Chimeric antigen receptor T cell (CAR T) therapy was a milestone in the treatment of relapsed and refractory B cell malignancies. However, beneficial effects of CAR T cells have not been obtained in solid tumors yet. Herein, we implement a porous microneedle patch that accommodates CAR T cells and allows in situ penetration-mediated seeding of CAR T cells when implanted in the tumor bed or in the post-surgical resection cavity. CAR T cells loaded in the pores of the microneedle tips were readily escorted to the tumor in an evenly scattered manner without losing their activity. Such microneedle-mediated local delivery enhanced infiltration and immunostimulation of CAR T cells as compared to direct intratumoral injection. This tailorable patch offers a transformative platform for scattered seeding of living cells for treating a variety of tumors.  相似文献

The magnetic anisotropy of Fe25Ni75 nanocrystallites in the range of 10∼20 nm was measured by the law of approach to saturation. The samples were prepared by mechanical alloying process, and the average crystal size was determined by X-ray diffraction. The effective magnetic anisotropy of these fine particles is found In an order of 106 erg/cm3 that is much greater than that of normal crystal size of particles. The dependence of magnetic anisotropy on the particle size was studied. It has been demonstrated that the strain anisotropy occupies the most of the total magnetic anisotropy, and the internal strain is a critical factor for their magnetic properties. The magnetic properties of as-quenched and annealed Fe73.5Ag1Nb3Si13.5B9 alloys were investigated. X-ray diffraction was used to monitor the structure changes of the samples under different annealing temperatures. It was found that by Cu addition, the crystallization temperature T x1 of Fe-Nb-Si-B alloy is decreased, whereas by Ag addition, the T x1 is increased. A small amount of α-Fe and Ag were obtained after the sample was annealed at 500°C for 1 h. With increasing the annealing temperature, Fe23B6, Fe2B, Fe3B were separated out and magnetic properties of the samples were impaired quickly. A mostly single nanocrystalline grain of a-Fe phase could not be obtained in the annealed samples.  相似文献

应用随机分组将600例手术病人两组,每组3000人。一组从手术开始即吸入纯氧,流量4-6升,至麻醉结束为止;另一组术中不给氧。两组术前术后抗生素应用、术中的麻醉方式及用药均基本相同。对两组病人性别、年龄、疾病构成、手术时间、术后肠道功能恢复、伤口感染、白细胞计数、住院时间和费用等多项变量进行统计学处理,两组的感染数、住院时间以及感染病人和非感染病人的白细胞计数、平均住院时间均有显性差异。术中给氧是预防外科伤口感染的使捷方法。  相似文献

Lymphatic filariasis continues to be the major cause of clinical morbidity in India and other developing tropical countries. One of the major lacunae in the effective management of clinical filarial cases is the non-availability of a suitable diagnostic test for confirming filaria aetiology in acute, chronic and occult clinical cases where microfilariae (mf) are not usually seen in peripheral circulation. Studies in our laboratory have shown the usefulness of filarial antibody and antigen assays using microfilarial excretory-secretory (mf ES) antigen in detecting microfilaraemic, acute and chronic filarial cases and in confirming filarial aetiology in occult infections. Diethylcarbamazine citrate (DEC) is the drug of choice for lymphatic filariasis. Different regimens of DEC have been explored in the treatment of microfilaraemic cases. Immunomonitoring has shown that the seroconversion of antigen and antibody positivity was found to be very helpful in determining appropriate period of DEC treatment for clinical relief and cure in clinical filarial patients and further they did not have recurrence in most of the cases. Optimal DEC (6mg/kg body wt/day for 21 days each month for 3–12 months) therapy was found to be very effective in acute and atypical clinical manifestations such as asthmatic bronchitis, pulmonary eosinophilia, monoarthritis, recurrent upper respiratory tract infections (URI), pneumonia (super imposed infections) in children and minimal hydrocele, epididymoorchitis, lymphangitis, lymphadenitis, acute abdomen, central serous retinopathy, tenosynovitis, pain and swelling in limbs and joints in adults living in filaria endemic areas.  相似文献
