Study on the expression and mutation of human telomeric repeat binding factor ( hTRF1 ) in 10 malignant hematopoietic cell lines

Objective: Detecting the expression and mutation of human telomeric repeat binding factor (hTRF1) in 10 malignant hematopoietic cell line cells on the base of determining its genomic structure and its four pseudogenes to clarify if hTRF1 mutation is one of the factors of the activation of telomerase. Methods: hTRF1cDNA sequences were obtained from GenBank, its genome structure and pseudogenes were forecasted by BLAST and other biology information programs and then testified by sequencing. Real-time RT-PCR was used to detect the expression of hTRF1mRNA in 10 cell line cells, including myelogenous leukemia cell lines K562, HL-60, U-937, NB4, THP-1, HEL and Dami; lymphoblastic leukemia cell lines 6T-CEM, Jurkat and Raji. Telomerase activities of cells were detected by using telomeric repeat amplification (TRAP)-ELISA protocol. PCR and sequencing were used to detect mutation of each exon of hTRF1 in 10 cell line cells. Results: hTRF1 gene, mapped to 8q13, was divided into 10 exons and spans 38.6 kb. Four processed pseudogenes of hTRF1 located on chromosome 13, 18, 21 and X respectively, was named as ψhTRF1-13, ψhTRF1-18, ψhTRF1-21 and ψhTRF1-X respectively. All cell line cells showed positive telomerase activity. The expression of hTRF1 was significantly lower in malignant hematopoietic cell lines cells (0.0338, 0.0108:_0.0749) than in normal mononuclear cells (0.0493, 0.0369:_0.128) (P=0.004). But no significant mutation was found in all exons of hTRF1 in 10 cell line cells. Four variants were found in part of intron 1, 2 and 8 of hTRF1. Their infection on gene function is unknown and needs further studies. Conclusion: hTRF1 mutation is probably not one of the main factors for telomerase activation in malignant hematopoietic disease. Transient expression of chicken alpha interferon gene in lettuce

We investigated the possibility of producing chicken alpha interferon (ChIFN-α) in transgenic plants.The cDNA encoding ChIFN-a was introduced into lettuce (Lactuca sativa L.) plants by using an agro-infiltration transient expression system.The ChIFN-α gene was correctly transcribed and translated in the lettuce plants according to RT-PCR and ELISA assays.Re-combinant protein exhibited antiviral activity in vitro by inhibition of vesicular stomatitis virus (VSV) replication on chicken embryonic fibroblast (CEF).The results demonstrate that biologically active avian cytokine with potential pharmaceutical ap-plications could be expressed in transgenic lettuce plants and that it is possible to generate interferon protein in forage plants for preventing infectious diseases of poultry.     

拟南芥热激因子HsfA1a部分氨基酸的表达与纯化

以构建的表达拟南芥热激因子HsfA1a C端氨基酸331到氨基酸486（简称△HsfA1a）的大肠杆菌Ecoli M15为材料,用异丙基硫代-β-D-半乳糖苷（IVFG）诱导表达△HsfA1a,再通过Ni—NTA—Agarose亲和层析纯化表达的△HsfA1a,通过SDS—PAGE电泳分析表达蛋白和纯化蛋白,获得了表达纯化的△HsfA1a．     

人格权与表达自由:孰轻孰重?

大众传播侵权表现出民事权利同宪法权利之间尖锐的冲突,其关键在于有关表达自由的宪法基本权利规范能否以及如何适用于大众传播侵害人格权的民事纠纷。表达自由基本权利只有内化为民法价值才可以发挥其作用,亦即宪法关于表达自由的规范对大众传播侵害人格权所产生的民事法律关系不可直接适用。对于上述民事关系,间接适用宪法的最佳途径是公共利益条款,即把有关表达自由的宪法规定转化成民法语言,以实现在民事案件处理中兼顾宪法精神。     

论宋贾昌朝《群经音辨》的语言本体研究价值

北宋贾昌朝所撰《群经音辨》是有宋一代一部重要的小学文献,也是我国古代继《经典释文》之后又一部重要的音义著作。自宋迄清的很长一段时间以来,一直备受学者们的肯定和重视。前一至六卷汇录了1100多个异读字头的2600多条音义资料,蕴含着丰富的直音、反切、词义、音变构词等语言研究资料,是研究先秦与汉魏六朝时期文献语言的语音、词义和音变构词法等的资料宝库,因而有着十分重要的语言本体研究价值。     

模糊控制分区方法研究

针对基本模糊控制解析式控制粗糙的问题,介绍了一种比较适用的模糊控制表(F.C表)分区方法,并在模糊控制解析式中引入动态调整因子,仿真取得了满意的控制效果。     

Negative Jealousy-Related Emotion Rumination as Consequences of Romantic Partner,Cross-Sex Friend,and Sibling Jealousy Expression

Negative jealousy-related emotion and rumination are examined as consequences of a close relational partner's jealousy expression. Specifically, relationship type (i.e., sibling relationships, cross-sex friendships, and dating partners) and three of Guerrero et al.'s (1995 Guerrero , L. K. , Andersen , P. A. , Jorgensen , P. F. , Spitzberg , B. H. , &; Eloy , S. V. ( 1995 ). Coping with the green-eyed monster: Conceptualizing and measuring communicative responses to romantic jealousy . Western Journal of Communication , 59 , 270 – 304 . [CSA] [Taylor &; Francis Online], [Web of Science ®] , [Google Scholar]) forms of jealousy expression (i.e., distributive communication, integrative communication, and negative affect expression) are compared according to negative jealousy-related emotion and rumination following a hypothetical partner jealousy expression situation. Siblings and dating partners reported experiencing more intense negative emotion than cross-sex friends after partner jealousy expression. Further, participants reported ruminating more after their partners used distributive communication compared with integrative communication or negative affect expression to express jealousy. Emotional intensity did not vary according to type of jealousy expression and rumination did not vary with regard to relationship type. Practical and theoretical implications for the study of negative emotion, rumination, and partner jealousy expression are discussed.     

Anecdotal episodes in the social context

This article examines poor, moderate and good anecdotal episodes in the social context. The research results suggest 1) the quality of anecdotal episodes is discriminated on the basis of energy and clarity concepts rather than the traditional delivery and content perspective, and 2) the energy and clarity of anecdotal episodes are related to observer's perceived social attraction and projected potential to achieve social and professional success. Tnese results suggest that anecdotal episodes merit further consideration in relational development research from the communication perspective.     

Mate Value Discrepancy as Predictor of Forgiveness and Jealousy in Romantic Relationships

Mate value discrepancy (MVD), the perceived difference in resource value between self and partner in romantic relationships, may impact both forgiveness and jealousy. One hundred seventy-nine participants rated their own and their partner's mate value, and self-reported forgiveness and jealousy. MVD was associated with forgiveness in romantic relationships in that the higher the value of one's mate in relation to self, the more likely an individual would forgive that partner's transgression. Similarly, MVD played a role with jealousy in that the higher the value of the partner, the more likely an individual experienced jealousy. Additionally, individuals were more likely to forgive transgressions when their partners had higher mate values than theirs, even when jealousy is experienced.
