To develop a gene therapy strategy for treating bovine mastitis, a new mammary-specific vector containing human lysozyme (hLYZ) cDNA and kanamycin resistance gene was constructed for intramammary expression and clinical studies. After one time acupuncture or intracisternal infusion of healthy cows with 400μg of the p215C3LYZ vector, over 2.0 μg/ml ofrhLYZ could be detected by enzymatic assay for about 3 weeks in the milk samples. Western blotting showed that rhLYZ secreted into milk samples from the vector-injected cows had molecular weight similar to that of the natural hLYZ in human colostrums. Twenty days after the primary injection, the quarters were re-injected with the same vector by quarter acupuncture and even higher concentrations of rhL YZ could be detected. Indirect competitive ELISA of milk samples showed that the vector injection did not induce detectable humoral immune response against hL YZ. Clinical studies showed that twice acupuncture of quarters with the p215C3LYZ vector had overt therapeutic effect on clinical and subclinical mastitis previously treated with antibiotics, including disappearance of clinical symptoms and relatively high microbiological cure rates. These data provide a solid rationale for using the vector to develop gene therapy for treating bovine mastitis. 相似文献

Single parenthood as a common psychosocial phenomenon seems to be regarded as one of the most significant issues in the psychological domain and needs to be taken into serious consideration due to emotional, psychological, and social problems created by it. With regard to the rapidly growing population of single parents and their children experiencing relationship-based problems, interventions which deal with relational issues are worth in-depth explorations. This study was primarily concerned with the betterment of single parent-child relationships. For the purpose of this research, a case study was conducted to probe into the impact of filial therapy-a relationship-enhancement family/play therapy--on (1) reducing children' problem behaviors, and (2) ameliorating parent-child dysfunctional relationships through improving parenting knowledge and skills. The intended data for the study was gathered through (1) pre-post play observation, (2) parent's self-reports, and (3) the reports obtained from a kindergarten trainer. The outcomes of this one-month long case study revealed that filial therapy has the potentiality in enhancing and rebuilding parent-child relationships through teaching single parents the skills necessary for coping with the problems accelerated by the newly developed situation, which might be seen as a result of single parenthood (e.g., childhood problem behaviors, parent-child relational problems, lack of adequate parenting knowledge and skills). In the end, this article concludes a few suggestions for future studies in this field. 相似文献

At Chiba University, gross anatomy laboratory sessions (“laboratories”) are required for physical therapy students. Though most physical therapy schools require their students to participate in laboratories so that they will better understand the structure of the human body, few data exist on the value of these laboratories specifically for physical therapy students. We administered questionnaires to physical therapy undergraduate students both before and after they participated in laboratories. Questionnaire items focused on student attitudes toward the laboratories and on human life and dignity. Data from 83 students were analyzed, with the following results: (1) 74.7% of students had a positive attitude about attending laboratories before doing so; (2) with few exceptions, students' attitudes about upcoming laboratories grew more positive after experiencing the laboratory work (P < 0.001); (3) laboratories caused students to contemplate the topics of human life and dignity; and (4) 83.1% of students hoped to participate in laboratories at least four times. These results indicate that laboratories reinforce physical therapy students' positive attitudes about laboratory learning and promote student reflection on human life and dignity. This study provides support for the implementation of multiple laboratory sessions using cadavers into a uniform curriculum for physical therapy students in Japan. 目的:对比不同保守疗法治疗运动员膝关节内侧副韧带(MCL)损伤的临床疗效。方法:2018年6月至2022年12月期间膝关节MCL损伤的运动员共72例,随机分为电针+推拿组(对照组)、电针+推拿+康复训练组(实验组),每组36人,进行为期4周的治疗。采用疼痛视觉模拟评分(VAS).Lysholm膝关节功能评分,对不同组的运动员治疗前后的疼痛、膝关节功能进行评定,并进行临床疗效评估。结果:对照组和实验组治疗后的有效率分别为76%和94%,VAS评分均显著降低(P<0.45),Lysholm评分均显著增加(P<0.05);治疗后,实验组的VAS评分.Lysholm评分均优于对照组,且差异显著(P<0.05)。结论:电针结合推拿和康复训练治疗运动员膝关节MCL损伤的疗效要优于电针结合推拿治疗方法,能够明显改善患者膝关节周围疼痛及膝关节整体功能。 相似文献
