藏獒基因组DNA提取方法的比较研究

摘    要:分别采用酚一氯仿法和改进的盐析法对藏獒血液基因组DNA进行了比较提取.并以提取的基因组DNA为模板进行了RAPD-PCR扩增.结果表明.酚-氯仿法提取的藏獒血液基因组DNA在浓度和纯度上均显著的高于改进的盐析法(P〈0.05),但两种方法所提取的藏獒基因组DNA都可用于RAPD分析.

Abstract:The methods of phenol-chloroform and improved salting out were used for whole blood genomic DNA Extraction of Tibetan Mastiff, and the RAPD fragments amplified with DNA template extracted by two methods.The results showed that the concentration and purity of genome DNA by the method of phenol-chloroform were significantly higher than the method of improved salting out (P〈0.05),but extracted genome DNA by the two methods could be used for RAPD analysis of Tibetan Mastiff.
