乳腺癌耐受蛋白介导5-氟脲嘧啶的耐受及机制探讨

AIM To filtrate breast cancer resistance protein(BCRP)-mediated resistance agents and investigate the mechanism,so as to provide valuable datum for optimization clinical chemotherapy scheme to tumor with evaluation marker of BCRP expression. METHODS MTT assay was used to filtrate BCRP-mediated resistance agents with PA317/Tet-on/TRE-BCRP cell of different expression levels of BCRP after treated with different concentration anticancer agents. High performance liquid chromatography(HPLC) was applied to measure relative dose of intracellular retention resistance agents. Nuclear DNA fluorescence dye,Hochest 33258, staining and flow cytometry were adopted to detect apoptotic cells after treated with drugs. RESULTS There were shown increasing durg-resistance to 5-fluorouracil,methotrexate, doxirubicin, pirarubicin,etoposide and mitoxantrone followed with increasing expression of BCRP on PA317/Tet-on/TRE-BCRP cells(P＜0.05, n=3),but shown sensitive to paclitaxel, cisplatin, vincristine, mitomycin and vindesine. There also was shown significant negative correlation between the intracellular retention dose of 5-fluorouracil with different expression of BCRP(r=-0.885, P＜0.05, n=3).There were shown parallel results of that decreasing cellular apoptotic rate with increasing cellular expression of BCRP after treated with 5-fluorouracil by fluorescence dye staining and flow cytometry(P＜0.05, n=3),and also shown significate rise of the apoptotic rate of BCRP expression cells after treated with Ko143 (P＜0.05, n=3). Every group of cells could be different extently blocked in phase of G0/G1 treated with 5-fluorouracil. CONCLUSION Resistance of 5-fluorouracil could be especially mediated by conjugated with BCRP and acted as drug exclude-pump substrate. Cellular ability resistant to 5-fluorouracil-induced apoptosis could be reinforced by BCRP expression.

Discursive Dancing: Traditionalism and Social Realism in the 2013 English History Curriculum Wars

This paper is an exploration of the debates surrounding the publication of a new National Curriculum for history in England. The draft curriculum was published in February 2013 and was withdrawn just 6 months later in the face of considerable opposition. This paper offers a tentative explanation for this example of a rare phenomenon: effective resistance to curriculum change. Using a socio-cognitive approach to discourse analysis, the paper explores the context models of the two antagonists in the contestation: new right traditionalism and social realism. While both context models are viewed as coherent, it is suggested that critics of the draft prevailed because they more fully comprehended the context model of their opponents and were prepared to adapt their strategy accordingly. The paper takes an analytical narrative approach to the contestation. Resistance to the draft is presented in two phases: an initial phase in which criticism was diffuse, instinctive and political; and a more effective mature phase in which opposition united around a depoliticised disciplinary defence of the subject in social realist terms. It is argued that this deft shift went unnoticed by Education Secretary, Michael Gove, rendering ineffective his attacks on his critics as ‘Marxists’ and ‘progressives’.     

外源海藻糖提高黄瓜抗盐性的初步研究

文章初步研究适宜浓度的海藻糖可以提高黄瓜的抗盐性.以中农16号黄瓜幼苗为供试材料,首先分别用0mM、5mM、10mM、和20mM的海藻糖随1/2Hoagland营养液根灌两次,随后在上述各营养液中加入180mM的NaCI对幼苗进行渗透胁迫3天,自第4天开始测试下列指标:幼苗叶片的相对含水量、离体叶片电导率、MDA含量、叶绿素含量、POD、CAT酶的活性.试验结果如下:5mM、10mM、20mM的海藻糖处理都可以提高高盐渗透胁迫下幼苗叶片的相对含水量、叶绿素含量、POD、CAT酶的活性;降低叶片电导率和MDA含量.而且,10mM的海藻糖处理效果最好,而20mM的海藻糖处理效果不如5mM的海藻糖的处理效果.研究表明:适宜浓度的海藻糖可提高在渗透胁迫下黄瓜幼苗的抗性,在5mM、10mM、和20mM三个浓度中以10mM的海藻糖的处理效果最好.     

藜蒿提取物抗氧化性能的研究

研究藜蒿提取物对食用油脂的抗氧化性能。结果表明，藜蒿用90℃热水提取、乙酯乙酯萃取而得到的抗氧化物质，产率高，抗氧化效果好。另外，对猪肉的保鲜也有一定的作用。
