Effect of interrupted endogenous BMP/Smad signaling on growth and steroidogenesis of porcine granulosa cells

Bone morphogenetic proteins (BMPs) play a critical role in the growth and steroidogenesis of granulosa cells (GCs). BMP signals act through membrane-bound heteromeric serine/threonine kinase receptors. Upon ligand binding, BMPs activate intracellular Smad proteins and regulate growth and apoptosis in various cell types. The objective of this study was to demonstrate the effects of BMP/Smad signal on growth and steroidogenesis of porcine GCs. A strategy of RNA interference (RNAi)-mediated ‘gene silencing’ of Smad4, a core molecule mediating the intracellular BMP/Smad signal transduction pathways, was used to interrupt endogenous BMP/Smad signaling. Results indicate that Smad4-small interfering RNA (siRNA) caused specific inhibition of Smad4 mRNA and protein expression after transfection. Interrupted endogenous BMP/Smad signaling significantly inhibited growth, and induced apoptosis of porcine GCs, while decreasing estradiol production. In addition, interrupted BMP/Smad signaling significantly (P<0.05) changed the expression of Cyclin D2, CDK4, Bcl-2, and Cyp19a1. These findings provide new insights into how BMP/Smad signaling regulates the growth and steroidogenesis of porcine GCs.

中国美术的审美特色是在漫长的岁月中形成的,它是和中国人的哲学观紧密相连的.这个审美特色缘起于诸子百家时期,在以后的发展和形成过程中又吸收了大量的外来思想,在各种思想的共同作用下逐渐定型,并在我国封建时代中到达了高峰.近代西学的进入给传统的审美思想带来了强劲的冲击,但百年的中西文化碰撞还没能让中国的审美特色发生质的变化,中国画的审美精神仍然是立足于中国人的哲学思想,发生变化的只是具体的技法.     

Investigation of blood pulse PPG signal regulation on toe effect of body posture and lower limb height

Objective: To study the regulation of blood pulse volume via photoplethysmography (PPG) signal detected from toe, while the lower limb is passively raised in different height positions. Methods: Use a modified non-invasive PPG technique to detect the blood pulse signal on toe with infrared (IR) photo sensor. A protocol consisting of two postures, i.e., supine and 45° reclining, was designed to conduct laboratory trial in this study. During the period of performing the protocol of these postures, the lower limb was passively raised from the heights of 10 cm to 60 cm randomly and individually with sponge blocks underneath the foot. Results: In the supine posture, the higher the foot was passively raised, the more the blood PPG signal decreased. In the 45° reclining posture, the blood PPG signal increased at the beginning and then decreased in the foot height position from 10 cm to 60 cm. In both postures the normalized AC signal changes significantly while the normalized DC signal changes little. Conclusion: The toe PPG signals can obviously indicate the regulated blood volume change with the designated postural procedures due to the heart level position.     

HIT—MU（2）的结构和复杂度

SAT问题(可满足性问题)是计算机科学的核心问题,研究SAT问题的方法很多,利用极小不可满足公式的性质来研究SAT问题是近几年兴起的一个热点研究方向. 本文主要利用1(,)*-消解和分裂方法研究了差为2的碰撞极小不可满足公式集()2(MUHIT-)的结构和复杂度.此前,只有G.Davydov, I.Davydova 和H.Kleine Büning对)1(MU和)2(MU的结构和复杂度得出了较好的结果.     

光谱法研究芦氟沙星与人血清白蛋白的相互作用

用光谱法研究水溶液中芦氟沙星与人血清白蛋白的结合反应，分别测定了295，310K两个不同温度下二者的结合常数心和结合位点数n，探讨了其荧光猝灭机制；根据热力学参数确定了芦氟沙星与人血清白蛋白之间的作用力类型，以氢键或范德华力为主，并运用Foster偶极-偶极非辐射能量转移原理。测定了其相互结合时授．受体间的距离r；采用同步荧光技术考察确定芦氟沙星对人血清白蛋白构象产生一定的影响．     

基于新鲜度和配送成本的易腐食品配送路径

为解决企业在制定易腐食品配送计划时难以权衡配送总成本与交付产品的新鲜度的问题,建立以配送总成本最低和交付产品平均新鲜度最大为目标,带时间窗的易腐食品配送路径规划模型。采用自适应差分进化(differential evolution,DE)算法求解模型,通过数值算例验证模型和算法的有效性。与基本DE算法和基本蚁群算法的求解结果进行对比,自适应DE算法的求解结果更优,收敛速度更快。求得的帕累托解集表明配送总成本与交付产品平均新鲜度相悖,增加少量的配送成本可以使交付产品的平均新鲜度得到大幅提升。对易腐食品保质期、时间窗宽度和车辆装载量进行灵敏度分析,为企业在不同配送情景下在配送总成本与交付产品的平均新鲜度之间的权衡提供参考。     

科技评估DIIS方法

科技评估在科技管理中发挥着重要的作用,从智库角度而言,其不仅是智库研究的重要任务,而且是智库研究质量的重要保障。然而,现有科技评估方法通常作用于评估问题的局部,缺少从辩证法及系统论角度对科技评估全过程方法体系的思考与认知。为此,文章将潘教峰提出的解决智库研究一般问题的DIIS理论方法运用于科技评估,据此提出科技评估DIIS方法,进而从DIIS视角出发将科技评估在科技决策咨询中的作用界定为六大功能:证据形成功能、衡量比较功能、诊断分析功能、前瞻预测功能、价值导向与判断功能和质量控制功能。最后以机构评估、项目评估、政策评估和研究质量评估为例,分析科技评估DIIS方法的实践应用,以期为我国科技评估实践提供一种方法指引与支撑。     

结合速度和干扰观测的船舶路径跟踪模型预测控制

针对船舶路径跟踪控制中的舵角需优化﹑舵幅和舵速受约束等问题,提出模型预测控制(model predictive control,MPC)算法。为处理系统高阶状态值不易测量以及环境干扰问题,设计高阶非线性观测器,同时对船速和包含模型不确定项和外界干扰的总未知项进行估计。以引入舵机响应系统的MMG模型作为预测模型,不仅能提高预测精度,而且更符合船舶运动控制的实际情况。仿真结果表明,所设计的控制器在风浪流时变干扰下仍能跟踪参考路径,舵角幅值小且其变化是光滑的,速度值和总未知项也均能被准确地逼近,验证了所提算法的有效性。
