A pair of two-component regulatory genes ecrA1/A2 in S.coelicolor

Two-component genes are kinds of genetic elements involved in regulation of antibiotic production in Streptomyces coelicolor. DNA microarray analysis revealed that ecrA1/A2, which mapped at distant sites from red locus and encode respectively the kinase and regulator, expressed coordinately with genes of Red specific biosynthetic pathway, ecrA1 and ecrA2 gene-disruptive mutants were constructed using homogenotisation by reciprocal double crossover. Fermentation data showed that the undecylprodigiosin (Red) level of production was lower than that of wild-type strain. However, the change of the actinorhodin (Act) production level was not significant compared with wild type. Thus, these experiment results confirmed that the two-component system ecrA 1/A2 was positive regulatory element for red gene cluster.

Genetic polymorphism analyses of a novel panel of 19 X-STR loci in the Chinese Uygur ethnic minority

The population genetic data and forensic parameters of 19 X-chromosome short tandem repeat (X-STR) loci in Chinese Uygur ethnic minority are presented. These loci were detected in a sample of 233 (94 males and 139 females) unrelated healthy individuals. We observed 238 alleles at the 19 X-STR loci, with the corresponding gene frequencies spanning the range from 0.0021 to 0.5644. After Bonferroni correction (P>0.0026), there were no significant deviations from Hardy-Weinberg equilibrium. The cumulative power of discrimination in females and males, and the probability of exclusion of the 19 X-STR loci were 0.999 999 999 999 999 999 998 091, 0.999 999 999 999 966, and 0.999 999 986 35, respectively. The cumulative mean exclusion chance was 0.999 999 992 849 in deficiency cases, 0.999 999 999 999 628 in normal trios, and 0.999 999 998 722 in duo cases. The high value of the forensic parameters mentioned above revealed that the novel panel of 19 loci had important values for forensic applications in the Uygur group.     

除草剂"Basta"对油菜植株和愈伤组织的生长分化的影响

通过研究除草剂“Basta”对3个油菜品种的植株和愈伤组织的生长及分化的影响,发现“Basta”对油菜植株和愈伤组织的生长及分化具有强烈的抑制作用,而且品种不同,抑制程度不同。并在此基础上找到合适的“Basta”筛选压,为以bra基因作为选择标记基因的基因转化试验莫定了基础。     

萤火虫萤光素酶基因转化花叶芋及表达的研究

将萤火虫萤光素酶基因重组到质粒pBR325中,通过“三亲交配”和同源重组,将萤光素酶基因整合到Ti质粒pGV3850:1103。通过改良的叶盘转化法,用萤光素酶基因转化单子叶植物花叶芋,获得转基因植株。对再生植株的胭脂碱测定、新霉素磷酸转移酶分析、DNA点杂交、萤光素酶基因活性测定,证明萤光素酶基因已转化到花叶芋中并进行了表达。

人类首例基因编辑婴儿试验:伦理失范和善治的迫切性（英文）

近年来,中国生物医学研究快速发展的同时,也在伦理治理方面带来了挑战。贺建奎公开声称其胚胎基因编辑婴儿诞生后,立即招致广泛的批评、谴责和激烈的伦理和法律大辩论。以生育为目的的胚胎基因编辑操作违反了公认的伦理规范,具体包括:较低的科学价值、不合理的风险-受益比、伦理审查不合规、并非真正的知情同意等。这说明主要依靠科研人员自律的机构伦理治理体系是成问题的,为此需要在不同层面上改进伦理治理水平,强化技术和伦理指南和法规。     

人类STR标记的研究进展

STR是由1～6个核苷酸的串联重复片段构成的,均匀分布于人类基因组中的简单重复序列,由于重复单位的重复次数在个体间呈高度变异性并且数量丰富、分布广且均匀、多态信息含量高、检测快速方便等特点,因此目前被广泛应用于人类基因定位、连锁分析、血缘关系鉴定、群体遗传学、系统发生树构建、遗传做图等方面.
