum Dose of l-Arginine in Regional Tumor Therapy

The purpose of this study is optimizing the l-arginine (l-Arg) doses on the basis of chemical structure in regional accessible tumor therapy to settle down a new protocol for the treatment of cancer. ³H-thymidine-based cell proliferation assay was performed in vitro on tumor cell lines of fibrosarcoma (FS), lymphosarcoma-ascitic and on normal cell line of NIH 3T3 after treatment with different concentrations of l-Arg in phosphate buffered saline (PBS). The cultures were harvested after 22 h and the incorporated radioactivity was counted to identify their histologic grades as described in earlier studies. In vivo therapy of murine tumors was conducted where FS cells injected subcutaneously at ventro-lateral position of mice. Various drug delivery schedules were injected into the centre of tumor base, once a day for 4 days. Tumor diameter and survivals were monitored where the day of sacrifice was considered for monitoring the survival period. By identifying the histologic grades of the treated cultures in vitro and in vivo by different concentrations of l-Arg, the corresponding energy of such concentrations were determined. An efficient model with a good fit (R² = 0.98) was established to describe the energy yield by l-Arg dose. The equivalence between the tumor histologic grade and energy of the l-Arg dose delivered in saline (PBS) environment is the optimum condition for regional tumor therapy achieves higher survival rate. The selective cytotoxicity to tumor cells with minimal damage to normal cells by l-Arg due to its chemical structure suggests to be considered the most promising drug for regional therapy of the accessible tumors like breast cancers of early stage with no distant metastasis.     

Hb Hope: A Rare Variant of Haemoglobin-Forged a Rise in HbA1c on Immunoturbidimetric Assay

Glycosylated hemoglobin (HbA1c) determination is a powerful means for assessing the evaluation and management of patients with diabetes mellitus. Hemoglobin (Hb) variants and chemically modified derivatives of Hb can affect the accuracy of measurement of HbA1c done by various analytical methods. We report a patient with a rare variant of Hb (Hb Hope) that caused an abnormally high value of HBA1c when assayed using immunoturbidimetric assay (“Tina-quant” 2nd generation assay) and also elucidate the nature of the variant.     

竹红菌甲素对人黑色素瘤B-16细胞的杀伤效应

从生物学角度上对竹红菌甲素(HA)光敏活性进行了研究，运用MTT法实验发现HA合并光照处理人黑色素瘤B-t6细胞，HA对B-16细胞具有明显的杀伤作用，使癌细胞存活率显著下降，结果表明HA具有光敏抗癌活性。
