榛子的立体培养及紫杉醇的提取

摘    要:这篇章着重比较分析了榛子的芽和幼茎愈伤组织中紫杉醇的合成能力。用高效液相色谱仪测定榛子,结果显示芽比幼茎含量更高。用2.0mg/L BA和1.4mg/C LNAA诱导幼芽,8天后的愈伤组织以100%的速度感应。较好的培养愈伤组织的方法是用0.5mg/L2,4-D、1.0mg/L NAA和0.5mg/L KT补充B5培养基。测得愈伤组织的生长速度和紫杉醇的含量分别是7.74g/L和0.13%。

Abstract:This paper makes a comparative analysis on the Corylus c allus inductio n in its bud and stem. The HPLC method is used to determine the Taxa l composing ability in Corylus callus. The result indicates that bud is a more accessible so urce of Taxal than young stem is. The callus could be in duced from bud in 8 days on MS medium supplemented with 2.0 mg/L BA and 1.4 mg/L NAA. The rate of callu s induction from bud is as high as 100%. The superior medium for the callus grow th is B_5 medium supplemented with 0.5 mg/L, 2,4-D 1.0mg/L NAA and 0.5 mg/L KT . It's growth rate and Taxal content are 7.74g/L and 0.13% respectively.
